Utilisation of electromigration methods in analysis of naturally occuring compounds

UTILISATION OF ELECTROMIGRATION METHODS IN ANALYSIS OF NATURALLY OCCURING COMPOUNDS Jan Honegr ABSTRACT A transient isotachophoresis-capillary zone electrophoresis (tITP-CZE) method has been developed for pre-concentration and determination of nine analytes: caffeic, chlorogenic, o- coumaric, p-coumaric, ferulic and protocatechuic acid, and kaempferol, quercetin and rutin. The effects of several factors such as control of EOF, pH and concentration of the running buffer, addition of organic solvents and their concentrations, addition of cyclodextrins, the duration of injection and the λ max of UV detection were investigated to find the optimum conditions. Under these conditions, the analytes were separated within 15 min. Linearity was evaluated for concentration range 2.5-37.5µg/ml with R = 0.9924-0.9983; the detection limits (S/N 3:1) ranged from 203ng/ml to 5.556µg/ml. The relative standard deviations of the migration times (peak areas) were between 0.79 and 1.01% (1.34 and 2.13%).

8 SHRNUTÍ Tato práce představuje možnosti využití tří účinných separačních technik založených na elektromigraci analytů, jako jsou kapilární izotachoforéza (ITP), kapilární zónová elektroforéza (CZE) a micelární elektrokinetická chromatografie (MEKC), v analýze farmaceuticky významných látek a to jak syntetického původu v matrici léčivého přípravku, tak především přírodního původu v extraktech léčivých rostlin resp. rostlinných silicích. V teoretické části je věnován poměrně velký prostor popisu těchto metodik a zejména jejich širokému uplatnění ve farmaceutické analýze přírodních látek (tabulky 1, 2 a 3) a dále možnostem ovlivnění separace elektricky neutrálních analytů vlivem přídavku komplexujícího činidla či povrchově aktivní látky do základního elektrolytu. Součástí práce je také přehledná rešeršní publikace (příloha I), shrnující uplatnění elektromigračních metod v kontrole kvality lékopisných rostlinných drog. První experimentální práce (příloha II) ukazuje na možnost použití kapilární izotachoforézy ve farmaceutické analýze ke kontrole obsahu léčivé látky. Stanovení významného sympatomimetika tetryzolinu a antihistaminika antazolinu v hromadně vyráběných nosních a očních kapkách ukazuje, že vyvinutá, optimalizovaná a validovaná ITP metoda poskytuje přesné a správné výsledky. Methodology of jazz drumming

The aim of the thesis is to analyze the methodical procedure of teaching percussion instrument performace in the context of jazz music. The absence of a similar work - in the domestic area, which would summarize the basic, yet crucial practices in a comprehensible whole was one of the main inspirations for this thesis. The first chapter describes the context and the process of formation of the drum set. The next chapter introduces the notation of jazz music and the way it is different from phrasing in practice. The next chapters deal with the various approaches to playing the ride cymbal, hi-hat and the bass drum. These chapters contain practical exercises. Next I point out the importance of drums as an accompanying instrument. The chapter on transcription tells readers how to handle new material and how to be able to work with it further. In the seventh chapter I present coordination exercises, which I have completed and compiled with this work. The last chapter deals with the analysis of Jim Chapin's Advanced Techniques for the Modern Drummer.
The food of pikeperch (Sander lucioperca) fry in ponds with different kinds of management

The aim of this study was a comphensive assessment of the food of pikeperch fry in the monoculture rearing from the stage of summer fry to the stage of autumn fry in ponds with three different types of management and assessment of the influence of individual treatments for pikeperch production. The experiment was performed in small ponds in experimental facility of FFPW in Vodňany. The variants of pond treatment were: a) ponds with additional prey fish (fry of common carp (C. carpio) and grass carp (C. idella)), b) ponds with installed substrate from heather, c) control variant without any support of natural food of pikeperch. All treatments had four repetitions. Rearing of pikeperch fry lasted 97 days. In all treatments, the opportunistic food strategy of pikeperch fry was recorded during the whole period of rearing and the main recorded food was macrozoobenthos, especially insect larvae. Bigger species of zooplankton had also been often found in the stomachs of pikeperch. Considerable development of macrozoobenthos in all experimental ponds was probably caused by the presence of submerged macrophytes. Thus, this factor made harder to estimate influence of installed substrates from heather for production of pikeperch fry and composition of its food. In the treatment with prey fish, the total length (F (9. 450) = 91.1; p < 10-3) and weight (F (9. 450) = 61.9; p < 10-3) of pikeperch fry were for the whole period of rearing significantly higher in comparison to the other treatments. In addition, the survival of pikeperch in the treatment with prey fish reached to 56 - 92,4 % at the end of rearing. In the treatment with substrate from heather, the total length and weight of pikeperch was significantly higher in comparison to the control treatment only in the first term of sampling. In the rest of pikeperch fry rearing period were the values comparable or higher in control treatment. Thus, for the rearing of pikeperch from the stage of summer fry to the stage of autumn fry is appropriate to stock prey fish in ponds. In comparison to installation of artificial substrate, the diversity of water environment, which is subsequently able to provide sufficiently wide natural food for reared pikeperch, seems to be more decisive.

The effect of periphyton on the development of macrozoobenthos in ponds

The aim of this study was to evaluate the effect of two types of artificial substrate enhancing periphyton development on macrozoobenthos community in ponds in which larvae of pikeperch (Sander lucioperca) were cultured to advanced fry. The experiment was divided into two time periods and was performed in twelve ponds in experimental facility of FFPW in Vodňany. In the first time period of the experiment (from the end of April to the beginning of June) there were three different treatments of the ponds established: a substrate from heather, a substrate from geotextiles and a control without any substrate. Each treatment had four repetitions. In the second time period of the experiment (from the middle of June to end of September) the substrate from geotextiles was removed and following options of the pond treatment such as the substrate from heather, releasing of prey fish into the four ponds and no support of periphyton (a control) were applied. In the course of both experimental time periods samples of substrate and bottom macrozoobenthos were taken in ponds each 3 weeks. According to statistical analysis, significantly higher abundance of macrozoobenthos was found on the substrate from heather in comparison to substrate from geotextiles (F (1, 6) = 12.897, p = 0.011). Mean values of the species abundance and the species biomass were always higher in the bottom macrozoobenthos than in the substrate macrozoobenthos. Mean values of diversity index were always higher in the substrate macrozoobenthos than in the bottom macrozoobenthos. The effect of the pond treatment on development of the bottom macrozoobenthos was not confirmed. The influence of the geotextiles on enhancement of fish production was rather negative. The substrate from heather increased the production of advanced pikeperch fry at 7 - 21 %, in the first time period of the experiment. Thus, next further research in this field can be beneficial regarding effective culture of some important fish species.
