Determination of mtDNA sequence variation in czech population for the usage in forensic genetics.

In forensic research and practice is used an analysis of nuclear DNA commonly. However, in some cases we can use only mitochondrial DNA. The mtDNA is maternally inherited and it is located in the each cell approximately in 500 copies. Barring mutation in special noncoding segments, the mtDNA sequence of siblings and all maternal relatives is identical. This unique haplotyp can be helpful in forensic cases, such as analyzing the remains of a missing person, where known maternal relatives can provide reference samples for direct comparison to the questioned mtDNA type. MtDNA is also very resistant to the external influences, so in the cases where the amount of extracted DNA is very small or degraded it is more likely that a DNA typing result can be obtained by typing mtDNA than by typing polymorphic markers found in nuclear DNA. If we want to have a certain probability to exclude or confirm the match of two samples, classify suspect or missing person into a population, we need some enough wide comparison by the population databases. Then we can study the frequencies of different haplotypes in particular population. Tracing the fate of cell populations from regressive tooth primordia during ontogenesis

(v anglickém jazyce) Development of tooth primordia in mice is an important model for study of odontogenesis. Several dental rudiments develop during the mouse embryogenesis. These structures develop in functional teeth in their phylogenetically older relatives. Similarly, we can initiate growth of teeth from these germs in some mutant mice. In my diploma thesis we have focused on the importance of rudimentary structures with odontogenic potential in postnatal individuals. As a model of development, we have chosen a cell population originating from rudimentary primordia MS (mesial segment) that develops in diastema of the lower jaw during the embryonic day 12.5. Using the inducible Cre-lox technology we have marked the cells which are part of the signal domain of primordia at this time. As a marker of these cells we have used gene Shh. We have found out that these cells persist prenataly and also postnatally. Further we have isolated this cell area and we have tested it using a variety of methods. We have shown that in the cells of postnatal individual are expressed markers of stem cells (Sox2, Bmi1, Gli1) and also genes for major enamel matrix structural proteins: ameloblastin and amelogenin. KEEPING DISTANCE AND COMPLETE FUSION INTO A CHARACTER

This thesis examines my method of portraying theatrical characters throught the main ideas and dramatic theories of Denis Diderot, Konstantin Sergeyevich Stanislavsky and Bertolt Brecht. I perceive the themes of blending with a character and keeping a distance from a character as a question of sense and sensibility in theater, i.e. a question of conscious acting as opposed to acting which stems from the subconscious. While constantly rehearsing new roles and searching for my theatrical self, I came across three mutually contradictory dramatic theories and began to feel a need to confront them. In this thesis, I compare Diderot's Paradox of Acting, Stanislavsky's Method and Brecht's approach to one another while commenting on them from the perspective of my own acting experience. I further elaborate on my acting experience in the chapter about Letovisko, my second graduation play at the Disk Theatre. It is in this chapter where I answer most of my questions related to my acting. Another acting experience described in this thesis is my work on two productions of Bertold Brecht's Drums in the Night. I compare a version me and my classmates rehearsed in our second semester and a different, altered version we created four years later
