Denisa Janů Supervisor: PharmDr. Ondřej Janďourek, Ph.D. Title of diploma thesis: Current possibillities of laboratory diagnostics of EBV Background: The aim of this diploma thesis was to get acquainted with the issue of Epstein-Barr virus infection and its laboratory diagnostics. Other aim was to divide the patients into groups according to the serological results, then evaluate the prevalence of EBV and evaluate the incidence of primary EBV infection in the tested group of patients for the years 2019-2022. Methods: Chemiluminescence immunoassay and indirect immunofluorescence were used to determine specific antibodies against EBV. In some cases, the examination was completed by determining the avidity of anti-VCA IgG antibodies and the microblot array method. Real-time PCR was used to detect EBV DNA. Microsoft Excel 2016 was used to process data and to create tables and graphs. Results: For the years 2019-2022, 14,736 patients were tested for the presence of specific antibodies against EBV in the infectious serology and virology laboratory of Vidia-Diagnostika, 263 patients for the presence of heterophile antibodies...
Laboratory diagnostics of Hepatitis B

Hepatitis B is an inflammatory viral disease of the liver. The etiological agent is a DNA virus from the Hepadnaviridae family. It is transmitted through the blood and secretions of an infected person. The greatest danger of the disease lies in the transition to chronicity. Chronic infection carries a high risk of developing liver cirrhosis and hepatocellular carcinoma. Hepatitis B is widespread throughout the world. The most reliable prevention is vaccination and avoidance of risky behavior and contact with persons with risky behavior, especially drug users and promiscuous persons. The aim of this bachelor thesis is to get acquainted with the disease of viral hepatitis B and to describe the current possibilities of diagnostics of hepatitis B markers using the COBAS e601 device from ROCHE by electrochemiluminescent immunoassay and quantification of viral DNA on the COBAS 4800 from ROCHE by real time PCR and evaluation of results using both methods. The set of examined samples comes from the laboratory of infectious serology and virology of the company VIDIA-DIAGNOSTIKA. The results show that HBsAg is the most investigated marker of hepatitis B. Positive findings make up only 0.67%. The ratio of HBsAg positive men and women in 2019 was almost balanced. Most HBsAg positive patients were aged 30-40 years. I also examined the prevalence of anti-HBc Ig total antibodies. The positivity of anti-HBc Ig total was confirmed in 4.27% of examinations. The ratio of anti-HBc Ig total positive men and women in 2019 was also almost balanced and the most positive were at the age of 30-40 years.
Algorithmization development tasks with minirobot Ozobot

This thesis deals with creating a set of exercises for Ozobot with focus on algorithmization. The exercises are meant to be used printed on paper. But they can be used with either tablet or computer monitor to a limited extent. The exercises themselves are build upon programming via graphic codes. Goals, methods and problems while creating the exercises are described in the introductory part. The minirobot Ozobot, its various versions, their differences and usability is described in theoretical part. Programing with graphic codes and programing in block enviroment OzoBlocky are described there. It also contains a part about current goals and trends in teaching informatics and computional thinking. Currently available exercises are also described there. The practical part contains created set of exercises divided in chapters according to their focus. Each exercise has a set goal, notes for teachers, possible expansions and changes made on it.
Equations at high schools

The Bachelor's thesis is focused on the issues of equations, taught on elementary and high schools. Students make numerous errors when solving them, which stem from the wrong ways of solving these equations, but more importantly from assembling them. The first part of the thesis is focused on the types of equations (linear, quadratic, ...) and the ways of solving them. In the second part is the usage of equations when solving given problems. These word problems are solved through the usage of equations and the through the student's judgement, that most schools don't develop. There are exercises at the end of the thesis.
