Prediction of mechanical properties of iron castings by means of numerical simulation
Filipek, Szymon ; Kotas, Petr (referee) ; Roučka, Jaromír (advisor)
The aim of this thesis was to design optimization of the amount of alloying elements (copper and molybdenum) in liquid alloy, which improve mechanical properties of the grey cast iron. An adjustment was made to exceed requirement for a minimum strength in the stressed area. The strength limit was exceeded by approximately 60 MPa. Foundry simulation software MAGMAsoft was used for theoretical optimization. Conformity between the mechanical properties evaluated by simulation and the measured values of hardness, tensile strength was confirmed in the first step of the experimental part. A draft of optimization was created in the second step, which led to savings of the alloying costs.
Various aspects of soil microbial ecology as revealed by phospholipid fatty acid (PLFA) analysis.
KOTAS, Petr
The PLFA profiling method was adopted and used to determine changes in microbial community structure and abundance along natural and human-induced environmental gradients. The presented studies were based on field sampling campaigns combined with targeted laboratory experiments. According to the aims of particular studies, microbial PLFA fingerprinting was combined with the auxiliary below- and aboveground ecosystem characteristics to identify the drivers of microbial responses to environmental changes or with 13C-labelling and metagenomics to obtain more complex information about running processes and involved microorganisms.

Thesis deals with furniture and interior ergonomics for the physically handicapped children in wheelchair. Provides information about the problems of disability and physically disabled children needs. Thesis defines the requierements for creating an amiable interior and barrier-free furniture. The thesis goal is design furniture and room for physically disabled child and define physically disabled child outreach possibilities.
Ensolisation-silylation reaction study of selected steroids
Hehenberger, A. ; Kotas, Petr ; Tříska, Jan
For the purposes of trace analysis of 1,4-androstadiene-3,17-dione (ADD), 1,4-androstadiene-3-one-17β-ol (Boldenone) and 17-β-estradiol in water, waste water, soil and for the further phytosterols transformation studies the enolisation-silylation reaction was studied using MSTFA and BSTFA with different catalysts as derivatization reagents. The different reaction conditions and stability of the products was also studied using GC-MS technique.

Raman Microspectroscopy of Individual Algal Cells: Sensing Unsaturation of Storage Lipids in vivo
KOTAS, Petr
We have demonstrated the potential of Raman microspectroscopy for the fast and spatially resolved characterization of the composition of selected intracellular regions in individual living cells of three algal species. In particular, we have focused on lipid storage bodies and quantified the degree of unsaturation of algal lipids (iodine value). To verify our results, we developed a method for extraction of storage lipids from highly resistant Trachidiscus minutus cells and determined the fatty acid composition using GC/MS. We have found Raman microspectroscopy to be a fast, versatile, and virtually non-invasive tool for evaluating the potential of different algal species for algal lipid engineering and industry.
Liquidation of Brominated Flame Retardants from Waste Water
Vajglová, Zuzana ; Vychodilová, Hana ; Kotas, Petr ; Jiřičný, Vladimír
In this work we focused on the degradation method with a high potential for degradation of polybrominated diphenyl ethers from liquid waste. Very good results were achieved with the chemical degradation methods using nanostructured iron. Electrochemical method gave the best results of degradation of PBDE. Within the frame of this work we have used for the first time an electrochemical microreactor whose application in this context has not been described in the literature. The obtained original experimental results confirm the significantly higher efficiency and speed of degradation in comparison with the classic electrochemical reactor and chemical methods of PBDE degradation.
