The development, constitution and functions of the gland apparatus in schistosome cercariae and endopeptidases in its contents

Flukes of the family Schistosomatidae are blood parasites with two-host life cycles involving aquatic snails as intermediate hosts and avian or mammalian definitive hosts. Cercariae, as invasive aquatic stages of schistosomes, enter the lumen of vessels by penetrating the skin of the definitive host. During their short life, cercariae possess a glandular apparatus consisting of three types of glands with external secretion: penetration glands, escape glands, and a head gland. These glands secrete granules containing, among other things, proteolytic enzymes, which play an important role in the process of penetrating the host's skin, but at the same time they occupy many other functions during the life of the cercaria and other life stages of schistosomes. This thesis summarizes basic knowledge about the anatomy, function and development of the gland apparatus of schistosome cercariae and further focuses on the representation of proteolytic enzymes in these glands, specifically endopeptidases. At the end, it briefly compares the different representation of endopeptidases in some members of the family.

The bachelor thesis deals with insects as a component of poultry feed from economic and marketing view. Thesis is divided into two parts, i.e. literature research, which include history, legislation, expert articles and studies considering insects in poultry feed. Second part of the thesis deals with insects feed from company Underground Food, market research, searching producer of this feed and price comparison of poultry feed with insects. In the end is proposed marketing strategy how to make poultry feed with insects visible on the market.

Calcium is the fifth most abundant element in the human body. In addition to its important role in bone homeostasis, calcium is involved in other processes such as blood clotting, muscle contraction and regulation of nerve excitability Magnesium is the cofactor of many enzymatic reactions essential for a row of physiological functions. The aim of this work was to propose a novel screening method for determination of the degree of calcium and magnesium chelation. In the first step the UV-vis spectra of the selected indicator o-cresolphthalein complexone and its complexes with calcium and magnesium were measured. Analysis of secretome from Trichobilharzia regenti cercariae and characterisation of selected peptidases

(English): Trichobilharzia regenti is a neurotropic parasite of birds from the family Schistosomatidae. Cercariae, the invasive stages of these trematodes actively penetrate the host skin employing excretory- secretory products (ESPs), which contain proteolytic enzymes able to disrupt host tissues and thus reach the successful transmission. The most abundant secreted enzyme responsible for cercarial penetration of the human schistosome S. mansoni is a cercarial elastase. This serine peptidase is well known for the degradation of skin proteins such as elastin, keratin, collagen or laminin. However, the active expression of the orthologue of this enzyme has never been found in the genus Trichobilharzia. For this reason, it was firmly believed, that cercaria of T. regenti uses mainly cysteine peptidases for the invasion of the host, particularly cathepsins, which were repeatedly identified in this life stage. To strengthen this hypothesis, we incubated T. regenti cercariae in the apparatus with the excised duck skin stimulating the release of their glands' content. The collected ESPs were further analysed by shotgun mass-spectrometry and for the first time, the protein form of cercarial elastase was identified. Excretory-secretory proteins of schistosome intravertebrate stages in context of their biology

Schistosomatidae is a family of blood-dwelling trematodes which cause serious disease in humans and warm blooded animals. Human schistosomiasis affects over 258 million people worldwide. The fundamental pathological manifestations of the disease are not caused by adult worms, but by responses to antigens released from eggs, which remain trapped in the tissues. All stages of these parasites release excretory-secretory products, which mostly comprise of proteins which are, to a certain extent, species- and stage-specific. These products have many functions that play roles in successful transmission of the parasite and its interactions with the host. Various approaches to the collection and analysis of these molecules do not always accurately reflect the secretory processes in vivo, because they always employ in vitro techniques. The research of these secreted molecules discovered many mechanisms, through which the parasite successfully modulates and evades host immune system. Despite the fact, that many of these molecules were identified as potential vaccine candidates, still no sufficiently effective vaccine has been developed. Variable Intake manifold for 16V Engine

Aim of the diploma thesis is to design intake ducting with variable cross-section for 16V diesel engine consisting from divided intake manifold and divided ejector. Cross-section variability is ensured by a swirl flap placed in the divided ejector. Next aim of thesis is to provide drawing documentation for production of samples. For designing was used PTC Creo system and software GeomCaliper. Also was taken account of manufacturing of the parts with sand-casting, especially to guarantee trouble-free taking out of models from the moulds, and taking out of cores from core moulds, and also for machinability of some elements. Introduction of thesis is focused on diesel engine emissions and ways of their reduction.
Microprocessor control of sensory platform

This dissertation deals with a development of MCU position steering application of two servomotors and with data process evaluation from a connected sensor for distance measuring as well. The connected sensor is adjustable as for azimuth and elevation. Data from the sensor will be useful both for instant evaluation according to preset criteria and for additional possible processing at PC. The development is to be carry out at MCU ATmega128. HMI is represented with keyboard, LCD and joystick. The result of the dissertation is to be adjusted for practical demo. This research has been supported by the Czech Ministry of Education in the frame of MSM 0021630529 Research Intention Inteligent Systems in Automation.

The outcome of the work will be proposing the concept of franchising for the company AUTO HORÁK. The concept is designed to allow a basis for its expansion.
Design technology for the production of custom advertising article

The project worked out in the framework of the Bachelor's study presents a proposal of producing technology of an advertising article in shape of a razor blade made from stainless steel 316L, in jewellery commonly called surgical stainless steel. Expected batch production is 100 pieces. In the thesis is compared technology for the production on a knee-type CNC milling machine with a laser cutting machine. A production drawing of a razor blade is given as a material for manufacture. A part of this project is an economic evaluation and a conclusion of suggested technology.
