Breath Alcohol Tester

The theme of bachlor’s project is design of simply breath alkohol tester. A breath alcohol test determines how much alcohol is in your blood by measuring the amount of alcohol in the air you breathe out (exhale).My task is interpret the possibility of measuring the alcohol content in the body. Describe the different methods and compare their accuracy and useability for practical measurements and also used in law enforcement.
Electrochemical analytical devices for thick-film sensors measurement

This work covers an area of electrochemical analytical systems for trace determination of dissolved species in water solutions with use of thick-film sensors. Several types of common electrochemical devices including special ones used for sensors are described and compared in this work. The main goal of this work is to find the suitable electrochemical arrangement with the best, reliable and reproducible current response of the thick film sensors. A new electrochemical arrangement, which should eliminate disadvantages of common devices and unite their advantages with optimization for thick film sensors, is designed here. For better current response of the sensor, suitable improvements and optimization of the new electrochemical arrangement prototype were done in this work. All measurements were done on standard electrochemical couple of potassium ferro-ferricyanide using the cyclic voltammetry. In the end of this work, all the devices are compared in term of their features and presumptions for the best detection limit of the thick film sensors.
Electrochemical detection of purine metabolites and their isosters

Electrochemical analysis of some important purine derivatives from catabolic cycle of purine, and allantoin is presented. Determination is based on the forming of purine Cu (I) complex on the mercury or carbon paste electrode. Electrochemical stripping by suitable change of potential results in voltammetric signal, which reflects purine concentration in the sample. Procedure was optimalized and calibration curves were measured.
A Preliminary Study of Modification of 7-Deazaadenine with a Complex of Osmium Tetroxide with 2,2 '-Bipyridine

Reactivity of a purine nucleic base analogue 7-deazaadenine (A*) in short oligodeoxynucleotide (ODN) towards an osmium tetroxide complex with 2,2'-bipyridine (Os, bipy) was investigated by means of electrochemistry, capillary electrophoresis and MALDI-TOF mass spectrometry. Electrochemical measurement, electrophoretic analyses and mass spectrometric analyses of reaction mixture proved that ODN with an A* residue yields an osmium adduct with properties similar to those exhibited by the well-known adduct of thymine.

Voltammetric Analysis of Anthraquinone-labeled Nucleotide Triphosphates and Oligonucleotides at Gold Electrodes

DNA labelling is used for increase of sensitivity of electrochemical detection. When the DNA or oligonucleotides (ONs) are chemically modified, their electrochemical responses can be changed depending on electrochemical activity of the introduced moiety. One option of DNA labeling is incorporation of chemically modified nucleotides using corresponding deoxynucleotide triphosphates (dNTPs) into DNA molecules with using primer extension (PEX). In our study we used anthraquinone labeled dNTPs and ONs with enzymatically incorporated anthraquinone labeled nucleotides. Thus synthesized ON stretch bearing the anthraquinone tags was electrochemically analyzed using voltammetry at gold electrodes.

Base-modified anthraquinone-nucleotide triphosphates were used for DNA labeling. Anthraquinone modified nucleotides were incorporated into DNA by primer extension with KOD polymerase. In cyclic and square wave voltammetry at HMDE or PGE antraquinone gave a well developed characteristic signal suitable for differentiation of the labeled DNA from unlabeled one.
Electrochemical analysis of DNA using switchable redox moieties

Labelling of DNA with electrochemically active moieties proved to be a convenient way to the development of electrochemical techniques for the sequence-specific DNA sensing. Through combinations of various labels differing in redox potentials, independent redox coding of different DNA sequences or individual nucleobases can be attained. Applications possibilities of electrochemistry in analysis of modified DNAs are further extended by facile monitoring of chemical conversion of reactive groups on DNA during post-labelling with ultimate redox labels. In addition, controlled in situ electrochemical conversions of specific intrinsic and extrinsic DNA components can be utilized to switch their electrochemical signals and improve signal resolution.
