Methodical aspects of genetic diversity of cattle

The aim of this thesis is to describe methods for assessing genetic diversity. At the beginning is described biological diversity and its distribution with focus on genetic diversity. Furthermore, mutations, segregation, recombination, migration, selection, genetic drift, and inbreeding are described. The work also deals with the polymorphisms including SNP, VNTR and microsatellites. They are described as genetic markers, marker assisted selection and genomic selection. For within-population genetic diversity the text deals with biometric methods, the part describing interpopulation genetic diversity is concerned to distances and software packages. Finally, the phylogenetic trees and thein types are described with examples of use in practice.

The aim of the thesis was to study the genetic variability of wheat genotypes with blue caryopsis using microsatellite markers. This analysis was followed by the identification and characterization of candidate sequences involved in the non-standard blue pigmentation of wheat caryopsis and by related bioinformatic analysis of these candidate sequences. Furthermore was studied gene expression of selected candidate genes during caryopsis maturation. In addition the blue pigment storage in aleurone layer of the caryopsis was examined microscopically and finally based on orientation screening of collection with blue aleurone layer genotypes were selected as the most perspective three genotypes according to good bread-making quality, that completed the comprehensive insight into the topic of selected bread wheat blue aleurone genotypes.

The aim of this thesis is to evaluate the possibility of using sunflower for phytoremediation technologies of soil polluted with over-limiting concentrations of heavy metals. Helianthus annuus L.was selected for our experiments due to its phytoremediation properties. As heavy metals, cadmium, which is abundantly contained in fertilizers, lead, of which concentrations in the environment greatly increased thanks to the using of leaded fuels, and silver, which can be considered as an environmental issue due to its using in nanotechnology, were used. Changes in the activities of chosen enzymes (ALT, AST, urease), concentrations of specific peptides (GSH, GSSG, PC) and the ability to receive and distribute heavy metals were used to evaluate the suitability of plants for phytoremediation purposes.
