Expression of genes for the conversion of nitriles and amides in Rhodococcus erythropolis

The strain Rhodococcus erythropolis A4 is a source of enzymes nitrilhydratase and amidase, that catalyse conversion of nitriles and amides. These enzymes are used in industrial biotransformation and bioremediation. Since it was difficult to carry out genetic manipulations aimed at increasing the production of these enzymes in the strain A4, the corresponding genes (ami and nha1 + nha2) of a related strain R. erythropolis CCM2595, in which both plasmid and chromosome manipulations can be routinely performed, were identified and analyzed in this diploma theses. The ami and nha1 + nha2 genes from the strain R. erythropolis CCM2595 were isolated and sequenced together with the flanking regions (5.5 kb in total). The organization of these genes and the expected regulatory genes was described in the strain CCM2595 and mechanisms of regulation of expression of these genes were studied. For the analysis of transcription of amidase and nitrilhydratase genes from both strains of R. erythropolis, the promoter-probe vector pEPR1 replicating in Escherichia coli and R. erythropolis was used. The need of personal service for people released from the execution of punishment - Znojmo region study

The diploma paper deals with the issue of postpenitentiary care. Concretely the diploma paper is focused on representation of postpenitentiary care in the Znojmo's region and on its following utilization or on eventual need of dissemination of this type of care in this region. In the theoretical part there is described the issue of postpenitentiary care in general followed by the outline of its distribution among the Znojmo's region. There are also presented definitions, brief history, references on legislature. The conclusion of this part is dedicated to characteristic and description of Znojmo's region from postpenitentiary care's point of view. The practical part is closely focused on the needs and opinions of people from postpenitentiary care's sphere in Znojmo's region both specialists and service users.
Support for small and medium sized enterprises in the Czech Republic from national programs and EU funds

The essence of the first part of this work (chapters 1, 2 and 3) lies in the brief introduction to the support for small and medium sized enterprises. After that follows presentation of key institutions (ministries, agencies, etc.) helpful during the process of obtaining support from national programmes. Except of basic services performed by given institutions, useful web links are also included. In the same way, particular European support programmes are presented. In the framework of second part (chapters 4, 5 and 6), backward bases are exemplified on the practical example of the company Strojírny Podzimek, Ltd. and their experiences. That case shows, what does the making use of the national support programmes mean practically. Likewise, business support from EU funds is described, namely on the case of company Frýdlantské strojírny Rasl a syn, Inc. Third part of this thesis (chapters 7 and 8) is making us familiar with resuming statistics about the amount of subsidies drawn from national programmes and from the EU funds. In both cases, brief forecasts of future development and approximate amounts of unused funds are stated.
Education Innovation Possibilities at the Faculty of Management

This work is divided into two parts -- the theoretical and practical ones. Main intention of the theoretical part was to provide comprehend views of current state of tertiary education in our country. For this purpose, particular chapters dealt with key terms, current problems and selected aspects of tertiary education. The outputs of the educational process, i.e. satisfaction of Czech labour market with graduates and the requirements of this market for graduates were not omitted. Main objective resulting from setting of this work, i.e. searching alternatives that may lead to innovative education at the faculty of management, was implemented within the frame of the practical part. First of all, research targets, research questions, research sample and ways of data processing were specified. The key chapter is the fourth one, presenting and interpreting the data collected. Outputs were assorted in tables or graphs, completed by appropriate comments. Crucial points were summarized in the end of the work. Especially the main research conclusions, benefits acquired from this work, recommendations rised for the faculty management and some other interesting findings.
