Deformation index for tablets of microcrystalline cellulose

Myslivečková Jana - Rigorózní práce - 2006 This work deals with deformation index of tablets of microcrystalline cellulose. Theoretical part describes the theory of tablet formation by means of the record force - displacement. It also characterizes microcrystalline celluloses. The aim of the work was to find out the influence of a filler on the parameter E2 of the compression process and on the parameter DE of the destruction process. Further to asses the influence of the relation between parameters DE, D and E2,DE and effect of the filler type on new parameter DI. Six various fillers were tested. Ceolus KG 802 Avicel PH 102, Avicel PH 200, Pearlitol 100 SD, Tablettose 70, and Emcompress. The results show that polymeric microcrystalline celluloses accumulate less energy at compression than crystalline compounds. However, they have higher value of D. Microcrystalline celluloses have the highest values of the parameter DI. A semilogarithmic dependence was found between the parameters D and DE.
Permeation of nanoparticles across sublingual membrane 4

Student: Lenka Horejšová Title of Thesis: Permeation of Nanoparticles Across Sublingual Membrane 4 The theoretical part describes the anatomy and physiology of oral cavity and sublingual drug administration. It also describes theory of nanotechnology and the characterization of nanoparticles with a focus on nanoparticle drug forms and there are also mentioned methods and mesuring device. . The experimental part describes in vitro permeation experiment of nanoparticles (Nano beads based on PD, Chromeon 470 marked, carboxylated). Permeation of this nanoparticles through porcine sublingual membrane (1 cm2) . Thesis compares nanoparticles transfer of phosphate buffer pH 6.6 and citrate buffer pH 6.6 to the acceptor phase consisting of phosphate buffer pH 7,4. First permeation experiment works with frozen sublingual membrane, a second experiment with the native membrane. The dimension of nanoparticles and aggregates of nanoparticles in dispersions were measured before the start of permeation by the dynamic light scattering. DSC of Lipidic Excipients

Title of Thesis DSC of lipidic excipients In this work was evaluated the interaction of the lipidic components with water by using DSC (differential scanning calorimetry). In the first part of this work, we used samples which contained a phospholipid (DMPE). The samples which we used in the second part of the work, contained cutaneous lipids presented by pseudoceramides (14S24 and H6524) and cholesterol. Our attention was especially payed to the vindication of the existence of different structure organizations of hydrated lipidic components. By the samples which contained the phospholipid DMPE, we registered the most of the phase transitions presented in the literature on our calorimeter by using slightly modificated conditions. In similar conditions of the measurement we registered only the crystallization peak from the different phase transitions by the commercially produced pseudoceramide H6524. By the synthetic pseudoceramide 14S24 by used conditions of the measurement we didn't prove any phase transitions.
Suppositories in current therapy I

Suppositories are a conventional dosage form. Although their research is not being developed much further, there are still some changes to be found. Some new adjuvants have been used, the old active substances have been deleted and replaced by new ones. The last survey of rectal preparations was created in 1998. This paper provides an overview of proprietary rectal and vaginal suppositories, which were available on the market in 2010. There is a list of such products, medicinal substances used and their percentage. There are adjuvants and packaging materials, too. We found values of the melting temperature of those suppositories, which were stored under different conditions. These values were correlated to times of deformation of the preparations. On the basis of the evaluated data we recommend monitoring of changes in the melting temperature and the time of deformation of the products DICLOREUM 100 and KETONAL.

Thermal analysis of interaction of epidermal lipids III Lenka Dvořáková, diploma paper, June 2008 Abstract The skin is the organ protecting the human body from unwanted influences from the environment. Physical chemical properties, structure and ordering of the epidermal lipids, mainly in the stratum corneum, play a key role in the barrier function of the skin. The major lipids are ceramides, cholesterol and free fatty acids in this layer. We are used the samples mimic such the most frequently referred composition of the epidermal lipids, contained pseudo-ceramid, oleic acid, cholesterol and cholesterol sulfate. Because the samples with oleic acids can at the higher temperatures influence the behavior of the particular constituents of the mixture, we are also performed the evaluation of the contained simpler samples which compound cholesterol and pseudo-ceramid. We are tried hydrated the mixtures at in vitro environment by various routes. And we are observed their characteristics and interactions by means of the differential scanning calorimetry. We are also deal with the evaluation of the particular raw materials and the double-barrelled mixtures. We are verified and in detail elaborated the operating sequences which are not completely unambiguously referred in the literature by means of these studies.
The use of DSC for assessment of content uniformity of metronidazol in suppositories

The aim of this work is the evaluation of extemporaneously prepared metronidazole-containing suppositories. Content uniformity, dispersion of the active ingredient, and the content in relation to the declared one was examined. The content of the active ingredient was determined by means of DSC. The preparations complied with the test of content uniformity. The content of the active ingredient was lower that the declared one.
Determination of the plasticity of lactose

Juhaňáková Jana - Rigorózní práce - 2006 This work deals with the determination of tablet plasticity made of Tablettose 70 using stress relaxation method. Theoretical part of the work describes preparation and characterization of the tablets. It deals in detail with the determination of residual plasticity of Tablettose 70, determination of the influence of the dwell time on residual plasticity and on the dependence of residual plasticity on compression force. Plasticity was measured by means of T1 - -FRO 50 device. From the results of the work it follows that the increase in dwell time results in residual plasticity of Tablettose 70. Tablettose 70 has the parameter A 49.679 % and tangent B -13.712% kN-1 .

Title of diploma thesis: Suppositories in current therapy II Suppositories as dosage forms are still in use at present. For certain medicinal substances they are even one of the few possible applications. Therefore it is still necessary to deal with their proper formulation and storage. In this thesis, we followed up the influence of storage temperature of suppository bases on its calorimetric characteristics. As a basic method of thermal analysis we used differential scanning calorimetry. We determined the content of the solid phase in the suppository bases stored at temperatures of 26 řC and 5 řC. We evaluated the changes that occurred within one year after the preparation of suppositories. The obtained data shows increase in share of solid phases during the storage and the major differences are just in storage temperature. Thus we consider appropriate to set maximum temperature limits for storage of suppositories.
Methodical study of surface tension of epidermal lipids

The theoretical part brings a brief overview of selected relevant papers dealing with measurements of advancing and receding contact angles. These studies deal with the methods of determination of free surface energy, determination of contact angles, estimation of free equilibrium surface energy components, changes in the level of components of free surface skin energy following washing, Marangoni effect in water, advancing, receding and equilibrium contact angles, measurement of dynamic adhesive tension and dynamic contact angles. In the experimental part, the Krüss K100 tensiometer equiped with the plate method was used to measure the surface tension and contact angles of distilled water at 37řC on the surfaces impregnated with examined substances in selected proportions and dissolved in volatile solvent. The measuring plate surface was impregnated with prepared samples in advance; the first samples: palmitic acid and oleic acid with cholesterol; the second samples: cholesterol sodium sulphate in mixtures with cholesterol and selected fatty acid, as well as in combination with cholesterol, 14S24 and oleic acid; the third samples: N-acetyl-D-sphingosin, N-hexanoyl-D- sphingosin, 12SG12, 12GV12 in the solution alone, as well as in combinations with cholesterol and in combination with cholesterol...
