Characterization of minor fruit varieties in terms of use in the food industry

The thesis deals with one group of small fruits, the genus Ribes, ie currants, and gooseberries. It assesses their physical and chemical parameters with respect to the possibility of use in these properties for the design of a new type of product – beverages. Titration, spectrometric and other methods were used for the evaluation of these properties. A total of fifteen of these methods were available (total juice yield, juice pH, total dry matter, soluble fruit dry matter, titratable acidity, formolic number, reducing carbohydrate content, D-glucose, D-fructose and sucrose content, total polyphenolic substance content, anthocyanins, vitamin C and total antioxidant capacity) and a total of nine analytes (copper, iron, magnesium, phosphorus, zinc, manganese, potassium, calcium and sodium) were selected for the determination of mineral elements. For two years (harvest 2014 and 2015), these parameters were monitored for color variants of currants (white, red and black) and gooseberries (green-fruited, yellow-fruited and red-fruited). A total of six white varieties were tested in the group of currants - Olin, Jantar, Primus, Blanka, Viktoria and Orion, eleven varieties of red currants - Junnifer, Jesan, Detvan, Rovada, Rubigo, J.V.Tets, Tatran, Losan, Kozolupský raný, Stanca, NŠLS 11/6 and eleven varieties of black currant varieties - Ometa, Démon, Triton, Ben Hope, Ruben, Ben Gairm, Ben Lomond, Moravia, Ben Conan, Fokus and Ceres. A total of five varieties of green-fruited gooseberries were tested - Zebín, Mucurines, Rixanta, Rodnik and Prima. Yellow-fruited gooseberries were available in four varieties - Lemon Giant, Invicta, Golden Fig and Darek. The red-fruited gooseberries included varieties - Rolonda, Alan, Karat, Karmen, Krasnoslawjanskij, Himnomacki Rot, Remarka, Tamara and Black Neguš. Using Tukey's test, an analysis was performed and by scoring individual varieties based on the results of individual analyzes, a list of recommended varieties was compiled for further work on the development of a new beverage. Unfortunately, in 2015 there were very few fruits of green-fruited and yellow-fruited gooseberries, so it was decided that the volume of fruit will fall on the testing of bioactive substances. Therefore, at the end of the work, it is not possible to objectively evaluate these varieties of gooseberries in the whole range of analyzes. Therefore, the point evaluation of varieties was based on the averages of individual varieties. The red-fruited varieties were available in full, and therefore a full evaluation could take place. The most significant differences between red gooseberry varieties were in the content of bioactive substances (the content of polyphenolic substances, anthocyanin dyes, and vitamin C) and antioxidant capacity. These results were provided to the food subject, which continued to work with these values. Based on this project, utility model No. PUV 2016-33171 and the resulting combined drink based on wine and fruit juice were developed.
Chromatographic determination of simple sugars in selected beverages

An important part of the drinks are simple carbohydrates, because this thesis focuses on this issue. The theoretical part describes the characteristics , production , metabolism and the use of simple carbohydrates such as glucose, fructose and sucrose. There is also the definition of drinks and their main divisions . The literature review is based on the experimental part of analyzing selected beverages by high performance liquid chromatography with refractometric detection coupled to columns or alkyl ion-exchange phases. In conclusion, the results were compared to determine and comment . Nowadays the emphasis is on the correct functioning of the digestive tract. An important factor that supports the intestinal microflora is a prebiotic carbohydrate - tagatosa This could become tagatosa whose properties , production, metabolism in the body and any use is also included in the search . In the experimental part is listed possible optimization analysis of samples containing tagatosy on both types of columns in the HPLC system. The result is again commented in conclusion.
Amount of selected biologically active compounds in Stevia rebaudiana

This bachelor thesis deals with plant, called stevia rebaudiana and analysis of phenolic compounds in the extracts from it. The theoretical part describes its botanical characteristics, chemical constitution and effect on humans health. This thesis is interested in sweetener, produced from stevia, comercionally used as E 960: steviol-glykosides, too. There is listed register of companies producing this sweetener and food products, where stevia is used in. The experimental part of thesis is focused to analysis of total phenolic content in stevia extracts, because leaves of stevia are primary used for sweetening of tea. The main aim of the thesis was find out, how is total phenolic content changed during extraction of leaves in time, in water with different temperature and in different samples.
Basic chemical characteristics of juices from selected red currants varieties

This bachelor thesis is focused on the determination of basic chemical characteristics of selected red currants varieties from the Research and Breeding Institute of Pomology Holovousy Ltd. The theoretical part concentrates on general information about red currant, such as history of cultivation, origin and its expansion. There is also mention of the general characteristics of currant, currant building and significance of red currant cultivation. Chapter "Individual varieties" deals with the analysed varieties of red currants, especially its description and its biological properties, followed by an overview of the main substances contained in red currants and their brief description. Bachelor thesis also presents examples of the processing and the utilization of red currant in the food industry. The final chapter provides information about the individual determinations of the chemical characteristics of red currants, which include juice yield, soluble solids content, total dry matter content, pH, titratable acidity, formol number and reducing sugars. The experimental part describes the specific conditions and methods of individual determinations, followed by a summary of the results and their discussion. Variety Jesan showed the highest juice yield (63 ml per 100 g) and the highest pH value (3,16). The highest soluble solids content (13,94 %) and the highest formol number (22,83 ml of 0,1 M NaOH per 100 g) was determined in a variety Jonkheer van Tets. The highest dry matter value was detected for variety Rovada (14,48 %). The titratable acids content was highest in variety NŠLS 11/6 (403,88 mmol•kg-1). The highest concentration of reducing sugars was found in variety Tatran (8,17 %).
Prebiotic sweetener tagatose

This sweetener, D-tagatose, has numerous advantages, which would eventually be able to use in the food industry. It summarizes the properties of Dtagatose, physical and also chemical, the possibility of production from lactose or bacterial action on Dtalitol. The basic information about the properties of Dtagatosy also discusses the actual metabolism of Dtagatose, prebiotic properties and their effect in the gastrointestinal tract. Dtagatose is a proposed new sweetener, because the possibility can not be missed in certain applications tagatose in a different types of foods for example confectionery. And positive qualities in comparison with sucrose. This comparison is made to examples of disease associated with excessive use of conventional sweeteners sucrose. This paper also discusses possibilities Dtagatose determination in foods by different methods.
Determination of the basic physical and chemical parameters of fruit juice from aronia

Theoretical part of bachelor´s thesis contains basic information about aronia (Aronia melanocarpa) and examples of their use. There is a description of analysed substances, which are contained in aronia berries, and their importance for human organism. Experimental part is focused on determination of minerals, saccharides and vitamin C in aronia juices, syrups, nectar and aronia wine. Elements were determined by inductively coupled plasma optical emission spectrometry. Liquid chromatography was chosen for analysis of saccharides and vitamin C. All of analysed products are significant sources of macroelements. The highest concentrations of elements were determined in the sample named Aroniový skorosirup, which was sugared by vaporized apple juice. Fructose and glucose were contained in all products. Sucrose was only in products which were sugared. The amount of vitamin C was lower in consequence of pasteurization and storage.

Detection of Oxidative Stress Using Electrochemical DNA Biosensors

Presented Diploma Thesis is focused on the development, characterization, and utilization of simple and inexpensive electrochemical DNA biosensor for the detection of DNA damage caused by oxidative stress. The initial part of the work is devoted to preparation and characterization of a large-surface carbon film electrode (ls-CFE) modified with carbon nanotubes (CNT/ls-CFE). Carbon nanotubes improve electrochemical properties of the transducer and increase the amount of adsorbed DNA on the electrode surface. Testing of the electrode surface modified with multiwalled carbon nanotubes (MWCNT) was performed using cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS) using a redox system [Fe(CN)6]4-/3- and using square wave voltammetry without any redox indicator. Carbon nanotubes have proved to be unsuitable material for our type of biosensor, but it can be used inanalytical chemistry for the determination of electroactive substances. The second part of this Thesis deals with the application of the prepared DNA biosensor for the detection of DNA damage by oxidative stress. Novel Electrochemical Biosensor for the Detection of DNA Damage Based on a Large-Surface Carbon Film Electrode

Presented Bachelor Thesis is focused on the development and utilization of a simple and inexpensive electrochemical DNA biosensor for the detection of DNA damage caused by chemical carcinogens. A large-surface carbon film electrode (ls-CFE), having several advantages, such as its fast preparation, a simple mechanical renewal of the electrode surface, a good reproducibility of measurements, an absence of problems connected with "electrode history", and simple chemical modification, was used for its preparation. The initial part of the work is devoted to the investigation of the composition of a carbon ink suspension, which was optimized and tested using a [Fe(CN)6]4-/3- redox system by cyclic voltammetry (CV), further to the optimization of the DNA biosensor (DNA/ls-CFE) preparation and to its characterization, which was performed using two electrochemical techniques - CV and electrochemical impedance spectroscopy (EIS). The second part of this Thesis deals with the application of the newly prepared DNA biosensor for the detection of DNA damage by model chemical carcinogens.
