Synthesis and application of new bipyridine ligands

2,2'-Bipyridines and their appropriate N,N'-dioxides form a significant class of heteroaromatic compounds, which has found application in various fields of chemistry and predominantly in asymmetric catalysis. One of the most powerful methods for their synthesis is cocyclotrimerization of alkynes with nitriles. A new variant of cyclotrimerization reaction - cocyclotrimerization of halodiynes with nitriles, which results in the formation of 2- and 3-halopyridines, has been developed. The reaction was studied on a wide range of substrates providing the pyridine products in good isolated yields. Formation of an unexpected product of halogen exchange reaction was observed during the course of the study and its origin was elucidated by experimental studies. The prepared 2-halopyridines were used as starting materials for syntheses of new chiral 2,2'-bipyridine ligands. The crucial step of their synthesis turned out to be the reductive dimerization of 2-halopyridines to the corresponding 2,2'-bipyridines. Application of the formed bipyridine ligands was then tested in various metal-catalyzed asymmetric reactions, namely Mukaiyama aldol reaction, hydroxymethylation, conjugate addition, C-H activation of indole and desymmetrization of meso-epoxides, in which one of the bipyridine ligands showed...
Synthesis of the C15-C20 tiacumicin fragment

In the diploma thesis, a method for preparation of the unsaturated C15-C20 tiacumicin fragment was developed. Tiacumicin is classified as a macrolidone antibiotic. Although three syntheses of tiacumicin have been recently published, this study has aimed to develop a novel pathway for its preparation utilizing catalytic reactions instead of stoichiometric ones. In the second part, an enantioselective allylation of (E)-3-jod-2-methylpropenal has been developed applying catalysis by a series of N,N'-dioxide catalysts. The asymmetric induction achieved in the allylation was up to 99% ee. This procedure allows relatively simple, straightforward and efficient preparation of wide range of natural products. The synthesis of π-electron systems suitable for transfer and retention of charges

The aim of my Thesis was to develop a general synthetic methodology for the preparation of long helicenes equipped with suitable functional groups that control their solubility or serve as anchoring groups for attachment to metallic surfaces, especially gold. The well-established transition metal catalyzed [2+2+2] cyclotrimerization of triynes was selected as the key scaffold-forming transformation in the synthesis of long helicenes because of its high regioselectivity, atom efficiency, functional group tolerance and general robustness. A modular approach was used for the preparation of the starting oligoynes, thus enabling a high level of their structural diversity. Individual resorcinol- based aromatic building blocks were interconnected by Sonogashira cross-coupling reactions, providing complex cyclization precursors encompassing up to twelve alkyne units pre-arranged for the multiple [2+2+2] cycloisomerization to produce three six- membered rings from each set of three neighboring alkyne units. Thus, a small series of long helicenes with up to 19 rings constituting the helical scaffold was synthesized. The quadruple cyclization leading to the longest oxahelicene prepared to date was performed in a high-temperature-high-pressure flow reactor at 250 řC in the presence of CpCo(CO)2. The set of...

Application of H-Bonding Catalysis in Organic Synthesis

Over the last 20 years, asymmetric synthesis has seen considerable progress, particularly in the field of catalysis. In addition to enzyme catalysis and transition metal catalysis, organocatalysis, catalysis using small organic molecules also plays an important role in the asymmetric synthesis. Chiral organocatalysts allow the preparation of structurally interesting and optically pure molecules via various activation modes. This work is focused on the use of organocatalysis based on the formation of hydrogen bonds in organic synthesis. Our study was devoted to the enantioselective organocatalytic reactions of ketimines leading to the formation of chiral vicinal centers. The first part deals with the organocatalytic enantioselective addition reaction of α- fluoro(phenylsulfonyl)methanes to ketimines derived from isatin. The reaction utilizes catalysis of a commercially available quinoline alkaloid cinchonine. A series of enantiomerically pure compounds were prepared containing two neighboring stereocenters in good yields of up to 97%, with diastereoselectivity up to 6: 1 dr and with enantiomeric excesses of 70-98% ee. This bachelor thesis is focused on the stereoselective Michael-alkylation reaction of alkylidene heterocycles using bifunctional organocatalysis. Within the thesis, suitable starting materials were first prepared and then the organocatalytic reaction leading to the formation of spirocyclic compounds was optimized. Under optimized conditions, a study of the scope of application of the method on selected derivatives was also performed and the optical purity of the products was determined. Synthesis of Enantiomerically Pure Helical Aromatics Such As NHC Ligands and Their Use in Asymmetric Catalysis

Various ways of preparing enantiomerically pure 2-amino[6]helicene derivatives were explored. Ni(0) mediated cyclotrimerization of enantiopure triynes provided (M)- and (P)-7,8-bis(p-tolyl)hexahelicene-2-amine in >99% ee as well as its benzoderivative in >99% ee. The stereocontrol was found to be inefficient for a 2- aminobenzo[6]helicene congener with an embedded five-membered ring. Helically chiral imidazolium salts bearing one or two helicene moieties have been synthesized and applied in enantioselective [2+2+2] cyclotrimerization catalyzed by an in situ formed Ni(0)-NHC complex. The synthesis of the first helically chiral Pd- and Ru- NHC complexes and their application in enantioselective catalysis was demonstrated. The latter shows promising results in enantioselective olefin metathesis reactions. A mechanistic proposal for asymmetric ring closing metathesis is provided.
