Structure and biocatalytic properties of nitrilases from filamentous fungi

Filamentous fungi are alongside bacteria a rich source of nitrilases that can be used for biotechnological purposes. There is a large number of sequences of putative nitrilases in databases, but few of these enzymes have been expressed, purified and characterized. According to the sequences in GenBank database, synthetic genes of putative nitrilases from Aspergillus niger CBS 513.88 and Neurospora crassa OR74A were designed, expressed in Escherichia coli and the nitrilases produced were purified and characterized. The effect of chaperone-assisted nitrilase expression was also examined using a set of commercial chaperone plasmids. The structural properties of nitrilases were examined using various methods, such as homology modelling, sedimentation velocity analysis or electron microscopy. Recombinant nitrilase production was compared to their production by native producers - filamentous fungi. The cultivation of native producers was rather difficult and nitrilase production was very low, therefore, the advantage of using prokaryotic expression system was highlighted. Synthetic nitriles and inorganic cyanides occur as pollutants from human activities (industrial wastes, herbicides, etc.). Both natural and man-made nitriles are poisonous to many organisms, the natural ones serving as a protection against enemies (e.g. herbivores). Their toxic activity is used in some herbicides. Cadastral documentation supersession in cadastral area Blansko

Master´s thesis is about cadastral documentation supersession by revising in part of the cadastral territory Blansko. A cadastral map made by Instruction A is revised to concept of a digital cadastral map. Current status of the data file of geodetic and descriptive information from the cadastre information system and records detailed measurements of changes based in the documentation cadastral department Blansko are used. Planimetric coordinates of cadastral map in vector format were focused in the field for control. Thesis contain a comparison of size of parcels owned paper and the graphic file.
Methodology of determination of free cyanide (CN-) in industrial wastewaters

This work summarizes the current state in the methodology of CN- determination particularly in industrial wastewaters. It also describes in detail the determination of cyanide ions after their enzymatic conversion into formamide, which is determined by HPLC (including the method of preparation of the enzyme).
Seeking Novel Nitrilase Enzyme (Nitrile Aminohydrolase, EC 3.5.5.1)

Our research group has created a library of novel nitrilases. In general, the enzymes are available from heterologous producers in amounts ranging from 10 to 20 mg of protein exhbiting 25-50 % purity. The enzymes differ from each other in substrate specificity, temperature optimum, enantioselectivity (arylacetonitrilases) etc. Preferential substrates are phenylacetonitrile, (R,S)-mandelonitrile, benzonitrile or cyanopyridines (depending on the enzyme).
