The Social Functioning in Retirement Home during the Covid-19 Pandemic.

This bachelor thesis deals with social functioning in the retirement home during the Covid-19 pandemic. Terms age and ageing are described in the theoretical part. The issues of social functioning, Covid-19 disease and restrictions associated with the Covid-19 pandemic are also discussed. The practical part of this bachelor thesis includes processing and assessment of results of survey. The bachelor thesis has aspirations to characterize social functioning of clients of the retirement home during the Covid-19 pandemic. To achieve this goal the strategy of the semi-structured interview was chosen. The main research question is: How the social functioning of clients of the retirement home has been changed due to the Covid-19 pandemic? Interviewees were chosen according to given criteria. The additional interview with the head nurse, who assessed social functioning of clients of the retirement home, is enclosed to give the bachelor thesis higher plausibility. The Open Coding Method was chosen to assess gained data. The research shows that every client of the retirement home was somehow affected by the Covid-19 pandemic. How much social functioning of clients was influenced, depends on their personalities and abilities to cope with difficult life situations. This bachelor thesis can be used as methodical aid for social workers working with the elderly people or as informational material for public people. It also brings feedback for the retirement home, where was the research held.
Integration of Children from Institutional Care to Independent Life

Leaving institutional care is an important milestone for young adults. After leaving the institute, they may find themselves completely on the fringes of society. Therefore, this work deals with the life of young adults after the end of institutional education. It maps their steps after several years of independent living. The work deals with the issue of completion of institutional education and subsequent independent life of young adults. The aim of the bachelor thesis is to find out how young adults are prepared for this transition to independent living. The work also finds out how the integration process is doing and what tools are used to facilitate it. The work is divided into theoretical and practical part. The theoretical part defines the basic concepts. The first concept is a social worker. It is presented here in all the forms that a young adult may encounter when leaving institutional care. Institutional education is defined here. Its individual types and divisions. Furthermore, the work defines the target group of young adults. In a separate chapter, the work deals with the children's home and the phase of leaving the facility with regard to the goal of the work. Some projects aimed at supporting the integration of children from institutional care into independent living are mapped here. Within the research, a qualitative method of secondary analysis of documents of young adults who left institutional care was chosen. This analysis is intended to show the success of young adults in independent living. The work presents the current form of their lives. The research results show involvement in the project. It also includes the evaluation of knowledge after completing a project that is to help with independent living. Research shows that, despite the equal opportunities that the children's home tried to prepare for children during their years in institutional care, everyone took a separate life differently. It is obvious that it depends not only on the possibilities and chances, but also on the personal attitude of each young adult, his habits and the background from which he comes to institutional education. Despite the individual approach and respect for the needs of individual young adults, facilities and special program staff, not every young adult will be able to start living a successful and satisfied life after leaving the children's home.
Social Pedagogue in Centre of Educative Care

Bachelor's thesis deals with the professional application of a social educator in an educational care centre. The aim of the work is to find out how the members of the centre's multidisciplinary team perceive the profession of social educator. Another aim is to find out how the staff of the multidisciplinary team views the possibility of involving a social educator in the centre of educational care. The theoretical section deals with social pedagogy, possibilities of studying this field. It is also focused on a social educator, his competences and possibilities of his application. The next chapter of the theoretical part is devoted to the educational care center itself and its multidisciplinary team. The practical part of the work is then devoted to research investigation. The methodology of the research, the research package and the reason for its selection are described here. The following chapters describe the progress of the research and the results and evaluation of the responses of each communication partner.
Atmospheric characteristics of white dwarfs

We have analyzed the ultraviolet spectra of 40 hydrogen-rich (DA) white dwarfs. These spectra have been obtained with the Far Ultraviolet Spectroscopic Explorer (FUSE) satellite and are publicly available via the MAST FUSE database. We derived the effective temperature and surface gravity by fitting the hydrogen Lyman spectral lines with model spectra. By applying white dwarf evolutionary models, we were able to determine the white dwarf mass, cooling age and absolute magnitude. These then allowed us to determine the distance to each star and its predicted gravitational redshift. We have identified several chemical elements in the white dwarf spectra. We determined the abundances of C, Si, P and S by measuring the equivalent widths of selected lines of these elements. Finally, we studied the spectra of J0623-376 and LM Com in more detail.
Preparation of tumour ligand B7-H6 with coiled-coil tag and verifying of its binding to receptor NKp30

Natural killer cells (NK cells) are part of innate antitumor immunity. These cells have the capacity to prevent viral infection or malignant transformation without prior antigen sensitization. Activation of NK cells consist of different recognition strategies. Mechanism of activation is based on down-regulated expression of MHC gp I molecules on the cell surface. NK cells possess both activation and inhibitory receptors that transmit activation or inhibitory signals which determine if NK cells are activated or not, and thus whether the target cell will be lysed or spared. NKp30 is a type I transmembrane receptor which recognize the stress-induced cell surface ligand B7-H6. Interaction of these two proteins leads to the initiation of immune response. The main aim of this thesis is the preparation of cell ligand B7-H6 with coiled-coil peptide tag in human embryonic kidney cell lines HEK293 GnTI- and HEK293T. Successful preparation of B7-H6 with coiled-coil tag on its C-terminus was verified by mass spectrometry. Its interaction with natural cytotoxicity receptor NKp30 was also proven by sedimentation analysis. Image of Most Region's in Czech Narrative Fiction of the 20th. and 21. century

This bachelor's thesis deals with the research of the image left by the city of Most, or more precisely the whole Most region in Czech prose from 1945 to the present, and does so by means of an interpretative analysis of nine selected works. In the thesis, fiction is approached as a primary source and thus, from the point of view of literary theory, the thesis ranks among context-oriented theories. The thesis is divided into a theoretical and an analytical part; the theoretical part summarizes the history of the area and the city of Most (old and new), with the emphasis put on major events that could be considered turning points of the 20th and 21st century for the city and the region. It also describes the literature both of the Most region and about the Most region. The analytical part contains an interpretative analysis of nine works - by the authors Hořejš, Jedlička, Páral, Volák, Cach, Šerberová, Brycz, Čechura and Bendová - taking place in the city of Most and its surrounding. The analyses themselves are introduced by an explanation of the selection of these works, which was made in such a way that it was both representative and also outlined how the city and its surroundings were depicted in literature. Aplikace age managementu v průmyslovém sektoru

Diploma thesis deals with an issue of age management and its introduction to the selected industrial companies. Literary research of the thesis defines general con-cepts related to human resources, specifications of older employees and their status on labour market and examines problematics of the age management in context of present demographic development in Czech Republic. In the thesis there is a survey amongst the employees of the selected companies. These surveys also contains an interviews with top management. In the final part of the thesis there are recommendations based on the findings which leads to the effective applica-tion of the age management in the industrial sector.
The role of FANCI phosphorylation in the Fanconi anemia DNA repair pathway

Fanconi anemia is an autosomal recessive disorder caused by mutation in one of Fanconi genes and it is manifested by developmental abnormalities, bone marrow failure, predisposition to cancer, cellular sensitivity to cross-linking agents and many other symptoms. Proteins encoded by Fanconi genes and some other proteins are part of Fanconi anemia pathway (FA pathway), which is responsible for DNA repair of an interstrand cross-link (ICL). The repair by this pathway requires monoubiquitination of FANCD2, which is induced and regulated by ATR dependent FANCI phosphorylation. The FANCI phosphorylation initiates the FA pathway but the molecular mechanism of this initialization is not known. Furthermore the proper function of entire pathway requires both: sequence of phosphorylation events of FANCI and monoubiquitination of FANCI:FANCD2 complex . The principle of this work was to study molecular mechanism of initiation and regulation of FA pathway by FANCI phosphorylation. Therefore phosphomimetic mutants of FANCI have been created to investigate their role in processes leading to FANCD2 monoubiquitination. The main aim was to reveal how the phosphorylation of FANCI affects DNA binding and also DNA binding of FANCI:FANCD2 complex. Gargoyles of the European cathedrals, their origin, and their meaning

The bachelor thesis deals with the origin and meaning of gargoyles as well as traditional mythology, where it explores the sources of inspiration for their creation. The work presents examples of the European steel cathedral builders' works, by means of which they describe the way of creation and creation of gargoyles in the environment of the stoneworks. It also looks at the history of gargoyles in architecture, taking into account their symbolic meaning as part of the Gothic cathedral. The practical part refers to the theme of the work, which follows the aesthetics and function of the original gargoyles. The work is made in 1: 3 scale, using clay modeling and subsequent firing. The thesis contains complete photo documentation related to theoretical and practical parts and sketch material.
The God's Eye View: Epistemic Ideal or Conformity with Standards of Justification?

(in English) In the Enlightenment liberation from the religious myth one can see a paradoxical shift that is characterized by the fact that the scientist in the new type of secularized culture takes over the role previously dedicated to the priest. She holds the position of a mediator between what is human and superhuman, subjective, imperfect and flawed and a distant truth to which human beings feel a specific kind of responsibility. As the "soft sciences" ceded territory to the exact sciences, it happened because we have not yet been able to give up the dream of a perfectly neutral God eye's view. The author wants to show that objectivity itself is not impartial, but that it is a name for different kinds of rationality, which are at different times considered to be epistemic virtues.
