Separation and purification of nucleic acids with the use of magnetic microparticles

Application of magnetic microparticles for nucleic acid isolation from real samples is a modern and fast separation procedure. This method is suitable for application in laboratory practice. The aim of the bachelor work was isolation of DNA from a dairy product (Activia sweet white) in the PCR-ready quality. DNA isolation was carried out by magnetic microparticles P(HEMA co GMA) in the presence of 2 M NaCl and 16% PEG 6000 or 8% PEG 6000. According to the declaration of the producer the presence of DNA from probiotic bacteria of the genera Lactobacillus and Bifidobacterium was confirmed by PCR.
Partial slip in liquids studied with high-frequency shear oscillations

Byl studován kontakt mezi koulí a deskou pod tangenciálním zatížením ve vodě. Jako zdroj střižných kmitů byl použit akustický rezonátor (křemíkové mikrováhy – QCM). Kontakt koule s povrchem resonátoru indukuje změnu resonanční frekvence a šířky pásma. Byla měřena změna frekvence f a změna šířky pásma v závislosti na amplitudě oscilací. S rostoucí amplitudou docházelo k poklesu f a růstem , což je chování typické pro parciální skluz. Díky aplikaci Cattaneo-Mindlinova modelu byl vypočítán kontaktní poloměr a třecí koeficient. Kontaktní poloměr při nízké amplitudě stoupal při zvětšujícím se normálovým zatížením. Tato závislost se dobře shodovala s JKR modelem. Třecí koeficient se nacházel v odpovídajícím rozsahu. Při zvyšování externí normálové síly, docházelo k nepatrnému snižování hodnoty třecího koeficientu. Toto chování je vysvětleno příspěvkem adhezivních sil k totální normálové síle. Výpočtem byly získány dva typy třecích koeficientů, první ze změny frekvence f a druhý ze změny šířky pásma . Tyto dvě hodnoty se spolu shodovaly z ± 20 % pro měření prováděná ve vodě, zatímco pro dvě měření prováděných na hydrofilním povrchu ve vzduchu se lišila. Tento nesoulad poukazuje na nedostatek Cattaneo Mindlinovy teorie a mohl by být vysvětlen přítomností kapilárních sil.
Historical collection of rare books in the Municipal Museum of Ústí nad Labem and assessment of its presentation

This bachelor thesis deals with the subject of presenting The Municipal Museum of Ústí nad Labem and its library assessing the accessibility of historic and rare prints that can be found within its premises. Firstly it introduces historical book collection of Ústí museum library and illustrates various possibilities how to present it to the public. Next part compares the access of historical books to the public among both museum libraries in Ústecký region as well as main museum libraries in every region of the Czech Republic. The practical outcome of this thesis is the recommendation how to become more effective in presenting historical book collections in museum libraries
Documentation of the use of personified virtues ornamental bands in the prague printing offices of the time of Emperor Rudolf II

The bachelor's thesis on the motif of allegories of Christian virtues in the form of female figures of Faith and Hope, Love and Justice, Wisdom and Temperance, Courage and Patience presents a mapping of the occurrence of the theme on ornamental bands in printing oficces in the time before the battle of Bílá hora. It is based on the premise of a traceable pioneering line of ornamental bands with the Virtues motif from the workshop of Michael Peterle Sr, inspired by the production of Melantrich and other collaborators and followers. It represents the popularity of the motif in art in general, with a subsequent narrowing to old prints and consideration of manuscript production. Includes a broader historical perspective and period context. The intention of the work was to evaluate the originality and artistic treatment of ornamental bounds and to contribute to a closer understanding of the transfer of decorative elements used by Czech early modern printers. The thesis is mainly prepared by the method of creative compilation of knowledge obtained through the study of relevant sources and historical analysis. The thesis introduces a brief historical development of the imaging and the stereoscopy itself, basic biological and optical characteristics of a human eye and vision and the principles of stereoscopic projection. Furthermore it occupies with the procedures of making stereoscopic drawings and photographs along with the methods of their observation and creating some necessary tools using generally available materials. The end of this thesis is devoted to the possibilities of not only practical usage of the stereoscopy, but mainly of its application in the descriptive geometry teaching. The thesis includes many stereoscopic pictures, some of them are viewable with the lens- glasses or the anaglyph-glasses accompanied in the end of this thesis. Ilustrator Virgil Solis Jr. In the service of czech printed Book

This diploma thesis explores the life and the work of the painter and illustrator Virgil Solis Junior (1551 - 1617?). Virgil Solis Junior was the son of the renowned German graphic artist Virgil Solis Senior. In 1567 Solis Junior came to Prague. Firstly, the thesis provides information about Solis's life and position among visual artists in Prague of the time. Further, it describes his potential professional involvement at the court of the Emperor Rudolf II. Virgil Solis was also an illustrator. Drawing from literature published so far, it seems that his only illustrated book was Diadochos, printed in 1602. This thesis, however, supposes that Solis worked on other illustrations during his stay in Prague. It explores and compares various woodcuts from the 16th and 17th centuries and thus compiles a list of printed volumes where Virgil Solis was the author of drawings or woodcuts. This thesis brings an interesting insight into the life of an artist living and working in the Prague of Rudolf II.
Bookbinding databases

The aim of this thesis is to analyze and compare bookbinding databases, such as Einbanddatenbank, ProBok, databases of the Bavarian State Library, University and City Library of Cologne and others, including several projects thematically focused on provenance research. First, the thesis will introduce the topic of historical bookbindings and provenance marks in general, and then compare individual databases, which will focus on their content, operating institutions or technical solutions as well as the relevance of these databases to Bohemian bookbindings research. A comparative analysis of individual projects based on checklist will be the method used for the practical part of the thesis. Keywords Databases, provenance, bookbinding, collections, digitization.

Book World Prague - 25 Years of the International Book and Literary Festival in the Context of the Sociocultural Changes in the Czech Book Trade

(in English) This diploma thesis focuses on the Book World Prague - International Book Fair and Literary Festival - in the context of the transformation of the Czech book market after 1989. Particularly, it aims to examine the role the Book World Prague played in rehabilitation of the Czech book market as for re-implementation of basic market mechanisms in book production and distribution. Individual chapters cover the following topics: characterization and typology of modern book fairs, the history of the Czech book market from 1948 the present day, the history of the origin of trade fairs, the specific history and developments of book fairs, and the milestones in the history of the Book World Prague. Central to the research are data collected by the author by interviewing ten professionals working in various fields of the book industry. The transcribed interviews served two major purposes: firstly, they were used as the primary source enabling the author to answer the research question. Secondly, they created a primary source material relating the history of the Book World Prague.
