Methods in presenting products of nature in the environmental education in the first years of primary school ? comparison and real possibilities

The thesis aims on the methods of presentation of products of nature in environmental education in the first years of primary school. It mentions the way of teaching and presentation of products of nature in the science subjects and aims on the analysis of the textbooks in focused schools. The results of the thesis contribute to the making of concept of how the presence of products of nature contributes to the knowledge and its retention in the memory of pupils in the first years of basic school.
Characterization and function of the transferrin from the tick \kur{Ixodes ricinus}

Iron metabolism is an important physiological process in ticks which has a great potential for an efficient tick control. This thesis is focused on the characterization and function assessment of the transferrin-related protein in the hard tick Ixodes ricinus. The full sequence Ixodes ricinus transferrin was determined and revealed that this protein is related to the insect transferrins of the type 2 (melanotransferrins). RNA interference silencing of tick transferrin did not affect tick feeding and survival suggesting that function of this protein is not directly involved in the iron transport.
Tests of thermal and radiation embrittlement of reactor steel 15CH2MFA

Results are presented of mechanical destructive and magnetic nondestructive tests of the 15Ch2MFA steel, used for pressure vessels of the VVER40 reactor type. Two series of samples were tested: (i) samples, embrittled by a special thermal processing, and (ii)samples, embrittled in a reactor by high fluences of neutron radiation. Results of mechanical impact Charpy tests are compared with results of Magnetic Adaptive Testing carried out on the two sample series. The existence of mutual correlation between the two types of examination indicates alternative of replacement of the mechanical destructive tests of classical surveillance samples by magnetic tests, which are nondestructive.
VOJTA PRINCIPLE IN THE PHYSIOTHERAPY OF LOCOMOTIVE DISORDERS BY THE CHILDREN´S NEUROLOGICAL PATIENTS

My bachelor thesis deals with reflexology in relation to treatment of children with cerebral palsy (CP). As the Vojta method has been considered the most effective for treatment of patients suffering from any form of cerebral palsy, I have chosen as the main objective of my thesis the evaluation of effectiveness of treatment with the Vojta method. The respondents were two clients of the Arpida center for rehabilitation of handicapped people, who were undergoing treatment for triparetic and diparetic form of cerebral palsy. In the theoretical part I described facts about the disorder, including its etiology, symptoms, clinical manifestations but particularly diagnostics, prevention and treatment. Subsequently, I described the essence of the Vojta method as a diagnostic and therapeutic system. The practical part used a method of qualitative research, based on a secondary analysis of data and direct observation. The monitoring included a standard observation test Gross Motor Function Classification System (GMFCS) and ADL test. To meet one partial objective of the thesis, i.e. analysis of experience of parents with treatment of CP using the Vojta method, I used a method of semi-structured interview. The examination of spontaneous mobility and the quality and quantity of body posture in the development of erective mechanisms has confirmed improvement in both the respondents after the applied therapy using the Vojta method, which has been demonstrated in the final kinesiology analysis. The case studies have shown that effectiveness of the therapy using the Vojta method is influenced by certain aspects, such as frequency, dosing, early beginning of the therapy, proper management of parents by the physiotherapist and the parents´ attitude to the therapy. The Vojta method represents a good basis for treatment of children with cerebral palsy but the rehabilitation shall be always an integral process. In conclusion of my thesis I have recommended to monitor clients in their development by using retardation quotient calculations.
QUESTIONS OF SAFETY IN THE INSTITUTIONS INTENDED FOR WATER HOLIDAY ACTIVITIES

``Water-related injuries{\crqq} are the second most frequent cause of death of children in the Czech Republic. My thesis addresses the issue of safety in facilities designed for aquatic leisure activities. Within the scope of the qualitative research with the aim to analyze technical standards, to ascertain their observance and afterwards to analyze the injury risks I used the data collection technique consisting in secondary data analysis, observation in combination with photographic documentation and semi-standardized interviews. For the research I selected a swimming pool in České Budějovice and Aquaviva Wellness Centre in Aurora Třeboň spa. The reason for this selection was creation of a pilot study monitoring safety in these facilities in South Bohemia. The Czech Republic has technical standards in place that are applied to minimize the risk of injury to the largest possible extent in facilities like that. They determine the procedures that have to be observed during construction and subsequent operation of facilities designed for aquatic leisure activities. The provisions defined by the standards are observed in both facilities but in their substance they are so generalized that it is only at the operators{\crq} discretion to what extent the safety is to be secured by them. They should be more specified and applied concretely to individual units of the equipment in order to create a safe environment. As far as risk places in the facilities are concerned, it may be stated that, regardless of compliance with the requirements defined by the standard, there is a large number of risks related in particular with slippery surfaces. Within the scope of the safety evaluation, it is also necessary to take account of hazardous behaviour of users of the chutes. The results of the research indicate that in Aquaviva Wellness Centre in Třeboň safety is secured and complies with the requirements of technical standards and the prevention requirements; the swimming pool in České Budějovice shows more serious defects {--} the standards are complied with but the surfaces are not covered with materials that would secure safety. On the basis of my research and other technical studies that confirmed seriousness of water-related injuries it is obvious that the issue of injuries in this field will require modification of safety measures and more precise specification of standards and thus elements such as materials of surfaces that would secure the required safety.
Analysis of selected genomic regions with dominant suppressors of the \kur{adgf-a} mutant phenotype in \kur{Drosophila melanogaster}

Adenosine is an important signaling molecule in various physiological processes in many different organisms. Dysregulation of the level of this molecule leads to many pathological consequences as Severe Combined Immunodeficiency in humans. Recently a genetic model using fruit fly Drosophila melanogaster was established to study effects of adenosine in this organism and a genetic screen for genes modifying the effects of heightened adenosine was started. This work presents partial tasks in this screen which include detailed characterization of selected regions which were identified as dominant suppressors of the phenotype associated with heightened adenosine. First task was to produce a smaller, molecularly defined deletion in range of originally identified bigger deletion. Second task included a testing of already existing smaller deletion for the suppressing effects. In the third task, a role of potential suppressor was further analyzed which included a verification of P-element insertions in this gene and their testing for suppressing effects. In the last task, the sequence for making a probe to characterize the expression of this gene was cloned
