Testing the synteny of genes in \kur{Yponomeuta evonymella}

The aim of this thesis was to create a physical map of the bird-cherry ermine, Yponomeuta evonymella, chromosome Z1 and chromosome 15 using BAC-FISH mapping. The synteny of genes was compared to the closest outgroup of Yponomeuta spp., Plutella xylostella, in order to determine whether the P. xylostella genome sequence could be used as reference in reference-assissted genome assembly of Yponomeuta spp.
Neo-sex chromosomes and adaptive potential in tortricid pests

Although most of moths and butterflies possess high number of small undifferentiated chromosomes, in the family Tortricidae extraordinary large Z chromosome has been observed. In this study, the physical mapping of this sex chromosome in the major pome fruit pest Cydia pomonella (Tortricidae) revealed a fusion between an ancestral Z chromosome and chromosome 15 in Bombyx mori reference genome. Furthermore, the sex linkage analysis by qPCR has shown that the rearrangement originated in common ancestor of tortricid subfamilies Olethreutinae and Tortricinae. As the Bombyx mori chromosome 15 contains several genes involved in detoxification pathways and insecticide resistance, this fusion could have facilitated adaptive evolution and radiation of tortricid pests.
Structure and evolution of plant sex chromosomes: The use of BAC library

Sex chromosomes of plant species Silene latifolia are the ideal model for evolutionary biology studies because they represent early stages of X and Y chromosomes history. We used fluorescence in situ hybridization (FISH) with bacterial artificial chromosome (BAC) clones in studies of chromosome structure to demonstrate that a majority of genes are located in subtelomeric regions of all chromosomes. Our results also indicate the absence of accumulation of main classes of repetitive sequences - retroelements and centromeric tandem repeats - in non-recombining parts of the sex chromosomes.
