Vliv stájového mikroklimatu na vybrané kvalitativní a kvantitativní parametry ejakulátů plemenných býků

The aim of this diploma thesis The effect of stable microclimate on qualitative and quantitative parameters of ejaculates of breeding bulls, where the effect of stable envi-ronment and age on the volume, concentration, activity and changes in morphology of sperm in ejaculates of bulls was evaluated. In the period from 1st March to 31st August 2014, there were done 154 collections of semen from nine bulls of Czech Pied cattle lairaged at the insemination center of bulls operated by a breeder's cooperative Impuls in Bohdalec. Immediately after the collections macroscopic and microscopic examinations have been performed for all samples, which included determining the volume, con-centration and activity of sperm. After evaluation of morphological parameters of bovi-ne sperm the percentage of morphologically normal sperm and pathomorphological da-maged sperm were evaluated. There were observed changes on the head, acrosome, connecting part and flagellum. Our results show that the biggest effect on the qualitative and quantitative parameters of semen for breeding bulls has age, the temperature along with moisture, where the greatest differences in results were found for the extreme va-lues. A value of temperature - humidity index (THI) has negligable effect, where there were the smallest differences in the obtained samples.

The aim of this study was to evaluate the differences in sperm quality in dogs of various breeds used according to the breakdown Federation Cynologique Internationale - FCI. The procurement ejaculate in 6 dogs from 3 different groups of FCI (Group I - Sheepdogs breeds II. group - Molossoid; VIII. group - Retrievers). Sampling was carried out in the laboratory department UCHSZ reproduction, FA, MENDELU. Samples of ejaculate were procured by manual handling in a preheated glass beaker at a temperature of 39 +- 1 ° C. The results were evaluated using conventional statistical methods. For macroscopic evaluation ejaculate was found that the largest volume reached dogs II. FCI group (15.0 ml), then the smallest dogs FCI group I. (8.0 ml). Regarding the sensory evaluation of semen can be stated that there was no deviation as color, odor or foreign matter from physiologically healthy sperm. The greatest activity showed sperm ejaculate obtained from dogs VIII. FCI group (90.0 %), the smallest dogs II. FCI group (70.0 %). The highest concentration of sperm in the ejaculate was detected in dogs VIII. FCI group (397.5. 103.mm-3), the smallest dogs II. FCI group (60.0. 103.mm-3). The highest percentage of morphologically normal sperm ejaculate reached in dogs I. FCI group (92.0 %), the lowest for dogs VIII. FCI group (89.0 %). From the obtained results we can conclude that the best semen quality achieved psi VIII. FCI group. In contrast, the poorer results achieved psi II. FCI group.
