Evaluation of the properties of new types of zwitterionic stationary phases in HILIC

PharmDr. Lucie Nováková, Ph.D. Title of the diploma thesis: Evaluation of properties of new types of zwitterionic stationary phases in HILIC The aim of this thesis was to compare six new zwitterionic Astra ZWIX columns (3 μm, 2,1 x 100 mm) from Chromservis s.r.o. Five groups of compounds were used for the analysis, namely nucleic bases and nucleosides, nicotinic acid derivates, hydrophilic vitamins, beta- blockers, and phenolic acids. These groups of compounds were also analysed on commercially available columns - BEH Z HILIC, SYNCRONIS, OMEGA SUGAR, BEH HILIC and BEH AMIDE (OMEGA SUGAR 3 μm, 2,1 x 100 mm; other columns 1,7 μm, 2,1 x 100 mm). The investigated parameters were retention time, peak resolution, peak symmetry, and peak width at half height. Optimisation to improve peak separation of nucleic bases and nucleosides, hydrophilic vitamins, and beta-blockers was also carried out. The mobile phase gradient was optimized, as well as the column temperature, and mobile phase flow rate. The selectivity of the columns was found not to differ within the batch. A quadrupole time-of-flight mass analyzer SYNAPT G2-Si was used for mass spectrometric detection. Continuous data were collected. PharmDr. Lucie Nováková, Ph.D. Title of the diploma thesis: Development of a sample preparation method for the determination of selected ochratoxins in urine by UHPLC-MS/MS The aim of the work was to develop a method for the determination of ochratoxins A, B, C, and  in urine with the lowest possible quantification limit, and subsequently to carry out the validation of this method, which would be applicable in clinical practice. The method of ultra- high-performance liquid chromatography in a reversed-phase mode in coupled with tandem mass spectrometry was selected for the analyse. Optimalization of sample preparation involved: micro-solid phase extraction in pipette tips (-SPE-PT), dilute and shoot (DAS), supported liquid extraction (SLE) and finally salting-out assisted liquid-liquid extraction (SALLE). The method recovery, the interference of the matrix and the limit of quantification of the given method were evaluated. Corona crisis through the eyes of high school and vocational school students

The bachelor thesis deals with the perception of the coronavirus crisis through the eyes of high school and vocational school students in the Czech Republic. The thesis aims to find out how students experienced and how they dealt with the coronavirus crisis. The qualitative research includes the statements of four respondents which have been interviewed using the method of a semi-structured interview. The data were analysed through qualitative open coding. The bachelor thesis defines key ideas and concepts that are intertwined with the respondents' accounts. Moreover, the thesis also focuses on the mental health and emotions of adolescents during the coronavirus crisis. Furthermore, the thesis describes the influence of the media, students' daily routines and leisure time along with distance learning put into action during the pandemic. Additionally, issues of family and friendship are brought up. The research looks into the lives of four young people during the global pandemic of virus SARS-CoV-2.
Impact of farm size on efficiency economy

The aim of the bachelor thesis is to evaluate the development of agricultural companies divided by size, not only according to the number of employees, but also according to the balance sheet total of the annual balance sheet. Another goal is to evaluate whether the size of the agricultural holding has an impact on the economic result. It is based on data from the Albertina database and monitors the development from 2015 to 2019. The theoretical part explains the specifics of agriculture and deals with the topic of financial analysis. In this work, the data are included only for agricultural holdings classified according to the CZ-NACE classification of activities. The methodology describes the Albertina database used and the methodological procedure explains the selection of financial indicators, the subsequent use of these indicators and their calculation from the average company divided by size in a longer time series. In the end, the comparison of the calculated indicators with other sizes of enterprises is also shown, as well as the development of selected indicators in the graphs.

The thesis deals with various valuation methods. The thesis is divided into theoretical and practical part. The theoretical part lists the functions of greenery in the environment and emphasizes how important the greenery in urban area is. Is created an owerview of terminology in the field of valuation and a description of methodologies suitable for tree appraisal. The practical part deals with the identification of public greenery in the selected area and data od individual tree species processes into tables. The results of individual methods are evaluated in the discussion. The aim od the thesis is to identify the public greenery in the urban area and to evaluate it using two selcted methods and to compare the results. A partial task of the thesis is to quantify the cost of maintenance of greenery in the urban area.
