Evaluation of the Financial Situation in the Company and Proposals to its improvement

This Bachelor thesis is focused on Krnovske opravny a strojirny s.r.o. company´s financial situation from the year 2015 to 2019. The theoretical part explains some ecomic indicators, formulas, and methods needed for financial analysis. In the analytical part, the methods that apply findings from previous part are described. The last part of the thesis deals with suggestions and adjustments to improve current situation of the company.
A financial plan of business development

This thesis is focused on the design of a financial plan for the comany Krnovské opravny a strojírny s.r.o. and is divided into three parts. The first part deals with the theoretical background, the knowledge of witch is necessary to create a financial plan. This part is followed by the practical part, which deals with the strategic and financial analysis of the selected company. In the last part, a draft corporate financial plan for the next four years is developer. The final part consists of its evaluation using selected financial ratios and sensitivity analysis.
Synthesis and evaluation of potential antitubercular drugs based on isoniazid

Isoniazid is a first-line drug used against tuberculosis. It is a bactericidal drug with a selectivity for Mycobacterium tuberculosis (Mtb.). The mechanism of action is primarily based on the blockade of mycolic acid synthesis and thus cell wall synthesis. The development of resistance is limiting the therapeutic potential of isoniazid and that is the reason for the development of its new structural modifications. This diploma thesis is focused on synthesis and evaluation of novel isoniazid analogues based on a hydrazone obtained from isoniazid and glyoxalic acid. The free carboxyl group was further modified by various amines to form amides. A total of sixteen substituted 2-(2- isonicotinoylhydrazono)-N-phenylacetamides were prepared and tested for their in vitro antimycobacterial activity on selected strains of mycobacteria - Mtb., M. avium and M. kansasii. The best activity against Mtb. was shown by (E)-2-(2- isonicotinoylhydrazineylidene)-N-(4-propylphenyl)acetamide and (E)-N-(4-butylphenyl)-2-(2- isonicotinoylhydrazineylidene)acetamide, their minimal inhibitory concentration (MIC) is 0.125 µM compared to isoniazid's MIC of 0.5 µM. They were also active against nontuberculous mycobacterium M. kansasii (MIC from 2 µM). Their activity against multidrug- resistant strains was lower due to...
Health risk and profit of genetically modified golden rice

People in developing countries very often suffer from severe vitamin A deficiency due to an insufficient and unbalanced diet. This vitamin is necessary, among other things, for the production of the visual pigment rhodopsin. According to the World Health Organization, up to half a million children go blind each year because of this deficit. Its deficiency also weakens the immune system and thus significantly increases the risk of death from various infectious diseases. The best solution to this deficiency would be if the vitamin was contained directly in the only food that these people get - in husked rice. There is already a special, genetically modified rice, into which has been introduced by a complicated and extensive modification of its DNA the entire metabolic pathway, which ensures the production of betacarotene in this crop. In the theoretical part I deal with the golden rice project and the origin of this genetically modified crop by the method of transgenesis, using bacteria of the genus Agrobacterium tumefaciens. Plasmids of this bacterium are able to incorporate parts of their genetic information into the target organism. Thanks to restriction enzymes, we can insert genes selected by us into plasmids, which we then introduce into a specific plant. But are these products safe? Opinions on these crops are very diverse, but one thing interests everyone: how to safely identify a genetically engineered plant? In the laboratory, these experiments can best be mastered on model material. Therefore, in the experimental part of my bachelor thesis I deal with the transgenesis of the model plant Nicotiana tabacum using a selected strain of Agrobacterium tumefaciens. These bacterial strains were provided to me from the private collections of the Institute of Molecular Plant Biology in České Budějovice. The aim of my work in the laboratory was to master the practical methodology of preparation of genetically modified plant and subsequent verification of the presence of introduced genes in the examined samples of the model organism. Specifically, genes for antibiotic resistance. DNA isolation, PCR amplification and electrophoretic assays were used for this purpose. The signal gene used in agroinfection was verified too. Also, it was monitored how many copies of the transgene were integrated into the research material during agroinfection.
A Comparative Analysis of Foreign News in the Main News Sessions of TV Stations ČT 1, Nova and Prima in 2010

Diploma thesis "A Comparative Analysis of Foreign News in the Main News Sessions of TV Stations ČT1, TV Nova and Prima TV in 2010" studies the way of producing foreign news by Czech TV stations in the year 2010. Also, it focuses on the question which countries were mentioned. The introductory part discusses the Czech and foreign literature and offers the overview of TV news production, international flow of communication and factors which influences the production of foreign news. It also introduces the main news sessions Události, Televizní noviny and Zprávy TV Prima. The analysis of foreign news service is divided into two parts, first inquires into differences in used types of reports, their representation in the main news sessions, the topical structure and other characteristics connected with the production of news. Then, the second part concentrates on the states which were mentioned in the foreign news and focuses on the impacts influencing the production of foreign news by the Czech TV stations. For both parts of the analysis methods of quantitative research were used. The thesis also studies the differences between the main news sessions of private TV channels TV Nova and Prima TV and ČT1 which offer public service broadcasting. A Child in an Armed Conflict in the Light of Article 38 of the Convention on the Rights of the Child The thesis aims to transparently elaborate the topic of protection of children in armed conflicts pursuant to the Article 38 of the Convention on the Rights of the Child as well as identification of the main problems associated with the topic and efforts to suggest their solutions. The thesis also focuses on monitoring and enforcement mechanism of individual international instruments. The body of the thesis is divided into four parts, each of several chapters. The first part of the thesis deals with the development of protection of children in armed conflicts until the adoption of the Convention on the Rights of the Child, and relevant international law documents on the protection of children in armed conflicts. The first part also includes a chapter with the identification of the main problems associated with the protection of children. Development of HPLC method for determination of vancomycine in clinical research

The chromatographic separation was carried out by KinetexTM C18 column, 2.6 μm particle size, 50 × 4.6 mm (Phenomenex, USA) in combination with potassium phosphate buffer (pH 4.5) and acetonitrile (90 : 10, v/v) as the mobile phase. As internal standard was chosen cefuroxime. For analytes determination UV detection at 220 nm wavelength was applied. After optimization of separation conditions, the method was applied to biological material. Samples were easily modified using the protein precipitation. The method was partially validated. Their capability to encapsulate both hydrophilic and lipophilic molecules, biocompatibility and biodegradability of lipids make them a suitable alternative for well-established drug carries. The aim of this thesis was to determine suitable ratios of composition of nanoparticles with acceptable properties (especially reduced size and polydispersity, high zeta potential absolute values), to investigate status and thermodynamic behaviour of the nanoparticles and lipids used and to examine drug encapsulation efficiency. Nanoprecipitation method was used to prepare nanoparticles from stearic acid as a solid lipid and in the case of NLC preparation isopropyl myristate as a liquid lipid was used.
