RNDr. Dalibor Šatínský, Ph.D. Title of Diploma Thesis: HPLC method development and validation for analysis of anthocyanins in highbush blueberries HPLC method for simultaneous determination of anthocyanins: delphinidin-3-O-galactoside, delphinidin-3-O-glucoside, cyanidin-3-O-galactoside, cyanidin-3-O-glucoside, cyanidin-3-O- arabinoside, peonidin-3-O-glucoside and malvidin-3-O-galactoside in 21 varieties of blueberries ('Spartan', 'Sunrise', 'Toro', 'Bluegold', 'Croatan', 'Northland', 'Duke', 'Gila', 'Jersey', 'Bluecrop', 'Herbert', 'Berkeley', 'Patriot', 'Darrow', 'Bluetta', 'Collins', 'Brigitta', 'November', 'Iranka', 'Bluejay' and 'Rancocas') was developed and optimized in this thesis. Kinetex PFP column, 150 x 4.6 mm (particle size 2.6 µm) was used for the analysis. The detection was performed by DAD detector at 520 nm, column temperature of 50řC using gradient elution with a mobile phase of acetonitrile/ 2 % formic acid at a flow rate of 1.0 ml/min. Comparison of selected vegetation indices and determination of suitability for yield description on agricultural field

Calculation of vegetation indices (VI) is an effective method how to evaluate a condition of a monitored vegetation. It is a non-destructive and low-cost approach and its results are crucial for planning agronomic management within a monitored field. Diploma thesis deals with the determination of suitability of selected VI to describe the yield variability related to topographic attributes and weather conditions. The study area was a 11.5 ha field. To derive selected VI LANDSAT 5 images with a spatial resolution of 30 m were used. These images were taken in years 2005 and 2011 for winter wheat and in years 2006 and 2010, when oat was grown. SW ENVI and ArcGIS were used to obtain specific values. Those were used for statistical analysis. The results indicated a strong relationship between all selected VI and yield, respectively topographic factor. Selected VI derived from LANDSAT 5 images can be used to describe yield variability over a monitored field.
Multifunctional house

The aim of the thesis is to project a family house with a workroom and to work out a plan. The house is being built on building site no. 1127/28 in the town of Hoštka. The project is developed for four to six residents, with the workroom (separated side entrance). Self-contained house ground plan is in the form of the letter L (47.57 × 50.85 ft.). Disposition of the projected single storey house is 5+1 with the habitable attic. There are entry foyer, hall, bathroom, toilet, living room, utility room, garage, workroom with kitchenette, toilet and multifunctional lobby on the first floor. Hall, bedroom, two other rooms and bathroom with toilet are located in the habitable attic. The house is projected on concrete strip.
