Establishment of an object is made of concrete foudations pass and quiffs.
Conservation genetics of the grey wolf in Central Europe
Valentová, Kamila Anna ; Hulva, Pavel (advisor) ; Galov, Ana (referee)
Conservation genetics of the grey wolf in Czech Republic and adjacent regions is studied in the present thesis. Analyses of twenty-one microsatellite loci, one sex-determining amelogenin gene and mitochodrial control region were used to verify species determination, identify individuals and estimate relationships between them, analyse population structure and estimate demographic trends based on samples collected between 2014 and 2021. Genetic detection of red fox and dog samples incorrectly assigned to wolves illustrates the hurdles of field monitoring of grey wolf. Direct evidence for the occurrence of F1 hybrids was not found. Wolves from Bohemia showed lower values of allelic richness in comparison to the ones from Western Carpathians, probably as a consequence of recent expansion. Geographic distances between detection sites of identical individuals were relatively small or moderate in this study, suggesting regular movements of animals within their home ranges. Only two long-distance dispersal events exceeding 300 km were detected. Results of parental analysis provided evidence of pack distribution within the studied area. Most relationships were detected between wolves in the northern region of Czech Republic where the first recolonizing wolf pack in 2014 was registered. Within the studied...
Testing the applicability of hacker typologies and models: A comparative case study of Fancy Bear and The Shadow Brokers
Valentová, Anna ; Hynek, Nikola (advisor) ; Střítecký, Vít (referee)
The Master's Thesis named "Testing the applicability of hacker typologies and models: A comparative case study of Fancy Bear and The Shadow Brokers" focuses on different categorization techniques of hacker groups. It explains how hackers are studied in the field of social sciences and theoretical and practical cyber security. The thesis aims to test the disciplines and applicability of their typologies and models on two cases - The Shadow Brokers and Fancy Bear, hacker groups representing a novel type of threat actor. Firstly, the theoretical section focuses on explaining typologies and models while also representing the trends and evolution of research on hackers. Secondly, a practical part of the thesis picks a few frameworks to be tested via the two cases. After the testing, the virtues and shortcomings of the frameworks are analysed, and the approaches of disciplines are compared. The practical part of the thesis shows there is no unified approach to studying hacker groups and almost all previous ones are not applicable to the two hacker groups cases. Therefore, based on the results, the dimensions for a new typology are proposed with the objective of creating a stepping stone for an applicable approach to studying hacker groups in security studies research.
Factors important for Cajal body formation
Roithová, Adriana ; Staněk, David (advisor) ; Valentová, Anna (referee)
This research describes the structure and function of nuclear domains called Cajal bodies (CB). CB contain proteins and factors involved in assembly and modification of snRNPs. These bodies are found in vertebrates and invertebrates and even plants. Not all cell types contain CB. Their number and size depends on the transcription activity of cell and cell cycle phase. This paper discusses the factors that affect the CB formation. One of the most important factors is the level of snRNPs and transcription activity. Recently shows that an important role in CB formation has coilin and other components phosphorylation. Other works show the influence of the environment. There is also discussion regulation of CB biogenesis, witch is not yet fully understood. Key words: Cajal bodies, coilin, cell nucleus, snRNP, pre-mRNA splicing, transcription
The relationship between chromatin organization and RNA splicing
Icha, Jaroslav ; Staněk, David (advisor) ; Valentová, Anna (referee)
It is well known that RNA splicing and other pre-mRNA processing reactions happen cotranscriptionally. Surprisingly, there were recently discovered some chromatin features that had uneven distribution between exons and introns, which directly links chromatin organisation to splicing. This work summarizes all the studies that detected these chromatin patterns on exons and discuss their inconsistencies. In these studies nucleosomes were found to be preferentially positioned on exons, specific histone modifications and DNA methylation were also enriched on exons. These local patterns of chromatin organisation were evolutionarily conserved from mammals (human and mouse) to worm C. elegans and fly D. melanogaster. Their findings indicate that the role for chromatin structure in pre-mRNA splicing is to promote exon recognition. There are two mechanisms proposed for this role of chromatin in splicing. The first one is influence on RNA polymerase II elongation speed, and the second is specific recruitment of splicing machinery. In the near future we can expect studies searching for concrete examples of these two mechanisms and assessing their significance. Indeed it was reported very recently that H3K36me3 regulates alternative splicing via the second mechanism.
Genetic interactions of PRP45 mutantion in S. cerevisiae
Valentová, Anna
Human SKIP/NcoA-62/SNW1 ortholog proteins are nuclear factors which have been found in connection with the regulation of gene expression and subsequent modifications of primary transcripts. Human representative SKIP acts primarily as a transcription factor during the processes involving differentiation, carcinogenesis and other cellular signaling. It operates in the signaling pathways, eg TGFß/Smad, E2F/pRb, Notch/CBF1 and nuclear hormone receptors. SKIP protein is also essential for pre-mRNA splicing and recently its contribution during the elongation phase of RNA polymerase II transcription and contribution to some HIV genes alternative transcripts formation has been identified. The ortholog of S. cerevisiae FUN2O/Prp45 has been shown to participate in pre-mRNA splicing, others processes mentioned have not yet been associated with the function of this factor. Pre-mRNA splicing is a complicated process leading to the formation of native mRNA lacking non-coding regions, introns. In terms of composition of the spliceosome subunits and biochemical principle of its function is this process very well described but despite this knowledge the specific roles of most splicing factors are not known. For Prp45 protein direct interaction partners were identified; these partners derive some of their possible role in...

Recommended videos on YouTube and the problem of information bubble
Valentová, Anna ; Šlerka, Josef (advisor) ; Wančová, Nina (referee)
The topic of this bachelor's thesis is the function of the reccomendation algorithm on the YouTube platform and its preferential treatment of misinformation and alternative content over reliable sources. It focuses on Czech-language YouTube. This thesis is also concerned with the theory of the filter bubble, to which this topic is also connected. The main question posed in this bachelor's thesis is whether the algoritm purposely encloses the user in the filter bubble or not. Executed research follows the method of gathering recommended videos propagated by Guillame Chaslot, and focuses on the analysis of the data acquired. The results, presented in charts and accompanied by sufficient commentary, are then compared and critically evaluated. I believe that the results will prove valuable for future research into this topic.
