This bachelor´s thesis was focused on study of biocompatibility of organic semiconductors. In introduction, organic semiconductors and their properties, benefits and features of selected representatives were described. In the following part biocompatibility definition and approaches for its assays including tests of cytotoxicity, material stability were presented. The last part was focused on overview of cell cultures, appropriate cultivation conditions and compounds of media for in vitro testing of biocompatibility. Experimental part showed the result of biocompatibility testing of materials. For this purpose murine 3T3 fibroblasts were used.

Detection viability of bacteria population using flow cytometry

This work deals with the detection of viability of Xanthomonas euvesicatoria using a flow cytometer. The center of this work was tp optimize the method and the treatment of bacterial cultures with high temperature at given time intervals. The heat-treated cultures were dyed with an intercalating dye called propidium iodide which marks the permeabilized membranes of the cells and thus provides information on the number of dead (apoptotic) cells. The evaluation of the results confirmed the assumption that the number of surviving cells would decrease with the increasing treatment time. A temperature of 90 ° C was used, which according to other works, seems to be the most evident for the Xanthomonas genus. Flow cytometry has proven to be a simple, fast and efficient method for the viability detection of Xanthomonas euvesicatoria.
Detection of viability of \kur{Pseudomonas syringae} pv. \kur{oryzae} using flow cytometry

The viability of Pseudomonas syringae pv oryzae was analyzed by flow cytometry and intercalating dye propidium iodide in this thesis. The treatment of bacterial cultures with high temperatures in time series was the basis of this thesis. Statistically evaluation of the data obtained acknowledged the tendency of decrease of number of surviving cells in the population, depending on the growing period of treatment.
