In the first part of this thesis, there is a short research dealing with modernization of old-type machines. There is also given a description of main types of milling machines. The next part includes characterization and parameters of the milling machine, that is the subject to the thesis. There are to be found designes and calculations of selected units in the following part. In consideration of the character of the thesis there are not given calculations of all units. The last part deals with economical evaluation.
Microstrip Directional and Hybrid Couplers - a laboratory experiment

The diploma thesis deals with the elaboration of a laboratory experiment with basic types of microstrip directional and hybrid couplers in the frequency range up to tens GHz. The thesis is concerned on branch line directional coupler, rat-race directional coupler, De Rondeho coupler, coupled-line coupler, and Lange directional coupler. Simulated results are confronted with measured results obtained by a skalar network analyzer. From overall findings the relevant laboratory experiment was configured and designed.
Utilization of Catalytic Hydrogen Evolution in Electrochemical Analysis of Nucleic Acids

Catalysis of hydrogen evolution (CHE) at mercury in the presence of proteins was discoverd shortly after the introduction of polarography. In contrast, unmodified nucleic acids have not been reported to produce distinct signals due to the CHE to date. Chemically modified nucleic acids bearing certain extrinsic groups produce analytically useful signals due to hydrogen evloution catalyzed by the respective modifications. These species include (a) transition metal complexes, and (b) non-metal catalytically active organic moieties. In addition, the CHE has been reported to be invoved in guanine reduction process at the mercury-based electrodes.
Electrochemical Detection of p53 Protein Interactions with Plasmid DNAs Modified with Cisplatin Using Immunoprecipitation at Magnetic Microbeads

Antineoplastic drug [cis-diamminedichloroplatinum(II)] (cisplatin) forms covalent adducts with DNA. Cisplatin-modified DNA can be determined sensitively using square-wave voltammetry at mercury electrodes. Tumor suppressor protein p53 binds to DNA in different modes, including sequence-and structure-specific ones and these interactions are influenced by modification of the DNA with cisplatin. In this contribution we present a simple immunoprecipitation technique with magnetic beads, followed by voltammetric determination of recovered cisplatinated DNA, for the evaluation of p53 protein binding to DNAs containing various target sites differing in their proneness to being internally modified with the platinum complex.
TAIL-LABELED OLIGONUCLEOTIDE PROBES FOR A DUAL ELECTROCHEMICAL MAGNETIC IMMUNOPRECIPITATION ASSAY OF DNA-PROTEIN BINDING

A novel assay for detection of DNA-protein binding has been developed. Oligonucleotides bearing or lacking specific binding site of the p53 protein were tail-labeled by two different modified deoxynucleotide triphosphates using terminal deoxynucleotidyl transferase. Electrochemical detection enabled to discriminate between sequence-specific and non-specific p53-DNA binding in a competition assay.
Directing and control of regional media in Czechoslovakia in the 70s and 80s of the 20th century based on the example of media reflection of culture events in Pardubice district

This dissertation thesis addresses the history of public relations of the Communist Party in Czechoslovakia in Pardubice district. It is focussed at the period of so-called normalization. Firstly, it discusses the historical context of the time for understanding the crisis of the system in 70s and 80s. The first chapters describe so-called Prague spring and military invasion in August 1968. Next chapters stress changes in politics and media after the election of Gustáv Husák to become First Secretary of the Communist Party in April, 1969. The second part of text is case study about Pardubice district. The main topic of dissertation thesis are newspaper of Pardubice district - Zář. But there are also chapters about radio, factory newspaper, regional Union of Czech Journalists or College in Pardubice and editors of student magazine. In the second part of case study attention is focussed on cultural section of newspaper Zář and its main topics - for example political anniversaries and the role of culture. In cultural section I analyze which information and how they were, or were not presented to public. Supervision and control over the media content was one of the main regime's tools to take over the culture area and its presentation.

The ČTK Law: The Halfway Transformation. Evolution of relationships between political and media sphere in the Czech Republic in the '90s of the 20th century focusing on Czech News Agency Dissertation describes evolvement of relations between Czech political and media sphere in the 90ties of 20th century, focusing on the ČTK transformation, in which media sphere was influenced by politics. The first three chapters describe the legislative process of drafting law for the ČTK (The Acts No. 123/1965 Coll., 310/1991 Coll., 517/1992 Coll.). The next two chapters deals with application of those legal norms and putting them into practice: illustrated by election procedure of the very first ČTK Council (1993), and by Czech government's attempt to reinstate the obligation to publish full-text of its statements, and to redefine the mission of the agency (1995). The last chapter summarize attitudes concerning change of the ČTK Statute and the possible privatization of the agency. The chapter relates to the term of the second ČTK council (1998), however above mentioned issues were discussed from the beginning of the 90ties already, because the lawmakers in the 1992 presupposed that the public service media statute of the ČTK, unlike the Czech Television and Czech Radio, should be the interim one. Measurement of general characteristics of microphones
