Methods of storage of honeybee semen: evaluation of honeybee spermatozoa viability with flow cytometry.

Assessment of semen quality (sperm viability) is an essential for the efficient honeybee semen storage. The motility of spermatozoa is not the ultimate parameter of sperm viability. Instead, the viability of spermatozoa is assessed by testing their structural or functional plasma membrane integrity. These assays are based primarily on the technique of microscopy evaluation of either fluorescent DNA dyes or hypo-osmotic "tail-coiling" reaction of spermatozoa. However, the technique of microscopy evaluation is time-consuming, subjective and often is not accurate. On the other hand, the technique of flow cytometry is rapid, objective and precise. Nowadays, this technique is affordable for many institutions, as the price of "benchtop" flow cytometer is relatively low. To best of our knowledge, there is currently no flow cytometry assay available for evaluation of functional integrity of honeybee spermatozoa. Here we report the establishing of novel flow cytometry assay for simultaneous evaluation of structural and functional plasma membrane integrity. DRAQ7, the new fluorescent DNA dye was used to test the structural integrity of plasma membranes. The hypo-osmotic "tail-coiling" reaction was used for flow cytometric assessment of functional integrity of spermatozoa plasma membranes. The combination of both is a valuable tool for rapid and precise evaluation of sperm viability in honey bees.
Causes of colony losses

This bachelor thesis is focused on the causes of bee colonies losses. In the last years the number of dead bee colonies increased significantly. Especially in the years 2013 and 2014, which this paper deals with, losses were really alarming (on the nationwide average up to 30%). The theoretical part of the paper firstly presents beekeeping in general. Then, the thesis deals mainly with diseases and parasites invading bee colonies. Lastly, the paper describes hygiene and good breeding practice, which helps to prevent diseases and which should be obvious to every beekeeper. The reason for this is the fact that by not following good breeding practice the beekeeper does not threaten not only his/her own breeding, but also the breeds of neighbouring beekeepers. The aim of the thesis is to compare beekeepers practice with the help of the data collected in questionnaires. The questions concern the varroasis disease, which afflicts the beekeepers in this country most often and which is (according to the latest researches) one of the main causes of bee colonies losses. Fifteen beekeepers filled in the questionnaire. None of them (with one exception) had great losses of bee colonies. Eleven respondents noted losses within 10%. The greatest losses noted the beekeeper owning the highest number of bee colonies. The beekeepers admitted one serious mistake, not following the information leaflet of medical products, which may have decreased the effectiveness of treatment. None of the respondents had removed the sealed brood before fumigation and aerosol treatment was done. Therefore not all mites in the hive could have been exterminated.
Database Design for Bee Research Institute

The goal of the bachelor thesis is to design a database for the customer Bee Research Institute, Ltd. according to his requirements and specifications. The next step is to create a functional application in Microsoft Office Access 2007 using created design. The thesis is divided into two parts. The first part concerns the theory of database design and the second part deals with the procedure for designing a database for the Bee Research Institute according to the methodology MMDIS. The thesis includes creating a database application with a user interface in Microsoft Access.
