Biosensors based on modified nanoparticles

This thesis deals with biosensors based on modified semiconductor quantum dots(AIIBIII). This work is divided into three main parts. The first deals with the theory necessary for the use of QDs in bioapplications, there are described methods of synthesis, application and bioconjugation of QDs . In the practical part, GSH-CdTe, CdTe-MPA and TGA-CdTe quantum dots were prepared by direct colloidal synthesis. These quantum dots were then modified by EDC in order to increase affinity to BSA (bovine serumalbumin). For the analysis of samples were used Infinite M200,Tecan and HORIBA Jobin Yvon FluoroLog detector Quanta . The third part is the summary of results and work achievements.
Synthesis of core/shell quantum dots for diagnostics

This thesis deals with biosensors based on modified semiconductor core/shell quantum dots (QDs) for diagnosis. The work is divided into four main parts. The first one discusses the theory required for the use of QDs in bioaplications, there are described methods of synthesis, modification, application and bioconjugation of QDs. In the experimental part, CdTe/ZnS QDs with core/shell structure were prepared, in which the core was modified by MPA, GSH and TGA. In the next step, these QDs were further modified using CDI, EDC and NHS as mediators in order to increase affinity to BSA (bovine serum albumine) and IgG (imunoglobuline G). Prepared conjugates were characterized by fluorescence spectroscopy (Infinite M200Pro, Tecan) and capillary electrophoresis (Agilent 7100).
Ageing of asphalt binders and mixtures

Diploma thesis is particularly focused on ageing of asphalt mixtures and asphalt binders. In the theoretical part an array of methods simulating short term and long term ageing consequently is introduced. The main objective of the practical part was to carry out the long-term ageing test method BSA (Braunschweiger Alterung). Thereafter, specimens testing for stiffness properties according to CSN EN 12697-26 were made. After conducting the measurement on those, asphalt binder was extracted and processed to follow-up testing. The testing consisted of a range of methods – Penetration CSN EN 1426, Ring and Ball test CSN EN 1427, Determination of the Frass breaking point CSN EN 12593, Determination of complex shear modulus CSN EN 14770 and Dynamic viscosity using a dynamic shear rheometer (DSR). The given results were assessed in the last part in order to evaluate the rate of the binder degradation caused by the method BSA.
Effects of ultrasound on biological materials

In this bachelor thesis I deal with effects of ultrasound on biological materials. In theoretical part is write up a literature search about effects of ultrasound on bimacromolecules namely deoxyribonucleic acid (DNA), proteins (BSA) and bacterial cells. Effects of ultrasound was investigated practically on genetically modified bacterias of Escherichia coli. These bacterias were sounded by ultrasound by various frequences for the same time and then their viability was measured on luminometer. The results for ever frequency were compared.

AFM study of FBS, BSA, Fn proteins adsorbed on H-/O-diamond

Cell growth indicates that Fibronectin is behind micro-array formation. Using AFM we measured conformations of particular proteins from Fetal bovine serum in solution and in air. AFM data are not convincing.
