Petra Matoušková, Ph.D. Title of diploma thesis: Construction of plasmid and probes for detection of P-glycoprotein in Haemonchus contortus Anthelmintic resistence in parasitic nematodes has become an alarming issue worlwide. Haemonchus contortus, a pathogenic nematode of small ruminants, has become resistant to all types of anthelmintic drugs. Important players of the anthelmintic resistence are ABC transporters, most notably P-glycoproteins. In Haemonchus contortus, 10 homologues of Pgp have been identified. Research of individual P-glycoproteins could reveal particular transporters involved in anthelmintic resistence mechanisms. The diploma thesis focuses on P-glycoprotein-9.2, whose constitutive expression is higher in resistant strains of H. contortus than in susceptible strain. The opening chapters of the diploma thesis deals with the topic of parasitic nematode H. contortus, anthelmintic resistence and description of ABC transporters. Nikola Rychlá Title of Thesis: Characterization of carbonyl-reducing enzymes in barber's pole worm Barber's pole worm (Haemonchus contortus), one of the most pathogenic parasites of small ruminants as sheep and goats, has developed resistance against all used anthelmintic drugs. Detoxifying enzymes, which represent the main defense system against harmful xenobiotics, can also contribute to the development of drug resistance, and they are not fully explored in this organism. This research is focused on the study of carbonyl-reducing enzymes in barber's pole worm using specific substrates and inhibitors. Subcellular fractions (cytosol and microsomes) were prepared from adult worms for following in vitro experiments. Protein concentration and reductase activity towards different substrates were determined in each fraction. Substrates were chosen based on knowledge of human biotransformation enzymes. Multidrug resistance (MDR) is one of the major problems associated with cancer treatment. A key determinant of MDR is increased efflux of drugs through membrane ATP- binding cassette (ABC) transporters, which are a family of transporter proteins. Their overexpression contributes to MDR by decreasing intracellular drug concentration due to the efflux of various drugs from cells. Targeting ABC transporters appears to be a promising approach to suppress drug resistance. Therefore, one strategy to reverse the resistance of ABC transporter-expressing tumor cells is the combined use of chemotherapeutic drugs with ABC transporter modulators to enhance therapeutic efficacy. Petra Matoušková, Ph.D. Title of diploma thesis: Plasmid construction for expression of UDP-glukosyltransferase from Haemonchus contortus The barber's pole worm (Haemonchus contortus), like other organisms, uses biotransformation enzymes to metabolize exogenous and endogenous substances. Important representatives of these enzymes are also UDP-glycosyltransferases, which catalyze the formation of a covalent bond between hexose activated by uridine diphosphate and the metabolized compound, thus helping to eliminate undesirable substances. Increased attention to these enzymes is due to the involvement of UDP-glycosyltransferases in the development of resistance to anthelmintics, which are used to treat hemonchosis. The study of their properties and functions can significantly help in solving resistance issues. UGTs are divided into several families and subfamilies depending on sequence similarities. The Church of St. John of Nepomuk in Štěchovice There are not many Art Nouveau constructions in Bohemia. One of them is the Church of St. John of Nepomuk in Štěchovice designed by the architect Kamil Hilbert. Although the construction is not built in the pure Art Nouveau style, its individual parts and elements are taken over for example from the Roman or Renaissance style, it remains probably the most popular architectural example of this style. In view of this fact it is interesting that neither the church nor the builder were devoted sufficient attention in the past. There are not many sources and literature available. Experts devoted their attention to Hilbert only in connection with the preservation of monuments the modernisation of which was his merit at the beginning. Hilbert's own creation remained on the edge of the interest, be it creation dating back to the end of the 19th century or the first decades of the 20th century. The aim of my work is to inform the readers about the personality of the architect Kamil Hilbert and to depict his most important construction, the Church of St. John of Nepomuk in Štěchovice.
