Viral vectors based on tobacco mosaic virus for the expression of recombinant proteins in plants.

(eng): Over the past few decades, plant viruses and vectors derived from them have become not only an integral part of the work in laboratories, but also began to be used commercially for the production of vaccines, drugs, enzymes and other proteins / peptides. One of the most commonly used viruses for the expression of heterologous proteins is tobacco mosaic virus. In this work, I would like to focus on plant viruses and vectors derived therefrom. Compare it with other expression systems used in plants and describe how their benefits could be further used and combined with modern methods of cloning. The expression profile of cathepsin L in developmental stages of Fascioloides magna

Our experimental organism Fascioloides magna is a digenetic liver fluke from Fasciolidae family which parasitizes in domestic and free-living ruminants of North America and Central Europe including Czech Republic. In Czech Republic this highly pathogenic worm causes a severe liver damage to cervids and bovids and the prevalence locally reaches up to 95%. The biology of F. magna including e.g. the characteristics of host-parasite molecular interaction and the functions of particular molecules produced by the parasite are not fully understood. According to results of our previous research the excretory-secretory products of F. magna adults contain number of molecules which play the crucial role in host tissue invasion, digestion and evasion of the host immune response. One of the most abundant is cysteine peptidase cathepsin L (FmCL). FmCL is supposed to play various key roles in biological processes of all stages during a life cycle and therefore we can suppose its different expression level in particular life stages. In order to define the expression level of FmCL we performed the pilot study with miracidia and adults where the qPCR method was applied. Industrial landscape. The possibility of theme in art and in art education

This diploma thesis deals with industrial landscape in the visual arts in terms of educational, historical and theoretical viewpoints. In the theoretical part, the author deals with the interpretation of various terms that are related to this industrial issue. At the same time she outlines the development of the industrial landscape, exploration of relationships and attitudes, mostly among the younger generation, to these complexes, and deals with this issue from the aspect of art, such as suggestions or industrial materials. Very interesting and more recent phenomenon is the activity called Urbex, whose interpretation, the world and Czech leaders and its development is briefly explained. Concurrently, this activity is supplemented by visual arts, by the theme of industrial buildings, by industrial material, by memories, by feelings and relation to this landscape of the author of the thesis. In the didactic part, which is crucial to this diploma thesis, the author has developed five preparations for units of teaching in art lessons, where she focused both the use of industrial materials and this theme . Indication of female gender in new names of professions

The bachelor's thesis Indication of Female Gender in New Names of Professions is divided into two parts. The first one describes the indication of female gender in linguistics theory and presents main suffixes used for the indication of female gender. The second part tracks occurrence of the expressions excerpted from the vocabulary Nová slova v češtině 1 a 2 and the article Pojmenování nových profesí u pozic v oblasti ekonomiky a podnikání (v rusko- českém srovnávacím plánu) by Ljuba Mrověcová (2009). The expressions were searched for in the corpus SYN 2010 and Neomat database. Lists of names, both found and not found in the sources mentioned above follow. The meaning of less known names is explained in the alphabetical vocabulary.
Freedom of speech and its limits

This diploma thesis deals with freedom of speech and its limits. Freedom of speech as the fundamental right which is necessary to proper functioning of a democratic state where the law rules. In this regard the freedom of speech has to be protected properly. Protection in Czech legal order is entrusted with the legal regulation of the major legal force, part of constitutional order, "Charter of fundamental Rights and Basic Freedoms". Freedom of speech as we know it today, was expressed after the Second World War as a prevention of similarly devastating conflict in the future. Certain desire for the free dissemination of ideas we can see in a deep histor when firs attemps of gaining freedom of speech was closely connected to religion. In the 19th century, when the freedom of denomination has found its stable place in the statutes of the most European countries, the freedom of speech has become to be undestood in connection to the freedom of the press, that was the only communicatory media. The function of the language semiosis in the Quentin tarantino's films

Diploma thesis "The function of the language semiosis in the Quentin Tarantino's films" deals with interception and description of the uniqueness of linguistic expressions in the films of one of the most important film directors and screenwriters - Quentin Tarantino. Through semiotic analysis it examines significant elements in movies and screenplays and describes the characteristic method of used expressions. It focuses mainly on characteristics signs and symbols as the main tool of semiosis, and their function in the film language. Diploma thesis focuses also on description of used semantic expressions, explanation of their occurrence in the author's pieces, and trying to capture the originality of the artist's expression. The final part examines Tarantino's role in the cinematography and tries to explain the causes of the great media success.

In my work I discuss the analysis of metodical procedures where I focus on the arrangement of emotional experience through active listening to music while performing physical activities. I mainly focus my attention on the feeling of the rythm and its' inner devision. The aim of my bachelor work is to suggest my own version of a teaching plan which is created with attention to the developement of relevant music abilities and to the age of the child. Transfer between verbal and algebraic descriptions. Pupils' strategies and problems

The aim of this thesis is to gain insight into the problems that pupils have when switching between verbal and algebraic expressions, through the analysis of their problem-solving processes. The work is divided into theoretical and experimental parts. The theoretical part of the paper contains three sections. The first describes the analysis of three selected Czech textbooks series for primary schools in terms of teaching the concept of variables and algebraic expressions. The next section deals with international comparative research TIMSS and the success of Czech pupils of year 8 in algebra. In the third section, results of international research concerning the issue of transition between verbal and algebraic expressions are described. The main part of the work is the experimental part, it aims to describe the difficulties encountered by pupils in algebra and to identify their likely sources. The basis for the own research was a set of 9 problems based on released TIMSS problems. The target group of pupils was year 9 of primary school, with whom the investigation was carried out via think-aloud interviews. Stability of the catalytic domain of neutral trehalase studied by differential scanning fluorimetry

This bachelor thesis is part of a project aiming for the crystallization of trehalase domain of neutral trehalase Nth1 from Saccharomyces cerevisiae. The main goal of this thesis is to optimize the purification protocol of two different constructs of yeast Nth1 and determine the optimal buffer for crystallography of these constructs using the differential scanning fluorimetry DSF. Trehalases are important, highly conserved enzymes found in many organisms. These enzymes belong to the class of hydrolases, subgroup of glycosidases and their common feature is hydrolytic cleavage of trehalose molecule into two glucose subunits. Trehalose is a naturally occurring non-reducing disaccharide which serve in yeast cells as storage carbohydrate and energy and stress metabolite. Trehalose and its hydrolysis is very important in the life cycle of the insect, as it is present as the main sugar component of insect hemolymph, so some trehalase inhibitors could be used as insecticides. By this time, however, only one structure of trehalase from a prokaryotic organism Escherichia coli was solved. For this reason it is very important to solve the structure of trehalase from a eukaryotic organism. Perception and expression of non-verbal vulgarity in context of gender

Vulgarity is becoming to be an increasingly discussed and interesting topic, although not deeply explored by science. This thesis includes a survey focused on vulgar gestures. The main objective is to compare differences in perception and expression of non verbal vulgarisms- gestures among 1) boys and girls 2) two different teenage groups, in terms of frequency, form and purpose of using the gestures. I mainly use a closed, semi-closed or open questions in the questionnaire, which also includes a parallel presentation of each gesture.
