Synthesis of 5'-UTR of the PSM-E variant of GCPII gene as a standard for RT-PCR

PCR method has recently become a key element for the syntesis of genes. In this work two-step DNA synthesis based on PCR was used in order to obtain 5'-UTR of the PSM-E splicing variant of glutamate carboxypeptidase II (GCPII) which should serve as a standard for qPCR in real time. Another aim of this work was to clone this sequence into the plasmid PCRII-TOPO PSM-E for the purpose of protein expression in insect cells in the baculovirus expression system. In the first PCR the middle section of the 5'-UTR sequence was synthesized. This product was subsequently amplified in the second PCR using the first and last oligonucleotide. The product of the second PCR was ligated into the plasmid pUC19 and E. coli was transformed by tis construct. Sequence correctness was confirmed by sequencing analysis. Subcloning of the insert from pUC19 to pCRII-TOPO PSM-E was carried out and the sequence was checked by sequencing analysis again. The obtained construct was used as a standard in qPCR in real time in order to determine the concentration of the samples of mRNA from the prostate tissues of oncologic patients
Coupling Of Laser Beam Into Optical Fibers

Our project deals with examination of coupling of laser beam into optical fibers. In the first part of our project theoretical knowledge essential to the practical part was gathered. Subsequently the effectiveness of 4 different manual coupling methods was measured. In addition, measurements to the numerical aperture in both single- and multi-mode fibers were completed. Finally, the obtained data were made into comparison of the efficiency and suitability of individual fibers and methods for manual coupling.
The UN on the Threshold of the 21st Century – The Convention for Protection of the Mediterranean Sea against Pollution (a case study)

The bachelor thesis deals with the Convention for Protection of the Mediterranean Sea against Pollution. The thesis analyses the influence of Barcelona Convention on the quality of the Mediterranean environment and defines the barriers to a more effective functioning of the Mediterranean Action Plan. It proves that the United Nations plays an important role in the field of international relations, which was also confirmed by the creation of the United Nations Environmental Programme. The bachelor thesis describes how the approach to the issue of environmental pollution at the international level has evolved since the 70s of the twentieth century. The last part compares the condition of the Mediterranean Sea before the signing of the Barcelona Convention with the current condition.
Cultural barriers of Sub-Saharan Africa in the period of globalization. Case study: The implementation of Millennium Development Goals

The aim of this theses is to identify cultural determinants which have influence on African lifestyle and to find out which political, economic and social consequences they could create. The output of research is based on the confirmation our hypothesis that the cultural gap, or rather the clash of traditional and modern Africa, together with discovered determinants influences some barriers which thwart the successful implementation of development aid.
Structural study of the DCL-1 receptor using mass spectrometry

DCL-1 (CD302) is a type I transmembrane C-type lectin receptor, which is expressed on monocytes, macrophages, granulocytes and dendritic cells. However, its extracellular domain lacks the amino acids motives essential for carbohydrate binding in the presence of calcium ions, suggesting that it does not have the classic binding capacity found in other C-type lectin receptors such as the mannose receptor. No exogenous or endogenous ligands have been identified yet, though. Due to internal colocalization with F-actin we can assume, that this unconventional lectin receptor plays a role not only in endocytosis and phagocytosis but also in the cell adhesion and migration. The receptor DCL-1 was first identified as a genetic fusion partner of human DEC-205 multilectin receptor in Hodgkin's lymphoma cell lines. The experimental part of this thesis deals with the characterization of disulfide bonds and data acquisition for validation of DCL-1 crystal structure. First the production and refolding conditions were optimized to obtain the highest amount of DCL-1 protein, precisely its extracellular domain. These optimal conditions were used to prepare the protein for in-gel digestion using specific endopeptidases in the presence of cystamine followed by LC-MS analysis. The tools of effective internal communicaton in times of social networks.

Nowadays, the term of communication includes many types of information sharing and the efficiency of this process is the art of understanding the individual components of the communication process and the proper use of communication tools. It is necessary to be aware of the change of the current people thinking which differs considerably from the previous generations and it relates to the change of communication channels and means of communication. Former thinking of people was based on a set order, while the world today is disorganized and complex. Formerly, there was a finite number of elements that interacted with each other, but there is an unlimited number of elements in a large interconnected style nowadays. There were predictable conditions in the past but there is a high degree of unpredictability today. It is more than obvious that mankind made a big step since the end of World War II and multinational corporations, organizations and associations have a greater influence than the government itself. It is just a matter of discussion to which extent the world will be interconnected in future and how people thinking and communication shift in the coming decades. The subject matter of this thesis is the way of internal communication and used internal tools, especially the analysis of the channels of communication across generations, the effectiveness of communication processes and ways of using nowadays. In the theoretical part, I would like to investigate a change in human thinking (according to research by the Canadian professor Don Tapscott), the differences between the baby boomers, X, Y and new generation Z. My used sources are the websites on social marketing and also the research of magazine Forbes about the youngest generation Z. Besides the change in thinking and different effects on different generational groups I examine specific communication tools of these groups and progress in this area. I draw current and also new challenges arising from the global communication based on resources from United Nations´ official publications and the World Internet Statistics. Furthermore, I am focusing on the internal communication tools and social media. Internal communications is still relatively marginalized in the Czech Republic. Few people realize how great power is hidden within communication and how it can affect not only the atmosphere and relationships in the workplace but also the actual results of the organization. For this reason I chose internal communication as a subject of research in the practical part. The empirical part of this thesis focuses on internal communication among students of the Faculty of international relations of the University of Economics in Prague and the leadership, which share information to the students. I will focus on the online student portal ISIS, intranet, personal approach of teachers and on social media of this Faculty. I evaluate the use of the channels and subsequent effectiveness of communication channels. Based on the results of the survey I will evaluate the hypothesis which presuppose different use and functionality of communication channels. My goal is to assess the selected communication channels based on the results of this research, compare them, evaluate the efficiency and in case of weaknesses suggest possible solutions to improve the situation. Hypothesis I: Tools of Internal communication such as ISIS, website and internet are used effectively by students and students receive through them properly and in a time all relevant information, news and documents to study. Hypothesis II. Personal approach of teachers and officials helps in solving students matters. Hypothesis III. The most helpful tools in sharing information and materials is now moreless social medium facebook than the official networks of the University.
The manager’s communication in a company

This bachelor thesis deals with the importance of managerial communication in the company. There are mainly stressed different forms of communication in the business environment, effective strategies of successful communication in the organization, crisis communication tactics, the most common barriers, conflicts and their overcoming. This work further focuses on the challenges and changes associated with the current trend of globalization, such as the differences in communication in multinational companies or the influence of technology. The way of communication in practice is demonstrated in the analysis of a questionnaire survey held among managers of Lafarge Cement, a.s. Primary data were obtained from the answered email queries. Five managers of the company participated in the survey. According to the evaluation of these expert answers it can be said that the theoretical knowledge of the forms of communication and effective strategic methods of communication often corres pond with practice. Obviously, the communication is not to be underestimated. In practice, proper communication is one of the most important factors for a successful company. Aforementioned managers also describe emergency communication, where they follow procedures and recommendations written in the literature dealing with this subject. The final analysis of managerial responses clearly shows that, although in practice the correct communication is under a great emphasis (training, motivation, evaluation), it is almost impossible to achieve perfect communication throughout the whole organization and managers in the company must constantly face barriers in this area and need to be further educated.
The Financial Literacy for Secondary Education

The thesis deals with financial literacy in secondary education and its possible improvement. Author describes the administrative way from planning to commissioning of requirements for students to practice. It is also mentioned the importance of financial education for society and individuals. The thesis mentions the basic documents on the issue and the role of relevant ministries and the banking sector. There is also solved support for teachers who teach financial literacy in secondary schools. The practical part of the thesis observed research of financial literacy at the secondary school, its organization and results. The findings are compared with the findings of quantitative research conducted for the Ministry of Finance in 2007. The aim is to determine whether students who learnt basic elements of financial literacy, have higher skills and are able to focus better on financial market than respondents in 2007. The conclusion of the thesis contains instructions and advices to do the learning process more effective.
