Possible association of polymorphisms in COMT gene with mental diseases

This thesis i focused on associacion of polymorphisms in COMT gene with mental diseases, primarily on polymorphism Val158Met. The theoretical part provides definitions of COMT gene and enzyme, catecholamines affected by COMT and definition of the polymorphism Val158Met. Next is listing of mental diseases, description and known connection with researched polymorphism. The practical part contains proceeding of PCR RFLP used to detect the genotype. The results were evaluated and connected to the theoretical part of the thesis.

Optimization of the PCR-RFLP method for the Val / Met polymorphism analysis in the COMT gene.

The topic of this bachelor thesis is the optimalization of the method PRC-RFLP for the analysis of polymorphism Val/Met in the gene COMT. The first goal of this thesis was to familiarization with factors of congenital risks for schizophrenia. The theoretical part is devoted to this matter, in which the disease of schizophrenia is defined. In the First part the history and etiology of this disease is mentioned. Then the clinical signs of schizophrenia, the diagnostics and threatment of the disease. The important part is also the explanation of the connection between the schizophrenia and polymorphisms in the gene COMT and the role of dopamine connected with it. Next to this issue, the connection of the onset of schizophrenia with other non-genetic factors is being explained. In the next chapters the function of the gene COMT and the role of the enzyme COMT is being described. Other chapters are devoted to the polymorphism val158/met in the gene COMT and the methods appropriate for detection of this type of mutation. Primarily the methods used in the practical part of this bachelor thesis are being developed. I have set as other goals of my bachelor thesis the optimalization of the method PRC-RFLP for the examination of polymorphisms Val/Met in the gene COMT, which are increasing the risks of schizophrenia and also the analysis of several samples of individuals with the incidence of schizophrenia in the family. The practical part of this thesis is dedicated to these goals, in which the above procedures, that helped me to archive my results, are being described. I have used the method PRC-RFLP and also the method ARMS-PCR. The suitability of the use of these methods is discused in the last part of this bachelor thesis.
