This diploma thesis deals with the reconstruction of the high-rise block of flats in the city district Brno-South. The thesis concentrates on the evaluation of the economical effectivity of this project using two ways of financing. It focuses primarily on planning of the individual finance flows of the project, which will arise in connection with the operation of the high-rise block of flats. The part of the thesis is also the calculation of the basic economic indicators in two ways of financing, by which is it possible to objectively assess the investment and evaluate the favourableness of the financing by own sources and financing by bank loan. In conclusion, the comparison of these two ways of financing will be performed.
Host immune response in cutaneous versus visceral form of leishmaniasis
Matějková, Barbora ; Leštinová, Tereza (advisor) ; Jelínková, Kristýna (referee)
Parasitic protozoans of the genus Leishmania circulate between vectors and hosts during their life cycle, in which they come into contact with the immune system. In the host body, infection can lead to the development of a disease called leishmaniasis. This can manifest itself in a number of ways, with the best known forms being referred to as visceral, mucocutaneous and cutaneous leishmaniasis. This thesis focuses on the host immune response during the cutaneous and visceral forms of leishmaniasis. In addition, attention has been given to host, vector and parasite factors that differ between cutaneous and visceral leishmaniasis forms and that may influence the different clinical manifestations. Attention is paid not only to factors related to immunity, but also to factors such as parasite inoculum dose sizes, temperature differences between skin and internal organs, parasite genetics, and others. Keywords - Leishmania, visceral leishmaniasis, cutaneous leishmaniasis, immune response, parasite, immunity
Factors affecting the protective effect provided by sand fly saliva immunization on the outcome of Leishmania infection
Jelínková, Kristýna ; Kolářová, Iva (advisor) ; Lipoldová, Marie (referee)
ENGLISH VERSION Leishmaniasis is a disease caused by protozoan parasites of the genus Leishmania, transmitted by its vector - female sand flies during the process of blood feeding. Preexposure to sand fly saliva prior to Leishmania infection affects the host immune system, leading to possible establishment of protective effect on the disease outcome. This effect is based on the delayed type hypersensitivity reaction to sand fly saliva which has been shown to peak about 48 hours after antigen encounter, leading to the more effective host immune response towards the Leishmania parasites. In this project, we found this protective effect as species-specific, additionally dependent also on the timing of the infection post last immunization. Results of presented experiments showed that infection of BALB/c mice with Leishmania major performed after 48 hours post last Phlebotomus duboscqi sand fly saliva immunization, did not confer protection in comparison to infection performed at one-week after the last exposure. Moreover, the effect of the sand fly saliva pre-exposure was at this shorter timing of infection rather diminished, supported by insignificant differences in the measured parameters compared to the unpre-exposed infected control group. Additionally, immunization with sand fly Sergentomyia...
The Evolution of the position of the enclitic se in Bohumil Hrabal's short story collections
Jelínková, Kristýna ; Mareš, Petr (advisor) ; Adam, Robert (referee)
This thesis tries to describe the evolution of the position of the encliticon se in Bohumil Hrabal's short story collections from two different periods of his literary production, specifically in Perlička na dně (collection of 1963, "Pearl at the bottom") and Krasosmutnění (collection of 1979, "Beautiful sadness"). The thesis should complete and evolve already existing observations that find Hrabal's substandard usage of enclitics an important attribute of his writing style and also that this usage gives his prose an aspect of spoken language. The first part of the thesis encapsulates main characteristics of his author's style - my summary leans on several studies from important literary theorists such as Milan Jankovič or Josef Štěpán. The next part serves also as summary, but this time it epitomizes studies about the unmarked position of enclitics, not only Jacob Wackernagel's Über ein Gesetz der indogermanischen Wortstellung (On a Law of Indo-European Word Order) but also some studies from Czech linguists, such as František Trávníček, Ludmila Uhlířová or Václav Ertl. The thesis uses electronic corpora as the base for the quantitative and qualitative analyses and draws conclusions on the usage of the enclitics se in both phases of Bohumil Hrabal's literary work. Key words: encliticon se, position...
The role of branched-chain amino acids in nutrition of strength athletes
Jelínková, Kristýna ; Caha, Jan (advisor) ; Vebr, Pavel (referee)
These are a significant group of essential amino acids - branched chain amino acids, including leucine, isoleucine and valine. There is evidence to suggest that supplementation of branched amino acids can be very beneficial for synthesis and reduction of muscle protein degradation in strength athletes. On the other hand, there are studies to find that BCAA supplementation is not useful for restoring damaged muscles and alleviating pain after exercise as expected. The aim of this work is to summarize the issue of branched amino acid metabolism and to compare their effects when used in the form of BCAA supplements in strength athletes. Keywords: amino acids, leucin, BCAA, fitness, strength, muscles
Time perspective and fear about the future of pupils
Jelínková, Kristýna ; Pavelková, Isabella (advisor) ; Škaloudová, Alena (referee)
The thesis deals with the relationship between the time perspective and the concerns of second grade elementary school students. In the theoretical part we briefly introduce the time perspective and various theoretical views on it. Furthermore, we will get acquainted with the theory of Zimbardo and Boyd, from whose theory of the time perspective we are based on and use their diagnostic tool to examine the time perspective. Then we will introduce fear, its various concepts and define it against anxiety and concerns. Subsequently, we will focus on the theory of J. A. Gray, which we have chosen to examine fear. In the empirical part of the thesis we will present our research sample, which are pupils of the 8th and 9th grade of elementary school. Our goal is to explore the relationship between the future and fear in these students, and we also want to chart the specific concerns they have, about the future. We used both quantitative and qualitative methods to examine our goal. Quantitative methods include the Swedish version of the Zimbardo Time Perspective Inventory (S-ZTPI) and the BIS / BAS questionnaire. Qualitative methods include open-ended questions, which we want to use to chart the specific concerns of the students and it will also help us to illustrate the results from the quantitative...
Interactions of human immune system with saliva of bloodfeeding Nematocera
Jelínková, Kristýna ; Kolářová, Iva (advisor) ; Janda, Jozef (referee)
In humans, the bites of bloodfeeding insects from the suborder Nematocera induce an immune reaction, both humoral and cell-mediated. Some antigens presented in the saliva of some insect families - Psychodidae, Culicidae, Simuliidae and Ceratopogonidae - are studied more deeply to reveal their immunomodulatory and antigennic properties. Most studies are focused on mosquitos (Culicidae) and sand flies (Psychodidae). Mosquito saliva elicits primarily IgG and IgE antibodies. The level of antibodies in the sera of bitten individuals reflects the length and intensitity of previous exposure to insect bites. Anti-saliva IgE antibodies play an important role in response to the mosquito bites and are frequently associated with allergic reactions. On the other hand, sand fly saliva elicits primarily IgG antibodies. Cell-mediated human immune response to mosquito bites is a neglected research topic. It has been proven that the saliva of sand flies (genus Phlebotomus and Lutzomyia) stimulates proliferation of PBMC from repeatedly bitten humans. Cytokine production by human PBMC and expression of costimulatory molecules in human monocytes, macrophages, and dendritic cells are also influenced by the presence of sand fly saliva. So far as we know, there are only few studies on human immune response to black fly...
