Economic cycle and municipal budgets
MACHOVÁ, Lenka
The main aim of this diploma thesis named Economic Cycle and Municipal Budgets is to assess the impact of the economic cycle on the revenue and expenditure part of municipal budgets in the Czech Republic. The practical part is divided into two sections. The first section shows the development of the economic cycle in the Czech Republic and the development of revenues and expenditures of Czech municipalities in the years 2006 to 2019. The second section is an econometric analysis of the behavior of Czech municipal budgets depending on the economic cycle. Data for the second section of the practical part are obtained from publicly available databases such as ARIS, ÚFIS, AMECO database and MONITOR of the Ministry of Finance of the Czech Republic. In total 6 239 municipalities of the Czech Republic were analysed for the reporting period 2010 to 2019. A total of 12 explained variables were determined, namely: tax revenues, personal income tax, corporate income tax, value added tax, real estate tax, non-tax revenues, capital revenues, received transfers, received non-investment transfers, received investment transfers, current expenditures and capital expenditures. The dependence between the explained variable and the explanatory variable is examined using the regression analysis of the panel data. The economic cycle was found to affect all of the selected explanatory variables. The main hypothesis of the diploma thesis is not rejected. The economic cycle affects the revenue and expenditure part of the municipal budgets in the Czech Republic.
Secondary metabolites in fungal pathogenesis
Veselý, Martin ; Čmoková, Adéla (advisor) ; Machová, Lenka (referee)
The ability of fungal pathogens to induce infection and later survive in its host is dependent on virulence factors. Often these factors are based on primary metabolites (hydrophobins, proteases, phospholipases, catalases etc.). Nonetheless many pathogen produced secondary metabolites are also involved in the infection process. Their true role during infection used to be rather undervalued. First part of this bachelor degree thesis aims to describe host-pathogen relation and afterwards introduce reader with some basic, commonly accepted virulence factors of fungi. In the second part of this thesis known roles of secondary metabolites are described in each stages of progressing infection. In the first stage a pathogen needs to create a suitable habitat on host tissues to ensure growth. This is in part done by antimicrobial substances (...). In later stages pathogen prevents phagocytosis of its spores by host (melanins). Successful infection is often accompanied with host tissue damage that is induced by production of cytotoxic substances (xanthomegnin, riboflavin) and immunoregulation of host immune system (gliotoxin, pseurotin). Last but not least there are virulence factors that ensure prolonged survival in host (siderophores). Fungal pathogens of poikiloterm animals are mostly not primary...
Reliability assessment of a hydraulic switchboard production proces
Machová, Lenka ; Maradová, Karla (referee) ; Blecha, Petr (advisor)
The Diploma thesis is focused on possibilities of an assessment of a hydraulic switchboard and its manufacturing process. Furthermore, it analyses methods individually which are suitable for quality assurance and production reliability, which are described in details. The follow-up work focuses more on the Statistical Process Control (SPC) method which is applicable in the early warning system and the Failure mode, effects, and criticality analysis (FMECA) method, applicable in the planning system. These two methods are further applied to the newly introduced process of machining the hydraulic switchboard as an effective tool not only for assessing the reliability of production, but also for its subsequent securing and maintaining in a relevant condition.
Virulence factors of the Trichophyton benhamiae complex
Machová, Lenka ; Čmoková, Adéla (advisor) ; Labuda, Roman (referee)
Dermatophytes are a group of fungi, some of which can cause skin diseases in humans and animals due to their ability to degrade keratinized tissue. Representatives of this group also include strains from the Trichophyton benhamiae complex, known to cause dermatophytosis especially of small rodents and rabbits. In the last decade, one of four populations of this complex has spread epidemically across Europe among guinea pigs and their breeders. To answer the question what stands behind the successful spread of this population, the gene expression and production of volatile organic compounds of epidemic and non-epidemic populations of T. benhamiae was investigated. Gene expression of three strains from each population was studied during growth in liquid medium and on ex vivo mouse skin models prepared according to a newly optimized protocol. RNAseq and RT-qPCR methods were chosen for the gene expression analysis. Based on the literature and the results of RNAseq preliminary analysis, several genes were selected for which specific primers were designed. The spectra of the produced volatile organic compounds of the same strains growing on sheep wool in vials were analyzed by GC-MS. While non-epidemic populations did not differ in gene expression and production of volatile organic compounds, the...
8D report as a tool for improving the quality of production
Machová, Lenka ; Řezníčková, Hana (referee) ; Hammer, Miloš (advisor)
The Bachelor's thesis is focused on the quality as a means of effective production. Furthermore, it demonstrates individual methods, which are relevant in quality improvement . The procedures and a method 8D that were chosen are described in detail. Such as beneficial funds for quality improvement. The method 8D is considered the most effective mode. In order to achieve these objectives is provided minutely delineation of individual steps of the strategy. The main objective of this work is an analysis of a specific example for business practice. Moreover, it elucidates solutions of the 8D complaint report usage.
Virulence factors of dermatophytes
Machová, Lenka ; Čmoková, Adéla (advisor) ; Dobiáš, Radim (referee)
Dermatophytes are a group of phylogenetically related microscopic fungi of the order Onygenales, the Arthrodermateceae family, capable of using keratin as a source of nutrition. Some species are important pathogens of vertebrates including humans. The aim of my bachelor thesis was to summarize the current knowledge about virulence factors of dermatophytes, taking into account their possible role in the host specificity. In the main part, I describe the virulence factors, pathogenesis mediating molecules, in the context of the individual stages of infection, such as adherence, degradation of the keratin, or suppression of the immune response. The study of these factors in general, deserves increased attention because their knowledge can lead to the development of targeted treatment. Some aspects of pathogenesis, e.g. keratolysis and fighting the immune system, are relatively well studied. However, we know almost nothing about the other factors, especially about those mediating host sensing and adherence, but they could provide important information on identifying the host specificity of dermatophytes. The final chapter is dedicated to a discussion about the potential ability of these virulence factors to influence the selection of the host. My thesis brings a comprehensive overview of the...
Factors influencing municipal capital expenditure
MACHOVÁ, Lenka
The main aim of this bachelor thesis (Factors Influencing Municipal Capital Expenditure) is to evaluate the capital expenditures of all municipalities of the Czech Republic in terms of amount, dynamics, structure and financing. The time series from 2006 to 2016 is used for the analysis of all municipalities. The analysis of the capital expenditures of a selected sample of municipalities (Vysočina municipalities) is also carried out, with attention being paid to the search for factors affecting these expenditures. The municipalities of the Vysočina region are analysed for the period from 2010 to 2016. A total of 8 factors were identified, whereas they could influence the capital expenditures of the Vysočina municipalities. The dependence between the explained variable (capital expenditure per capita) and the explanatory variable (individual factors) is examined using regression analysis of panel data.
Security of Debts
Machová, Lenka ; Jelínková, Martina (referee) ; Musilová, Helena (advisor)
The key focus of my thesis is to determinate guidelines for protection against liabilities within a company. The theoretical conclusions are subsequently applied in the practical section using a sample of a specific company. The proposal establishes a direction to take in order to fully safeguard the specific case including guarantee documents.
