Charles University Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Student: Nikola Martínková Supervisor: PharmDr. Anna Jirkovská, Ph.D. Consultant: Mgr. Petr Jošt, Ph.D. Title of diploma thesis: Induction of oxidative stress in skin keratinocyte cells Keratinocytes are part of the epidermis and represent the majority of cells in the upper layer of human skin. When these cells are damaged by mutagenic substances, changes in the composition of the genetic material may occur, and even cell death may occur. Such substances that induce changes in DNA and have mutagenic potential include the chemical warfare agent sulfur mustard. The purpose of this study was to compare the HaCaT keratinocyte cell line with the experimentally derived 3HSM4 cell line in response to hydrogen peroxide-induced oxidative stress. The 3HSM4 cells were prepared by selecting resistant clones of HaCaT cells that were repeatedly exposed to the cytotoxic effects of sulfur mustard. The results of the comparison show that 3HSM4 cells proved to be more resistant to hydrogen peroxide. When glutathione (GSH) synthesis was blocked in both cell types, the cytotoxic effect of hydrogen peroxide was the same. Although 3HSM4 cells were shown to be more resistant in terms of cytotoxicity, they were found to have much...
Limity supervize u pracovníků v sociálních službách

The aim of the diploma thesis was: "To identify what limits perceive supervisors at group supervision meetings of social workers in social services." The main research question was: "What are the limits of supervision of social workers from the perspective of supervisors?" A qualitative research strategy, a method of questioning and a semi-structured interview technique were chosen. To achieve the goal, the main research question was divided into areas: identification (6 partial research questions), formal requirements of supervision (2 partial research questions), supervision (4 partial research questions) and benefits of supervision limits (2 partial research questions). The research group consisted of supervisors associated under the Czech Institute for Supervision (ČIS). Taking into account the set strategy, 6 interviews (snowball method) were conducted, which already showed that the sample had been saturated. In order to be involved, the involved informants were required to provide supervisory services in the field of social work and to have at least one year of experience. The interviews were recorded on a dictaphone, from which a verbatim transcript was made. I subsequently coded the transcripts transcribed in this way (processed by the cluster method). Within the first area, i.e., identification, there were identified these clusters: the humanities direction of education, supervision training, psychotherapeutic training and preparation for limits. Within the second area, i.e., the formal requirements of supervision, the following clusters can be specified: contracting, formal limits, content limits, supervisor under supervision. In the area of supervision, which was identified as the third area, I present the following identified clusters: self-reflection, naming limits and ethical aspects. The last, fourth area, i.e., the benefits of supervision, brought the following clusters: personal development, re-contracting, emotional intelligence. The results of the thesis can be used in the training of supervisors, or for the development of already qualified supervisors in practice.
Advanced age of Roma seniors and its issues
MARTÍNKOVÁ, Nikola
The bachelor thesis focuses on age of gypsy seniors. The main focus of my bachelor thesis is to describe the age of gypsy seniors, their needs and subjective perception of a social status in a family. Also to find out reasons why the gypsy seniors are not being placed in a retirement home and to describe the way how the gypsy people take care of their seniors. The bachelor thesis consists of expressions such as gypsy, age, social status and social work. It also describes the history of gypsies from the beginning of their mentioning until our present. I focus on gypsy family, their tradition and the age of seniors. At the age I focus on the main needs and occurring changes. My research goals was to find out reasons the gypsy seniors are not being placed in the retirement home. I chose three age categories of gypsy seniors for my research. The first category are seniors from 65 to 70 years, the second category are seniors from 71 to 80 years and the third category are seniors from 80 years +. During my research I found out that I am not able to obtain the third category since I had the informants based on the snow ball technique, therefore no one from my informants knew any senior older 80 years. This is the reason why my is my research based on two categories only. Next the research focuses on the second category. This category is about offsprings of the gypsy seniors. Half structured interviews were done with eight gypsy seniors and their eight offsprings. The result of this work was to find out the opinion of the gypsy seniors and their offsprings on the retirement home. Also how the gypsies take care of their seniors and what needs the seniors have. I hope and believe that everyone will benefit from the knowledge of this bachelor thesis.
