Genetické aspekty melanomu

The aim of this diploma thesis is to summarize findings about the cellular, molecular and mainly genetic characteristics of malignant melanoma, which is one of the most harmful human malignancies with steadily increasing incidence in recent decades. Malignant melanoma is a highly complex disease in which a huge number of genes play a role. The experimental part is devoted to the detection of mutations in the CDKN2A and CDK4 genes and polymorphism C609T in the NQO1 gene in participants with melanoma or pancreatic cancer in the family.
Gene NQO1 polymorphism analysis using the PCR method

This bachelor thesis deals with the detection of polymorphism of the NQO1 gene by PCR-RFLP methods. The thesis describes the exact methodology of detection of polymorphism commonly used in the clinical laboratory. The methodology including DNA isolation, determination of DNA concentration, PCR, RFLP, and electrophoresis. The detection was performed in 120 patients in genetic laboratory GENLABS s.r.o. in České Budějovice. The results of the analysis were evaluated and compared with the literature.
