Němečková, Lenka ; Semecký, Vladimír (advisor) ; Louda, Miroslav (referee)
Increased incidence of benign prostatic hyperplasia (BPH) has been associated with age of men. The quality of lives gets worse in men with BPH. Adenocarcinoma of the prostate (CaP) is the most common malignant disease in aged men, younger men as well. In 2005 the incidence of prostate cancer has exceeded the number of occurrence of lung cancer in the Czech Republic. Prostate specific antigen (PSA) has found widespread usefulness in diagnosis of prostate cancer and BPH, too. PSA is organ-specific marker only, therefore, new options (modify PSA, new markers) have been searched to improve CaP detection. Histological estimation of prostate biopsy specimens validates the diagnosis of CaP. Gleason score (GS) determining histology of biopsy tissue affect treatment strategy. Frequently GS of biopsy can be varying GS of final preparation after radical prostatectomy. Therefore, we analyzed the correlation between the prostate biopsy GS and GS of final preparation in patients undergoing radical prostatectomy. Almost half of cases in this group of 88 patients treated in The Clinic of urology in Faculty Hospital of Hradec Králové was identical to GS of prostatectomy specimens and same part of cases was undergraded. High grades correlated with elevated PSA levels. Amount of patients had the PSA levels over PSA...

The introduction of immunohistochemistry in laboratory practice for prostate disease Němečková Lenka ABSTRACT Increased incidence of benign prostatic hyperplasia (BPH) has been associated with age of men. The quality of lives gets worse in men with BPH. Higher numbers of reported malignant tumours of prostate with age of men, too. Adenocarcinoma of prostate (CaP) was the most frequently diagnosed oncological disease in males after other malignant neoplasms of skin. Physical examination and laboratory methods, especially the measurement of prostatic specific antigen (PSA) in serum, are applied in the diagnosis of CaP. The histological examination of tissue sample obtained by needle biopsy of the prostate confirms the diagnosis of CaP. The immunohistochemical examination of histological sections helps to differentiate malignant and benign prostatic glands in tissue of prostate usually using antibody anti - high molecular weight cytokeratins (HMW CK) highlighting basal cell of benign prostatic glands. Basal cell is missing in malignant glands. Method of immunohistochemistry was introduced and is used in the diagnosis of CaP in the histology laboratory at department of Pathology in Oblastní nemocnice Náchod (ONN).
