Design of colagen capilary beds phantom for cells accumulation testing and blood vessels blocation

This bacalor thesis describes the adhesion of cells marked with iron nanoparticles to the capillary wall which is exposed to magnetic force. First, it characterize the bloodstream and speed of flowing blood. It also discusses the general characteristic of the nanoparticles useful in medicine, description of used microscopes and lastly proper methods of cultivation cells in vitro. The practical part tests the adhesion of the cells at different speeds, in different sized capillaries using magnetic forces.
Cell Viability Measurement

This master's thesis deals with the ways of cell viability determination and its measuring. The first part of this thesis has a brief introduction into the theory of cell viability, also the methods of cell viability measurement and different types of fluorescent probes are described. The thesis further includes a brief description of the image processing methods, as well as the fluorescent microscope and applied filters are described. In the practical part the system implemented at a graphical programming environment LabVIEW used for the cell viability evaluation is described, functions of applied units are explained. The program is checked on model data.

The thesis covers the theory of fluorescence principles, fluorescence methods and indicators for measurement of concentration of intracellular calcium. The procedure of measurement by means of fluorescence microscope, camera and indicators is also mentioned. Literary solution is presented. Master‘s thesis deals with the methods and system for measurement of concentration of intracellular calcium in LabVIEW. The thesis covers the theory of fluorescence principles, fluorescence methods and indicators for measurement of concentration of intracellular calcium. The procedure of measurement by means of fluorescence microscope, camera and indicators is also mentioned. The theory dealing with confocal microscopy is presented and two techniques are compared. Literary solution is presented. The practical part describes the program with a graphical user interface used for acquisition and analysis of data measuring dynamics of intracellular calcium concentration. The experimental part describes the preparation and measurement of cells and evaluation of experiments.
