Hmyzí anti-stresové reakce vůči přírodním toxinům

The aim of this bachelor thesis was to characterize the effect of Bt (Bacillus thuringiensis) toxin on adipokinetic hormone (AKH) levels in the brain, on the activity of digestive enzymes (lipases, proteases, glucosideases, amylases) in the midgut and on nutrient levels (lipids, proteins, glycides) in the haemolymph of the firebug (Pyrrhocoris apterus). Bt toxin was applied by injection to the adult males 24 hours before monitoring of the above mentioned physiological characteristics. The results showed that Bt toxin reduced AKH level in the brain, modulated level of nutrients in haemolymph, but did not change activity of digestive enzymes in the midgut.

The aim of this bachelor thesis was to characterize the effect of one insect pathogen (entomopathogenic nematode Steinernema carpocapsae) and one venom (from the honey bee Apis mellifera) on digestive enzymes (amylases, proteases, lipases), and on adipokinetic hormone production in the brain of the cockroach Periplaneta americana . The experiments were done with the adult male and female cockroaches from a colony kept in the Insectary in the Institute of Entomology BC CAS. The pathogen/venom were applied into the cockroach body by injection 24 hours before the processing. After that the activity of digestive enzymes in the midgut, and AKH level in the brain were monitored. The results showed that the nematode has no effect on enzymes, and on AKH level as well. On the other hand, the bee venom partially increased the activity of digestive enzymes, but the role of AKH in this process in not completely clear.

The main goal of this bachelor thesis was to characterize changes of adipokinetic hormone (AKH) production in the brain and corpora cardiaca of the firebug Pyrrhocoris apterus during the infection caused by entomopathogenic nematode Steinernema carpocapsae. The effects of the pathogen were investigated by determination of AKH level in the CNS using competitive ELISA test and immunohistochemical localization with a specific antibody against the P. apterus AKH (Pyrap-AKH). The results confirmed presence of majority of AKH in corpora cardiaca and smaller amount also in brain. The infection increased AKH production in both monitored organs, however, in brain the elevation was more intensive than that in corpora cardiaca.
Úloha adipokinetického hormonu v metabolismu základních živin u octomilky obecné \kur{Drosophila melanogaster}

The aim of the thesis was evaluation of various metabolic characteristics in the fruit flies Drosophila melanogaster with deficiency of adipokinetic hormone (AKH) production, and with adenosine receptor dysfunction. The experiments were done with a goal to evaluate involment of AKH and adenosine into control of the metabolic pathways. For that measuring of basic nutrients, level of Drome-AKH, mortality and some others characteristics in the fruit flies during starvation were performed. Results revealed the effect of AKH on metabolism of storage nutrients, however, the role of adenosine was unclear.
Studium adipokinetické hormonální odpovědi na infekci způsobenou entomopatogenní houbou druhu \kur{Isaria fumosorosea}

The goal of this bacholar thesis was to find out how to elicite the infection by entomopathogenic fungus Isaria fumosoroesa in the firebug Pyrrhocoris apterus. When firebugs were successfully infected by blastospors of these fungus, the level of AKH in the central neural system and metabolic activities were measured. The results revealed that the infection caused decrease of AKH level in the CNS and activation of energy sources (lipids) in haemolymph. The flow cytometry analyses indicated certain changes of hemocyte numbers and populations in the hemolymph after the infection.
Adipokinetic hormones and the energetic metabolism in the spruce bark beetle (\kur{Ips typographus} L.)

The aim of the thesis was to elucidate the occurrence of adipokinetic hormones in the spruce bark beetle (Ips typographus) by competitive ELISA and LC/MS analysis. Total lipid and carbohydrate content as well as levels of glucose, trehalose and glycogen were measured to characterize physiological changes during particular phases of the life cycle.

The aim of the thesis was to analyze and to quantify the adipokinetic hormones (AKHs) of the spruce bark beetle Ips typographus. The RP HPLC was employed for the AKHs identification, and a competitive ELISA for their quantification. Those methods indicated that the Peram-CAH-I and Peram-CAH-II could be the AKH representatives, and that their levels fluctuate during a year. The results also suggested that the hormones might substitute themselves within certain periods of the year.
