Measurement of Spectral Characteristics of Fluorescent Voltage-Sensitive Dyes

This bachelor thesis deals with optical measurement of substances properties in practical service of absorption and fluorescent spectroscopy. The work specifies the principle of fluorescence, which has direct association with spectral characteristics measurement of so-called voltage-sensitive dyes (in this instance di-4-ANEPPS). This dye is used for contactless (optical) imaging of organisms electric activity in practical service of its voltage dependent spectral shift after binding itself on cell membrane. However, the right application of this fluorescent dye is dependent on too many factors – one of them, discussed and measured in this work, is di-4-ANEPPS spectral dependency on its immediate environment.
Measurement uncertainties in photometry

This master's thesis is focused on problematic of measurent unceartainties in photometry. Theoretical part describes photometric measururing instruments, their uncearnities and limitations. In the second part of this thesis there is a mathematical description of calculating uncearnities in laboratory conditions. In practical part there are calculations of uncearnities for photometric instruments at light laboraty VUT Brno.
Possibilities of utilization of nitrogen compounds from liquid suspensions generated in waste management

This thesis deals with the removal of ammoniacal nitrogen from biogas plant waste to produce a new usable product. In the theoretical part of the thesis, attention is paid to waste management from the perspective of biogas plants, the problem of nitrogen, and its removal. The thesis contains, among other things, theoretical calculations that include distribution and precipitation curves. The practical part deals with the correctness of these curves, as well as the precipitation of ammonium ions to form ammonium magnesium phosphate and stripping. The practical part includes quantitative and qualitative analyses of the individual components involved in precipitation. Nitrogen removal was performed on the model and real samples.

The bachelor thesis focuses on the evaluation of water quality in the Velička River. The Velička is located in the Hodonín district in southern Moravia. Its total length is 40.2 km. Its source is on the western slope of Velká Javořina and flows into the Morava River near Strážnice. The water flow was monitored in eight profiles at two-month intervals for one year. The monitoring had two phases. The first phase consisted of taking a water sample and measuring some of the water quality parameters in the field using an instrument kit from Hach Company. These were indicators of pH, conductivity, oxygen content, and water temperature. The second phase was carried out in a laboratory by measuring the quality parameters from the water samples using a spectrophotometer. The concentrations of total phosphorus, sulphates, nitrate nitrogen, and chlorides were monitored. The quality of the water in the river was evaluated according to the ČSN 75 7221 standard and government regulation No. 401/2015 Coll. Total phosphorus was evaluated as the most unfavorable qualitative parameter.

The work is aimed at monitoring water quality in Příseckém creek. Water samples were collected at six stations, distributed over the entire length of the water flow. Subscriptions were carried out in bi-monthly intervals for a period of one year. The objective of this work is to determine the quality of the water according to the selected indicators and the subsequent evaluation of the influence of anthropogenic activities in the vicinity of the flow (for example, agricultural activities, reckless behavior of citizens or the influence of recreation). In determining the water quality were as indicators in the field of measuring selected values of pH, temperature, electrolytic conductivity and content of dissolved oxygen. The collected samples were in the laboratory of the Institute of applied and landscape ecology of Agronomic faculty of Mendel university in Brno determined the value of the concentration of phosphates, nitrate nitrogen and sulphates. The concentration of the phosphates were required to be recalculated on total phosphorus. The detected values of monitored indicators were evaluated according to the standard ČSN 75 7221 and annex no. 3 to government regulation 401/2015.

This master thesis depicts process of composting of food waste using an electric composter. Electric composters are eligible solution for waste management of catering facilities, that are in accordance with current legislation. The problematics of the food waste is being described using the literature review. The main goal of this thesis is quality evaluation of the final product of the composting process. The main focus is on evaluation of enzymatic activity of compost named arylsulfatase, which corelate with sulphur cycle. The enzymatic activity itself is being measured by spectrophotometer UV/VIS Lambda 25 (Perkin-Elmer, Shelton, CT, USA) using 5 mMp -Nitrophenyl sulfate as a substrate. Thesis also studies pH values of compost and phototoxicity (by Phytotoxkit test), that demonstrate an effect of final product of composting on white mustard seeds (Sinapis alba L.).

The bachelor thesis deals with the monitoring of the quality of the water flow Nešůrka. On the flow were selected six sampling points along the entire length, on which there was a monitoring of the water quality for a period of one year. The aim of this work was to assess the water quality according to the selected indicators. Survey of values was divided into measurements in terrain and laboratory analysis. In the terrain were measured pH value, water temperature, electrical conductivity and dissolved oxygen concentration. The samples for laboratory analysis were taken at the same time during the measurement in the terrain. The laboratory work was done with the help of the spectrophotometer and the values of nitrate nitrogen, reactive phosphorus, sulphates and iron were find out. The concentration of reactive phosphorus was necessary to recalculate to total phosphorus. The measured values were evaluated according to the standard ČSN 75 7221 Classification of surface water quality and according to annex No. 3 to the government regulation No. 401/2015 Coll.
Monitoring lesních studánek v katastrálním území Bílovice nad Svitavou

The Bachelor thesis is focused on monitoring water quality in forests springs in the cadastral area of Bílovice nad Svitavou. The purpose of this work was field monitoring and laboratory chemical analysis in one-year period, in two – months intervals. The monitoring was possible because of transferable apparatus Hach 30d, including probes for measuring pH, water temperature and electrolytic conductivity. The laboratory part was done at the Institute of applied and landscape ecology, Faculty of Agronomy, Mendel University in Brno. Laboratory chemical analysis content determination manganese, sulphates, iron, nitrites and nitrates were done by spectrophotometer Hach DR4000. The values and concentrations of selected indicators of water quality were compared with the hygienic limits of physical and chemical indicators given in regulation č. 252/2004 Sb., as amended. The results of work will be provided to national register of springs in Czech Republic, on the website www.estudanky.eu
