Checker of User Experience Guidelines

The aim of this work is to create a tool for automated checking the correct characteristics of the user interface. The thesis includes a list of rules from GNOME, KDE and OSX, which are suitable for automatic control. The tool verifies the background elements of the user interface while the user operates the program under test. The rules are ranked according to the severity and outcome of the program is also overall score. The final part contains testing of normally available programs. The application is appropriate for verifying a graphical user interface in development.
Information System for e-Learing Courses Testing

A long-term trend in modern digital education is the shift from static content to interactive e-learning courses. The focus is often on the form of information delivery rather than the content. The testing part of development tends to be ineffective, as there is no easy way to obtain information about student behaviour. This paper addresses this problem by creating a testable web application, xTester, that allows tracking key metrics of developed e-learning courses communicating through xAPI. The solution includes an enhanced course template in Articulate Storyline and a browser add-on to facilitate linking of created courses to the xTester application. General issues and specific stages of development are described, from specification of user requirements, interface design and implementation, to testing. The frontend of the web application is based on the Vue framework, the backend consists of a simple stateless Rest API in Node.js with a SQLite database.
Regulatory mechanisms of ornithin transcarbamylase and beta-glucocerebrosidase gene expression and their relevance to diagnostics

5 Abstract Definitive diagnosis of inherited metabolic disorders commonly depends on the measurement of enzyme activity (which is often complicated) and/or molecular genetic testing. Yet even the standard mutation analysis can bring false negative results in the case of gross chromosomal rearrangements or incorrect regulation of gene expression due to the mutations in regulatory regions. In the present study I focused on characterization of complex mutations affecting the gene encoding ornithin transcarbamylase (OTC) followed by studies of regulatory regions of OTC and GBA (the gene encoding β-glucocerebrosidase). In the first study we identified 14 novel mutations including three large deletions in a cohort of 37 patients with OTC deficiency (OTCD). Subsequently we evaluated clinical significance of all these mutations. We also found a heterozygote carrying a hypomorphic mutation and manifesting OTCD most likely due to unfavorable X-inactivation which was observed independently in three different peripheral tissues. In order to evaluate the clinical significance of a promoter variation c.-366A>G found in a family with mild OTCD we identified three alternative transcription start sites (TSSs) of human OTC and delimited the promoter. Function and regulation of ETV4 and MSX1 transcription factors in colon cancer progression

Colon cancer causes approximately seven percent of all cancer-related deaths in the world and presumably due to modern lifestyle, it is also one of the most frequently diagnosed cancers. The inefficiency of standard treatment indicates the need for intensive research of molecular mechanisms of cancer development. The canonical Wnt signaling pathway is essential for maintenance of the progenitor phenotype of stem cells in crypts of the intestine and controls repopulation of the epithelia, in physiological conditions. However, aberrant activation leads to tumor formation. Although Wnt signaling in cancer has been subjected to thorough investigation, there is still a lot of questions concerning further branching of the pathway. As a model of Wnt/β-catenin triggered colorectal cancer, we use mice with mutated APC, which is the tumor suppressor involved in this pathway. Previous expression profiling of the intestinal tumors from relevant mice revealed two transcription factors: ETV4 and MSX1 which are significantly overexpressed in cancer cells. In this project we elucidate whether the overexpression is really tumor restricted and Wnt dependent or there is a crosstalk with another signal transduction pathway.
