Title: Acipimox during Short-Term Exercise Exerts A Negative Feedback of Growth Hormone on Ghrelin Secretion in Patients with Bulimia Nervosa and in Healthy Women: The Role of Lipolysis Objective: Eating disorders, such as bulimia nervosa (BN) and anorexia nervosa (AN), are characterized by abnormal eating behavior. The main features of BN are binge-eating and inappropriate compensatory methods to prevent weight gain. The appetite-modulating peptide ghrelin is secreted by the stomach and shows a strong release of growth hormone (GH). A potential GH-ghrelin feedback loop between stomach and the pituitary has been recently reported. Acipimox (Aci), an analogue of nicotinic acid, inhibits lipolysis in adipose tissue (AT) and reduces plasma glycerol and free fatty acids (FFA) levels. Exercise and Aci are stimulators of GH secretion. We suppose that a negative feedback from increased GH levels during exercise may play a role in reducing plasma ghrelin levels. We surmised that altered baseline activity and exercise-induced activation of the sympathetic nervous system (SNS) results in excessive stimulation of lipolysis associated with negative energy balance and may lead to abnormal AT metabolism in patients with BN. The aim of this diploma thesis was to evaluate the occurrence of ketones and free fatty acids in milk of high-yielding cows. Ketosis is a very common metabolic disorder in dairy cows resulting in reduced milk production, fertility disturbance and more frequent occurrences of other diseases. Ketosis is not only a serious health problem, but also an economic problem, that can be avoided by balancing the feeding dose respecting the physiological needs of energy delivery after birth and at the beginning of lactation of high-yielding cows. The monitoring was carried out in 2016 at Chyšná, which is one of the centers of Agribusiness Košetice a.s. Owerall were 1407 cows monitored during the 6 utility tests (from January to June 2016). The occurrence of subclinical ketoses was defined by the content of ketones in milk (acetone and beta-hydroxybutyric acid). Critical was the period of the first 12 days of lactation, when the occurence of subclinical ketoses was the highest, the occurence of this disease was lower than in other breeds. At Chyšná breeding, the low incidence of subclinical ketoses is mainly influenced by zootechnical work and by working with the feed consultant and the company for the production of compound feeds. As a precautionary process, regular feed analysis and examination of free fatty acids and ketones in milk can be recommended.

Psychrotrophic lipolytic bacteria and content of free fatty acids in bulk samples of cow?s raw milk

The objective of the study was to analyze the relationship of psychrotrophic lipolytic bacteria in bulk samples of cow?s raw milk and their metabolic activity based on the content of free fatty acids and identifying the factors that may influence their levels. The contents of free fatty acids (FFA) and counts of total bacteria, psychrotrophic lipolytic bacteria (PLiBC) and somatic cells were determined in 150 samples of cow?s bulk raw milk on 20 farms with three different milking technologies in South Bohemia during 2008?10. FFA were determined using an extraction-titration method. Within the compared technologies, the highest mean values of FFA (38,8 mmol?kg-1; P<0,001) and PLiBC (696 CFU?ml?1) were observed on farms with pipeline milking in stalls. The lowest mean FFA level (15,4 mmol?kg-1) was determined on farms with an automatic milking system. Medium values were determined on farms with parlour milking. From the aspect of the inhibition of an increase in psychrotrophic bacteria, and mainly in psychrotrophic lipolytic bacteria in conditions of cold storage of raw milk the temperature of 4°C seems optimum as it markedly inhibits the growth of mesophilic and psychrotrophic bacteria and at the same time the increase in the values of free fatty acids is slower at this temperature compared to the temperatures of 6,5 and 10°C.
