The Proposal for Development of Family Wine Shop in Prostějov

The diploma thesis deals with evaluation of the current state of the Vinotéka Rek family business and results in a strategy of its further development. The first theoretical part is based on the theoretical backgrounds focusing on the development and growth of entrepreneurship in particular small and family businesses, the context of strategic management within the development of companies and also on selected analytical and research methods that examine the internal, external and market environment of the company. The second, analytical part of the thesis contains the practical application of the methods to the selected company, the identification of key factors and impacts on the company and the evaluation of the overall results for the subsequent selection of the appropriate development variant. Third, the part of proposals builds on the results of the analyzes and processes the selected variant of the development strategy, including the implementation steps and the benefits of the proposals.
Design, Creation and Implementation of Software Applications in the Corporate Environment

True random number generator

The goal of this bachelor thesis is to apprise with true random number generators and their realisation. At first, random numbers and true random number generators are described and principles, details and properties are explained. The thesis also describes the use of generators in computers and in IoT devices and random sequences testing. The part of this thesis is description and realisation of computer generator and description of smartphone generator. The source of entropy used in this work is cursor move. Generated sequences were tested by the NIST STS and compared with sequences from other generators.

This Bachelor Thesis deals with elaboration of requirements on metrological system in the production process of vibro-pressed products for pavement coverings and masonry. The thesis represents a research of legal regulations and norms. It identifies researched metrological quantities in the production process and control activity. The goal is to make an overview of measuring devices and proposal of their justification periodicity.
Extremophilic microorganisms

This bachelor’s thesis deals with the extremophilic microorganisms. There is described extreme environment in which these microorganisms occur, as well as several structural and chemical adaptations to the environment. In most cases, adaptation to extreme environment requires changes in the composition of membrane lipids, nucleic acids and proteins. Enzymes with stability in extreme environment are used in chemical, food, pharmaceutical and textile industry. There is also mentioned the application of extremophiles in the biodegradation of organic materials. This bachelor’s thesis is mainly focused on hyperthermophilic microorganisms, which have been most researched.
Verification of fermentative process of grape juice

This thesis deals with identification of yeasts isolated during spontaneous fermentation of grape juice. For analysis the Pinot Noir grape variety grown in the integrated vineyard was chosen. In the theoretical part of this thesis basic information about yeasts are described. Genera of yeasts that occurs during fermentation process and methods based on PCR are also described. In this thesis PCR-RFLP method was used for identification of yeasts. The amplification of the 5,8S-ITS rDNA sequence was performed by the polymerase chain reaction with use of the primers ITS1 and ITS4. The restriction analysis was performed by applying five restriction endonucleases: HaeIII, HinfI, Taq?, AluI, MseI. The amplicons were split into fragments which length and number are typical for the particular species. These fragments were identified by agarose gel electrophoresis and electrophoreograms were evaluated by BioNumerics software. Dendrograms representing genetic similarity of isolated wine yeasts were created by using UPGMA cluster analysis.
Encapsulation of active substances into nanofibers and possibilities of their application

The master thesis was based on the optimization of the production of nanofiber covers and to gaine the product for subsequent functional use. The production of nanofiber covers was made by electrospinning and forcespinning from selected materials. Polyhydroxybutyrate, gelatin, chitosan and alginate were used as starting materials. After successful optimization, these materials were enriched with active ingredients ampicillin and ibuprofen for the functionalized use of covers for more effective wound healing. The theoretical part was focused on the issue of skin, healing processes, types of wounds and nanofibers, the characterization of selected starting materials for the formation of nanofibers was also mentioned. The practical part was based on the lengthy optimization of the preparation of fiber covers and later enriched with active ingredients. Furthermore, combined covers made of different materials with contents of both active ingredients were designed. This was followed by the characterization of all prepared covers from the point of view of stability in the short and long term. The gradual release of active ingredients was determined spectrophotometrically and by hifh performance liquid chromatography. It was also important to determine the antimicrobial activity of selected active substances. At the end of all testing, combined coatings containing both active ingredients were used for safety testing with human keratinocyte cells (HaCaT). Safety testing was based on determining the viability of human cells using the MTT test, to verify the LDH test. A scratch test was also performed, a wound healing test after the application of devised combined covers.
The use of dynamic non-destructive methods for determining the strength of cement from cement plant Hranice

Nondestructive testing methods are employed as a tool for evaluation of parameters of building materials, especially to ascertain strength properties and to determine alteration occured during durability tests. An ultrasonic pulse velocity method is one of the most used nondestructive methods. A resonance method is employed among the nondestructive methods less. The aim of this work is to establish the calibration relationships between parameters of nondestructive testing and compressive strength value and flexural tensile strength value, respectively. Moreover, applicability of the calibration relationships is also discussed.
Determination of antimicrobial activity of berry fruit extracts

The goal of this bachelor´s thesis was the determination of effect of model sample of chokeberry extract (Aronia melanocarpa) on selected types of microorganisms (bacteria Bacillus cereus and Escherichia coli, yeast Candida glabrata). Two commercially available samples of natural extracts from raspberry and cocoa were selected and analysed for comparison. Theoretical part is focused on characteristic, composition and properties of mentioned plant materials, there are also listed their uses in pharmacy, medicine and cosmetics and possible methods of determination of antimicrobiological activity. Analysed commercially available samples of raspberries and cocoa don´t show any antimicrobial activity to selected microorganisms with the exception of 100% raspberry extract on Candida glabrata. There was growth promotion of colonies in the surroundings of the extract in other microorganisms. Ethanol extract of chokeberry fruits showed growing antimicrobial activity with increasing concentration of the extract. The strongest potential was observed in yeast Candida Glabrata and in bacteria Bacillus cereus. The bacteria Escherichia coli was completely resistant.
