Depiction of side effects of Covid-19 vaccines in Czech television news
Zelbová, Judita ; Klabíková Rábová, Tereza (advisor) ; Křeček, Jan (referee)
This thesis focuses on how the side effects of vaccination against covid-19 were presented by media during the pandemic. The aim was to find out how the side effects were reported by Czech national television stations in their evening news programmes, what were the differences in presentation between the stations and which experts were invited to comment on the topic. The thesis primarily uses qualitative content analysis and to a lesser degree quantitative content analysis. The thesis shows that three of the four TV stations studied (ČT1, Nova and Prima) approached the presentation of the topic in a very similar way conceptually, TV Barrandov being quite different in this respect. All TV stations presented side effects on the basis of substantiated data, with an emphasis on information obtained from relevant organisations (European Medicines Agency, Czech State Institute for Drug Control) and experts (mostly epidemiologists, immunologists and physicians of various specialties). Contrary to the expectation, the coverage of the side-effects proved to be relatively marginal and only became notable in the spring of 2021, when a larger number of adverse reactions occurred with the AstraZeneca vaccine.
Viral proteins that facilitate RNA replication
Černý, Prokop ; Bouřa, Evžen (advisor) ; Faltová, Lenka (referee)
In recent years, the pathogenic human coronavirus SARS-CoV-2 (Severe Acute Respiratory Syndrome Coronavirus 2) has caused a global pandemic of highly infectious disease COVID--19 (coronavirus disease 2019). According to the World Health Organization, as of August 20, 2023, there have been nearly 770 million confirmed cases and nearly 7 million deaths associated with the disease. However, only a small number of effective antiviral drugs are currently known to help treat the disease in already infected patients. Therefore, in this work, the interaction of host 14-3-3 proteins with the nucleocapsid protein of SARS-CoV-2 was studied as a potential biological target for the rational design of new antiviral drugs. The importance of this interaction in the virus life cycle has not been fully elucidated, but it probably has an important regulatory function affecting multiple processes in the infected cell. The SARS-CoV-2 N protein contains several binding motifs for 14-3-3 proteins in its sequence, two of which (at positions S197 and T205), most likely responsible for the protein- protein interaction, were studied in this work. Microscale thermophoresis was performed to determine the affinity of each of these phosphorylation sites on the nucleoprotein for the 14-3-3ζ protein. The key phosphorylated residue...
Behind the front line, Rehabilitation during Covid-19 pandemic
Novotná, Klára ; Vaněk, Miroslav (advisor) ; Wohlmuth Markupová, Jana (referee)
The thesis is based on 13 oral history interviews the impact of the Covid-19 pandemic on the health care sector (specifically rehabilitation medicine and physical medicine). Narrators are health care professionals who are part of multidisciplinary rehabilitation care in different positions and can therefore reflect the pandemic from different perspectives. In addition to the impact on the professional life, the thesis also describe the subjectively perceived impact on the narrators' personal lives. The interviews were recorded in early 2022, after 2 years of living with the pandemic, when strict restrictions were still in place. The thesis provides testimonies of the lived experience of the pandemic among health workers behind the so-called 'frontline', who have not received as much attention in the media. It also compares the experiences of the narrators in the context of Czech realities and compares them with the results of similar international studies on pandemics among health professionals.
Výuka výchovy k občanství a základů společenských věd během pandemie covid-19
ŽIDOŇ, Zdeněk
Between 2020 and 2021, the whole world was dealing with a coronavirus pandemic that had a major impact on education. An unprecedented number of pupils and students found themselves in distance learning for which they were unprepared. The purpose of this paper is to determine what impact distance education has had on the teaching of civics and social studies. In the beginning of the thesis, the concept of key competences will be discussed and the quality of teaching of civics and social studies during traditional face-to-face teaching will be analysed. In the next part of the thesis, the course of the pandemic in the Czech Republic and its impact on the whole educational system will be analysed in detail. In the final part of the thesis, the author will try to answer the question whether it is possible to teach these subjects fully during distance education and will investigate the experience of distance learning of teachers of these subjects in České Budějovice.
The digital divide and the spread of the fake news during the COVID-19 pandemic
Misík, Dominik ; Wolák, Radim (advisor) ; Soukup, Martin (referee)
This thesis focuses on two main topics: the digital divide and the spread of misinformation during the COVID-19 pandemic. The theoretical part is dedicated to the first mentions of the digital divide and introduces some of the main people who dealt with this area. The thesis follows up with the measurement and for a better understanding there are presented the data associated with the digital divide in 2021. In the next part, basic concepts from the field of disinformation are described, which are placed not only in a historical context, but also in the current reality. An example could be fake news that appeared during the COVID-19 pandemic. The next chapter deals with defense techniques against disinformation, both domestically and internationally. The results of a global study by the World Health Organization, which investigated ways of obtaining and working with information about the COVID-19 pandemic, are presented at the end of the theoretical part. In the practical part, the findings from this study were applied to the Czech Republic, with the help of a quantitative method in the form of a questionnaire survey with more than three hundred respondents. The obtained data are then partially compared with the results from an international WHO study. The aim is to gain a basic comparison between...

Typology of domestic violence events, their motivation and conditions in the so-called coronavirus era
VACKOVÁ, Adéla
The theoretical part of the thesis explains the concept of domestic violence, its manifestations, the characteristics of perpetrators and victims, and legal protection within the Czech legislation. This part is important for a better understanding of the whole issue and for better orientation in this area. The main objective of the thesis is to assess the changes in the functioning of helping organizations in the České Budějovice during the covid-19 pandemic compared to the standard state and any disruption of the usual course of action in providing assistance during the pandemic measures. Emphasis is placed on determining the impact of quarantine on partnerships and family relationships during a period of increased mental stress. The main goal is also to determine whether there has been an escalation or a decrease in domestic violence during this time. The work will involve analyzing data obtained from intervention centers in the České Budějovice, which will provide important information about the situation regarding domestic violence during the pandemic. The results of this work will serve to improve understanding of this issue and improve support for victims of domestic violence. It is necessary to ensure that all necessary resources and support are available for domestic violence victims. This work can contribute to greater awareness of this issue and improve conditions for those affected by it.
Impacts of the COVID-19 pandemic on streaming services and their users
Měska, Tomáš ; Kruml, Milan (advisor) ; Štoll, Martin (referee)
This thesis examines the impacts of the COVID-19 pandemic on streaming services and their users. It aims to shed light on how the impacts of the coronavirus pandemic, as well as the government measures and lockdowns that stifled social life and locked people in their homes for weeks, affected the use of streaming platforms by their users. This thesis focuses on the main characteristics and types of streaming services, the basic concepts associated with them, and looks at the production of new content as well as their distribution models. The subsequent section discusses the coronavirus and its impact on social behaviour, and then in particular the impact on the film industry in terms of production, content, cinema and film festival operations and of course the streaming services themselves. The final segment of the thesis then analyses and interprets the data collected in the questionnaire survey. Respondents' answers provide the basis for finding solutions to the research questions, which identify which streaming services users use, how many they use concurrently, how much time they spend watching streaming content, what categories of content they watch, or in what ways they use the services, for example in terms of devices or in comparison to other types of media.
Structural and functional characterization of inhibition of a coronaviral methyltransferase.
Ivanovská, Dana ; Bouřa, Evžen (advisor) ; Faltová, Lenka (referee)
Coronaviral methyltransferases participate in the modification of the 5'-end of viral RNA. Their enzymatic activity not only ensures efficient mRNA translation, but also allows the virus to escape the recognition of the innate immune system. This work is focused on the SARS-CoV- 2 methyltransferases (the methyltransferase domain of the nonstructural protein 14, MT14, the nonstructural protein 16 and its cofactor - the nonstructural protein 10, nsp16/10), which represent attractive molecular targets for therapeutic intervention. The aim of this work was to structurally characterize the coronaviral methyltransferases in complex with various small molecules. The recombinantly prepared proteins were purified and subsequently subjected to crystallization trials. The obtained crystals of the nsp16/10 heterodimer in complex with sinefungin were soaked in a solution containing a S-adenosyl-L-homocysteine analogue. Crystals suitable for X-ray crystallography of MT14 in complex with two different inhibitors were obtained by optimizing the identified primary crystallization conditions. The aquired structural data of the MT14 inhibitory complexes will serve as a basis for the design of new small molecule inhibitors targeting the S-adenosyl-L-methionine binding site. Keywords: methyltransferase, nsp14, nsp16,...
