Comparison of differential leukocyte count set by the analyzer Beckman Coulter LH 750 and by microscope in the group of pediatric hemato- oncology patients

In my thesis I focused on comparing a differential leukocyte count (DIF) determined by the analyzer and by microscope in the group of pediatric oncological and hemato-oncological patients. Thesis is divided into theoretical and practical parts. The theoretical part includes an explanation of the process of hematopoiesis, the development of blood cells and the characteristic of time series, a decription of hemato-oncological and oncological diseases which occur most frequently in the monitored group of pediatric patients, and their treatment. In a practical part is described a determination of the DIF by the analyzer Beckman Coulter LH 750 and by microscope Meopta. I measured a blood count with the differencial leukocyte count, I made a blood smear and I evaluated microscopically it, in the selected group of pediatric patients. The results were reported in the tables and statistically processed using paired t-test in GraphPad Prism and were created graphs. An aim of this thesis was to explore and evaluate further the differences between the two methods. In 19,32% of patients in the test group was not evaluated the DIF by the analyzer . Thesis is mainly focused on laboratory diagnosis of inherited types of anaemia. Anaemia or anaemia syndrome is a pathological condition characterized by low levels of haemoglobin and hematocrit under the physiological limit. Clinical manifestations are variously severe depending on development of disease and type of anaemia. Anaemia patient suffer from reduced oxygenation of tissues which determinates many symptoms of tissue hypoxia. Other symptoms are associated with hypovolemia and hyperkinetic circulation that can occur. In haemolytic anaemia, patients have symptoms of intravascular or extravascular haemolysis. Title: Study of new prognostic markers in patients with chronic lymphocytic leukemia Chronic lymphocytic leukemia (CLL) is the most common leukemic disorder of adults in Western hemisphere. It is characterized by clonal proliferation and accumulation of morphologically mature lymphocytes in bone marrow, peripheral blood and lymphatic tissues. Clinical course of CLL is extremely heterogeneous with some patients living for decades without need of therapy while others succumbing to the disease within several years. Thus, there has been great interest in identifying prognostic markers that could be used to distinguish patients with an aggressive form of CLL, because they might benefit from early intervention. The process of angiogenesis has been shown to be crucial for growth and metastasizing ability of solid tumors. Angiogenesis in "liquid" tumors was supposed to be less important, nevertheless numerous recent studies have shown enhanced angiogenesis in many hematological malignancies including CLL. Ilona Fátorová Title of diploma thesis: Possibilities of platelet count evaluation by analyzer Sysmex XN in a group of patients with thrombocytopenia This diploma thesis deals with the problems of platelet count evaluation in thrombocytopenic patients. There are several principles of determination of platelet count, which are currently used. The aim of this thesis is to assess and analyze three of them (impedance, optical and fluorescence method) in 400 patients. The theoretical part includes topics like function and development of platelets, principles of their count determination and pathology associated with low platelet count. In the experimental part the obtained data is graphically and statistically evaluated to analyse the agreement between three measurements. Methods: Analysis of complete blood counts was performed by the analyzer Sysmex XN-10. Platelet counts were obtained from PLT-I, PLT-O and PLT-F methods. Some blood samples required a microscopic control of platelet morfology. Results: The obtained data was statistically evaluated with help of Wilcoxon test and Bland- Altman plots.
