University of Agriculture in Prague. Architecture of University Buildings in 1960s
Kopecká, Kristýna ; Novotná, Eva (advisor) ; Schmelzová, Radoslava (referee)
The architecture of university buildings in the '60s of the 20th century The agricultural university in Prague The bachelor thesis is aimed at premises of the Agricultural University in Prague-Suchdol. It thoroughly deals with a history of education in the agricultural branch and its anchoring to a university level, places where it was taught in the past and most significant personalities connected to the field. The following chapters are devoted to the contest for building up these extensive premises, which were the largest planned realization of the capital city in those times. The jurors rated overall 24 projects, that were given over and are introduced. These ratings are then presented together with ratings of the contest in a contemporary press and introduction of all the projects. Location of the premises and the victorious project by Jan Čejka are presented subsequently. I dealt there with the architecture, the technical description of the particular buildings and urbanistic solutions of the premises, also the newer realizations are presented. In the conclusion I attend to the original works of art that were preserved in the premises up to this day. Architekcture ČSR '60s agricultural university Jan Čejka
ARTĚL - The Idea, Activities and Noteworthy Figures in the Association
Skopalová, Markéta ; Rakušanová, Marie (advisor) ; Novotná, Eva (referee)
Synopsis The thesis opens up by reviewing the situation in arts and crafts in the wake of the 20th century. Imperfectly managed transition to mass production caused lower quality. As a result, movements for raising quality arose in Europe. These arts and crafts societies are introduced. The British Arts and Crafts Movement, the Werkstätte in Dresden, the German Werkbund, and the Wiener Werkstätte influenced the Czech scene the most. They became models for the first Czech society of this kind, the Prague Artěl. The history of Artěl is discussed, in particular its creation, development, participation in exhibitions, and links to other organisations. In its final part, the thesis introduces Artěl's most important members and their works. Artel, Prague art group, Arts and crafts, Applied art, 1st half of 20th century
Hrádek u Nechanic - Local Architecture and Its Source of Inspiration
Sieglová, Kateřina ; Kuthan, Jiří (advisor) ; Novotná, Eva (referee)
HRÁDEK U NECHANIC - ARCHITECTURE AND ITS INSPIRATORY SOURCES Chateau Hrádek u Nechanic was built by František Arnošt, the count of Harrach (1799-1884) as a new representative residence of the county family and also as a Harrach's earldom administrative centre. The foundation-stone of the carcass was layed in the year 1839 and the ceremonial laying foundation of the chapel took place two years later. The construction was officially completed in the year 1857 but works in interiors were practically proceeding during the whole second half of the 19th century. Subject of this work is architecture of the chateau Hrádek u Nechanic and its inspiratory sources. We talk about first large aristocratic building in the whole Habsburg Imperium that was in such an integral resemblance influenced by the english Gothic Revival phenomenon - an architecture, an interiors culture and a lifestyle. As an author of building plans is despite many obscurities considered Edward Buckton Lamb (1806-1869). It makes from count František Arnošt of Harrach propably the only one developer in the Habsburg Imperium who gave an order of a project for his chateau directly to an english architect. During the construction were made changes and adjustments by the vienesse architect Karl Fischer who took also a control of bulding. At the...
The detection of protein covalent complexes with DNA using fluorescent microscopy
Melicharová, Růžena ; Jirkovská, Anna (advisor) ; Novotná, Eva (referee)
Charles University Faculty of Pharmacy in Hradec Králové Department od Biochemical Sciences Candidate: Růžena Melicharová Supervisor: PharmDr. Anna Jirkovská, Ph.D. Title of thesis: The detection of protein covalent complexes with DNA using fluorescent microscopy Anthracycline antibiotics are present one of the most potent antineoplastic drugs. The mechanism of their action is complex. They are reported to intercalate to DNA, form DNA adducts and interact with topoisomerase II (TopII) as its poisons. Catalytic cycle of TopII is interrupted when anthracyclines stabilize the covalent complex of DNA and TopII and that causes cell damage. However, using of anthracyclines is limited by several adverse effects e. g. myelotoxicity and cardiotoxicity. The mechanism of cardiotoxicity is still unclear but may be associated with poisoning of the TopIIβ isoform. Unlike the TopIIα, TopIIβ is present mostly in quiescent cells as cardiomyocytes. Furthermore, the only clinically approved cardioprotective drug dexrazoxane belongs to TopII catalytic inhibitors. Nevertheless, the details of the dexrazoxane-afforded protection are unclear. This thesis was aimed to optimize the TARDIS (trapped in agarose DNA immunostaining) assay to detect and quantify covalent cleavage complexes, compare different ways for analysis of...
Influence of benzoxazinedione derivatives on selected mycobacterial enzymes
Švidrnochová, Michaela ; Novotná, Eva (advisor) ; Krátký, Martin (referee)
Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Mgr. Michaela Švidrnochová Supervisor: RNDr. Eva Novotná, Ph.D. Title of Thesis: Influence of benzoxazinedione derivatives on selected mycobacterial enzymes Tuberculosis is an insidious infectious disease with more and more increasing incidence in certain regions. It is difficult to treat by the available means because of increasing cases of multidrug resistance, which is defined as a resistance of M. tuberculosis at least to two primary antituberculotic drugs - rifampicin and isoniazid. That's why is necessary to search for new substances which interfere with the metabolic pathways of Mycobakterium tuberculosis. Enzyme inhibitions in different levels of these pathways are considered to be a target of the new potencial antituberculotic drugs. In this work, influence of monosulphur and disulphur derivatives of 3-(4-alkylphenyl)-2H- 1,3-benzoxazine-2,4(3H)-dione on enoyl-ACP-reductase, dihydrofolate reductase, isocitrate lyase and FtsZ was tested by thermal shift assay, spectrofotometric method and light scattering. The results showed that the dihydrofolate reductase is not a drug target of selected inhibitors. Isocitrate lyase was the most inhibited by sulphur derivative 3-(4-terc-...
Detection of His-tag and FLAG-tag recombinant proteins
Javorská, Lenka ; Zemanová, Lucie (advisor) ; Novotná, Eva (referee)
Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Bc. Lenka Javorská Supervisor: RNDr. Lucie Škarydová, Ph.D Title of diploma thesis: Detection of His-tag and FLAG-tag recombinant proteins Recombinant proteins are proteins produced in genetically modified organism by recombinant DNA technology that enable insert DNA sequence of target protein in expression cell. This technology allows preparation of actually any protein in any quantity without difficulties associated with demanding purification of native protein from tissues. Recombinant proteins are produced often in form of fusion proteins with suitable peptide tag (e.g. His-tag, FLAG- tag) that can help with their detection and enable simple purification. The aim of this diploma thesis was to introduce and optimize universal method for the detection of recombinant proteins labelled with His-tag (6x histidine) and FLAG-tag. The model recombinant protein with His-tag was enzyme carbonylreductase 1 (CBR1) produced by expression system Escherichia coli. For FLAG-tag labelled protein were chosen enzymes DHRS12, HSD11β1, RODH4, DHRS3, DHRS7, DHRS8 produced by insect cells (strain Sf9). SDS polyacrylamide electrophoresis for protein separation and immunochemical detection after Western...
Olga Karlíková and Her Artwork in State Institutions
Váňová, Martina ; Rakušanová, Marie (advisor) ; Novotná, Eva (referee)
ANGLICKÁ ANOTACE A theme of the bachelor thesis is a live and work of art a Czech artist Olga Karlíková (1923-2004). Her work have not been lined up theoretically, and treating of her comprehensive monograph and listing her art-works in municipal and private hands is an incentive issue and deal for a renowned historian of art. The aim of my work is to partially contribute to exposure her work. A list of her works in municipal institutions hands is a part of the thesis. Olga Karlíková born in 1923 in Prague and there she live through her childhood. In 1948 she graduated at Academy of Arts, Architecture and Design in Prague, where she studied at Prof. Antonín Kybal (1901-1971), and she gained a stance on tapestry because of him. She worked in Ústav bytové a oděvní kultury - ÚBOK (Institute of Quarter and Dress Culture), where she used to design a quarter textile. She dedicated herself to tapestry the most. There used to be an open and creative atmosphere, and Jinrřich Chalupecký spent there a short episode, for example. Jiří and Daisy Mrázkovy were her long-time and close co-workers. Karlíková was a solitaire artist above all. She created without a support of collective programmes. She acted in a group Bilance for a short time, in totalitarian Czechoslovakia was a member of a Svaz Československých výtvarných...
