This work is focused on theoretical and practical study of genetic diversity in carrots species. The aim of this work was to analyze the DNA polymorphism in Daucus carota L. by using molecular biology methods based on DNA isolation, PCR and electrophoretic separation. A total of 20 samples were analyzed by using 10 microsatellite markers. It was detected a 50 alleles with an average of five alleles per locus and alleles size between 180 and 380 bp. Further were calculated statistical parameters DI, PI and PIC, which confirmed the high value of polymorphism of used SSR markers. Dendrogram was compiled, identifying the relationships between different varieties of carrots. The results were compared with scientific publications dealing with the same or very similar problems.

This thesis includes the study of the literary as well as practical aspects of DNA variability in selected species of Apiaceae family. Among the objectives were also an examination and a verification regarding the transferability of microsatellite markers designed for carrot and cumin to related species. The practical part was elaborated by using methods of DNA isolation, PCR amplification and electrophoretic separation on both agarose and polyacrylamide gel. Using a total of 13 SSR markers in 15 varieties of various representatives from the Umbelliferae family (carrot, celery, parsley, parsnip and dill), 98 alleles of the size differing from 140 to 400 bp have been found. Then, binary matrices were compiled and characteristics for each of the markers used were calculated, the values of which demonstrate high utility for the study of genetic variability in all species except dill. Based on the results, the dendrogram was developed, indicating the close and distant relationships between individual varieties and sorts of crops. The results were compared with similarly focused expert studies.

Bachelor thesis is focused on description of important content substances in Daucus carota L. Experimental part is focused on monitoring and evaluating of content substances in nine picked cultivars: 'Kardila', 'Rubína', 'Karkulka', 'Jolana F1', 'Kráska', 'Katlen', 'Jarana F1', 'Koloseum F1', 'Jitka F1'. Vitamin C, dry matter, antioxidants, minerals, refractometric dry matter, nitrates, carotenoids and roughage were determined. Aim was to summarize all results from experiment and compare them with other literary sources. The best results had 'Kráska' variety and the worst had 'Koloseum F1' variety. According to these results, it is recommended to continue in research.
