Odběr spermatu pomocí katetru a jeho využití při výtěru štiky obecné (Esox lucius L.)
PLAŇANSKÝ, Tomáš
The aim of this diploma thesis is to compare quality of northern pike sperm collected by different methods. First method is collection of stripped sperm by abdominal massage of the belly. Second method is collection of stripped sperm with special catheter to eliminate sperm contamination by urine. The last method is collection of testicular sperm. Differently collected sperm was evaluated and compared its quality. The main observed parameters were sperm volume, spermatozoa concentration, spermatozoa motility and velocity and osmolality of seminal fluid. Sperm samples were used for eggs fertilization. In fertilized eggs, the fertility of eggs and larvae hatching rate were observed.
Hormonal stimulation of ovulation of grass carp (Ctenopharyngodon idella)
MUSIL, Martin
Hormonal stimulation of ovulation in grass carp (Ctenopharyngodon idella) In 2015, an experiment with hormonal stimulation of ovulation in grass carpwas performed using dopamine inhibition of secretion luteinizing hormone (LH) by mGnRHa ([D-Tle6, Pro9, NEt] -mGnRH), with or without metoclopramide. Three experimental groups of females were injected with the following hormonal preparations: group A - mGnRHa (12.5 mikrog.kg-1) + metoclopramide (20 mg.kg-1), group B - mGnRHa (12.5 mikrog.kg- 1) Group C - 0.9% NaCl. For group A injected mGnRHa with metoclopramide, ovulation only achieved one of the females (14.28%), the fecundity index was 5.66% and the fertility was only 0.1%. The luteinizing hormone concentration peaked after application of the hormonal preparation and, unlike group B, declined with imminent ovulation. In Group B, the application of pure synthetic mGnRH induced ovulation in 57.14% of injected females. Fecundity index was 11.25 +- 4.77% and average fertility reached 60.25 +- 11.22%. The level of luteinizing hormone reached its peak only at the time just before ovulation itself. The results of the experiment did not demonstrate the use of mGnRHa with metoclopramide as the most effective. However, LH levels have shown a strong dopamine inhibition of LH secretion in this species.
Effects of hormonal treatment during artificial reproduction to produce to fingerlings of weatherfish (Misgurnus fossilis)
HOUDA, Ondřej
In this M. Sc. thesis, the influence of selected commercially available hormonal preparations based on hCG (Pregnyl, Chorulon) and GnRH-a with dopamin inhibitor (Ovopel, Dagin, Ovaprim) and their benefits were studied, compared with homogenate of carp pituitary (CPE), during controlled reproduction of weatherfish (Misgurnus fossilis, Linnaeus, 1758) in artificial conditions on the quality of the produced fry. The goal of this thesis was assess the suitability of individual commercially produced hormonal preparations for controlled reproduction of weatherfish and the possibility to replace with them unstandardized use of homogenate of carp pituitary. It was found that use of various hormonal preparations and dosage may to in some extent affect hatchability and survival rate of larvae (fry), the size of the yolk sac of larvae, and the amount of energy deposited in body tissues of larvae. However, demonstrable difference in studied parameters for the application of different preparations on the different hormonal basis (hCG versus GnRH) was not reported. The highest rate of hatching larvae (93.3 ? 3.8%) was observed in the groups treated with preparation Pregnyl 3000 IU - kg-1. In terms of overall survival of the larvae (from deployment of hatched eggs until the end of the experiment) have proven to be the most effective medication Pregnyl 3000 IU - kg-1 (69.9 ? 12.1 %) and Dagin (60.56 ? 11.71 %). Within the thesis has been demonstrated no relationship between hormonal treatment used and the growth of larvae (in terms of achieved total length of body and dry weight). The amount of energy deposited into the body tissues of larvae (gross calorific value) was comparable in all groups, except Dagin preparation, which was recorded impairment losses. For the best, most effective and commendable for control of reproductive hormone preparation carp pituitary homogenate replacing the terms of the assessed indicators can be seen the preparation based on hCG and that is Pregnyl at a dose of 3000 IU - kg-1.
The Questions of the Beginning and the End of Human Life
ČADOVÁ, Marie
This work deals with the problems of conception, gravidity and abortion in the first part and in the second part than deals with problems of death and dying. It concerns two cut-off points, which are component parts of human life. The first part surveis the biginning of human life and tries to find answers for questions, which have a connection with the conception, artifical insemination, surrogative maternity and abortion. It describes paradoxes, which comes from such situations. It appreciates this paradoxes in the wiev of medicin, ethics, psychology and law. The second part describes the end of the human life in several forms. It broods killing, suicide and euthansia in aspects of the psychology, medicine, law and ethics. In the end it describes the palliative care as an alternativ to euthansia.
