Selfconcept of individuals placed in the diagnostic institute

This diploma thesis is focused on individuals placed in a diagnostic institute and their self- concept. The aim was to evaluate and describe their view of themselves, whether they perceive positively or negatively, in which area they evaluate above average and where they move on the edge of below average. We consider the conception of ourselves to be the essence on which it is necessary to work on, build it and try to strengthen it if these individuals are to be corrected and their current behavior changed. The first part of this work clarifies the theoretical concepts related to the issue and the second part includes the research itself to determine the self-concept of individuals placed in a diagnostic institute. The evaluation of self-concept was based on a questionnaire survey (Questionnaire of self- concept of children and adolescents Piers Harris) and analysis of products of mental activity (drawing a tree). The results of the questionnaires show that the respondents from our research sample do not perceive positively, on the contrary, on many scales they show values corresponding to the average, below average to significantly below average. The elements resulting from the analysis of the drawings only highlight and confirm the results of the questionnaires.
Support of social and emotional development of shildren with ADHD in pre-school education

The thesis focuses on children in pre-school educational institutions who are diagnosed with ADHD (Attention Deficit Hyperactivity Disorder), as well as on the methods of supporting their social and emotional development. The thesis consists of two parts, the theoretical and the practical one. The theoretical part deals with the basic information of ADHD, children's development and the methods of working with children suffering from this syndrom. We can find there three main chapters. The first chapter deals with pre-school children's development. The second chapter concentrates on the definition, causes, occurrence, symptoms, diagnosis and treatment of ADHD. Finally, the third chapter focuses on the methods of working with ADHD children, creating a positive environment, integration of these children, general reccomendations for working with ADHD children and on educational methods and techniques of controlling their behaviour. The practical part investigates how these methods and techniques are used by teachers in pre-school educational institutions. The theoretical knowledge gained from literature is completed by practical findings which I have obtained from interwieving five kindergarten teachers. Analysis of amino acids and polyamines by HPLC and CZE

(EN) This work is focused on two specific classes of amines. First group comprises of cyclic polyaminocarboxylates from the DOTA (1,4,7,10-tetraazacyclododecane- 1,4,7,10-tetraacetic acid) family, which are important reaction intermediates in magnetic resonance imaging (MRI) contrast agent synthesis. In the development of new contrast agents, carboxyl groups are very often protected with tert-butyl ester groups. Among these derivatives, tBu3DO3A is of the highest importance. Therefore, reverse-phase high performance liquid chromatographic (RP-HPLC) and non- aqueous capillary zone electrophoretic (CZE) methods were evaluated for qualitative and quantitative analysis of tBu3DO3A (1,4,7,10-tetraazacyclododecan-1,4,7- tris(tert-butylacetate) and two typical reaction by-products, i.e. tBu4DOTA (1,4,7,10- tetraazacyclododecan-1,4,7,10-tetrakis(tert-butylacetate) and tBu2DO2A (1,4,7,10- tetraazacyclododecan-1,4-bis(tert-butylacetate). These optimized methods were successfully applied to monitoring of real reaction mixtures during synthesis of a new MRI contrast agent. No further sample pretreatment was necessary. Electrophoretic determination of rivaroxaban

Capillary electrophoresis is a method used in pharmaceutical analysis because of its low cost, speed and environmental friendliness. This diploma thesis deals with development of electrophoretic method for rivaroxaban determination. After several optimizations, a method suitable for rivaroxaban determination inside its dosage forms was developed. Optimizations involved change of background electrolyte's composition from aqueous solution of low molecular weight organic acids to non-aqueous solution of acetic acid and cetyltrimethylammoniumbromide in acetonitrile in multiple steps. Final conditions of analysis included background electrolyte composed of 1M acetic acid and 40mM cetyltrimethylammoniumbromide in acetonitrile, sample injection carried out hydrodynamically by pressure of 5,0 kPa for period of 3 s and subsequent insertion of separation voltage of 30,0 kV for entire duration of analysis. Capillary content was mobilized by pressure of 0,50 kPa for entire duration of analysis. With aforementioned optimizations a selective method for determination of dosage forms of rivaroxaban was obtained. This method provides limit of detection 0,0056 mg/ml and limit of quantification 0,019 mg/ml and is linear in 0,01 - 0,40 mg/ml range with a recovery of 93,2 %. Keywords capillary electrophoresis,...
Initial conditions of primary schools for implementation of preventive activities of the PBIS model

Diploma thesis deals with the current methods of implementation of primary prevention of primary schools in the Czech Republic. The aim was to determine the degree of preparedness of primary schools for the implementation of a world-wide research-proven program of positive support for students behavior in the school environment (PBIS) in order to prevent risky behavior of students in schools. The first part of the thesis is devoted to theoretical knowledge about students with risky behavior, preventive and interventional procedures of primary prevention applied in schools and a thorough description of the PBIS method. We used a qualitative method of semi-structured interviews for the research survey. The evaluation took place on the basis of interviews with pedagogical staff at four primary schools. The analysis was performed using open coding. Questions were asked about the use of preventive measures by primary schools and the answers provided an insight into which elements used correspond to the PBIS method. The results of the research show similarities in the use of preventive elements in the field of physical environment of the class and in the positive formulation of the rules of conduct. Separation of chiral compounds

This bachelor thesis deals with the chiral separation of newly synthesized orthoconic antiferroelectric liquid crystals using ultra-high performance liquid chromatography. These studied compounds differ in the alkyloxy-spacer length, in the presence and/or position of the fluorine atom on the phenyl ring and in the chiral center. The separation took place in a reverse separation mode using a chiral stationary phase based on derivatized amylose. First, the effect of mobile phase flow rate and column temperature on separation efficiency was exanimated. The effect of the composition of the mobile phase on the enantioseparation were studied at the optimum temperature and flow rate. The mobile phase was based on acetonitril, while deionized water was added gradually using isocratic elution. Furthermore, the effect of chromatographic conditions and the structure of the studied analytes on resolution and enantioselectivity was investigated, concretely the presence and position of fluorine atom on pfenyl ring, the length of alkyloxy-spacer and the effect of divergent chiral center. Under optimized chromatography conditions we succeeded to separate all examined substances on the base line. Analysis of Nicotine or Opium Alkaloids in Historical Pharmaceutical Relics from 18th Century

Belianský M.: Analysis of Nicotine or Opium Alkaloids in Historical Pharmaceutical Relics from 18th Century. Bachelor Thesis. Prague, Charles University, Faculty of Science 2020. Abstract: Two samples of historical relics of pharmaceuticals dated to the 18th century were analyzed; the first sample contained tobacco, the second laudanum, i.e., a preparation containing opium. HPLC with UV or mass detection was used for analysis. Separation of the alkaloids was performed on X Bridge® BEH C18 column using binary gradient elution. By comparison with a modern tobacco sample and on the basis of data from the literature, the presence of nicotine degradation products, especially 6-hydroxy-N-methylmyosmine, was found in the historical sample. Because the nicotine content in the historical sample was very low, it was not possible to quantify it. The analysis of opium analytes in the second historical relics revealed the main opium alkaloids (cotarnine, morphine, meconin, papaverine, and noscapine) as well as their degradation products. Main alkaloids were also quantified. The ratio of noscapine to cotarnine was determined, which according to the literature is an indicator of the age of opium pharmaceuticals.
