Vývoj molekulárních markerů pro chromozom W u modráska \kur{Polyommatus icarus}

This thesis focuses on development of molecular markers for the W chromosome in the blue butterfly Polyommatus icarus. In this blue butterfly, karyotype races with different constitution of the neo-sex chromosomes were reported. The markers were used for PCR sexing of the early developmental stages to obtain sex-specific total RNA samples for testing a role of sexually antagonistic selection in fixation of neo-sex chromosomes.
Analýza repetic v genomech vybraných druhů modrásků rodů \kur{Polyommatus} a \kur{Lysandra}

This thesis focuses on the analysis of mobile elements in the genera Polyommatus and Lysandra (Lepidoptera) with a potential impact on karyotype fragmentation. The presence of selected mobile elements in genomes of 15 lycaenid species was tested by PCR. Moreover, the same method was used to detect these elements in 13 selected ant species, which may present a source for lateral gene transfer in myrmecophilous blue butterfly species. Selected transposable elements were localized in pachytene nuclei using fluorescence in situ hybridization. The results of this thesis suggest a patchy phylogenetic pattern of studied repeats which can be partly explained by mobile elements spread through interspecific hybridization and horizontal gene transfer among studied Polyommatus and Lysandra species.
John Hus - Library and their Progression in Centuris

Following dissertation deals with the analysis of verbal symbols expressed in two works of Jan Hus: "O poznání cesty pravé k spasení (Dcerka) a Knížky o knězi kuchmistrovi". The first work is based on several manuscripts. The second is preserved in only one old print. This work first introduces the reader to the time of book origin, with charakter and ideas of Jan Hus. After that it is trying to define the term symbol. Each book is analyzed in terms of content and time of occurrence. Author divided symbols into several categories. His work deals with symbolism of characters, animals, body parts and abstract.

Catholic Reformation in the Prague Towns after the Battle of White Mountain. Prague's New Town in a context of the process of the catholic confessionalisation Abstract Olga Fejtová This thesis' methodological ground is a conception of both clerical and secular politics of recatholisation in a relation to Prague's New Town within the Post-White-Mountain period as part of a process of confessionalisation, i. e catholic confessialisation, within the paradigm that was introduced into European historiography in 1980s . The recatholisation within Bohemian lands in relationship to the paradigm of confessionalisation invokes a strategy of state and clerical power. Its enforcement and exercise in political practice were illustrated by normative materials that represented attendant signs of a development of the catholic confessionalisation. The target of this thesis is firstly to describe the progress of politics concerning recatholisation in a relationship to one of Prague's towns by using contemporary decrees of both secular and clerical origin. Furthermore, it shall investigate issues echoing within and from life of the New-Prague-Town's society of the 17th century. This within both public and private spheres of burgess' life. The thesis "Production and Evaluation of Fruit Wines" was prepared in 2016/2017 at the Institute of Postharvest Technology of Horticultural Products at the Faculty of Horticulture of the Mendel University in Brno. The thesis deals with the production of fruit wines with a focus on their fabric composition. In the practical part, based on analytical and sensory analyzes, samples of wines, laboratory samples of wines from the wine-growing and retail networks were evaluated. The data obtained were statistically compared and appropriately interpreted. The currant and cherry wines were sensitively evaluated above the apple. And currant and cherry wines have shown a higher antioxidant capacity.
Implementation of New Methods for Studying the Molecular Genetic Basis of the CADASIL Disease

CADASIL is a neurodegenerative autosomal dominant hereditary disease with late onset. Main symptoms are migraines with aura, cerebral ischemic events, cognitive impairment and dementia. The disease is caused by a mutation in the NOTCH3 gene. The major mutation type changes the number of cysteine residues in the EGF-like repeats of the Notch3 protein. In Czech Republic, currently used methods for molecular genetic analysis of the CADASIL disease are Sanger sequencing and MLPA. But there are patients with CADASIL-like symptoms who were not confirmed by these methods. Therefore, the aim of this thesis was to implement transcript analysis by Sanger sequencing of cDNA PCR products and quantitative real-time PCR (qPCR) to analyze gross deletions and duplications to clarify the molecular genetic basis of the disease. By transcript analysis, the existence of the transcript variant X1 was experimentally confirmed in control samples. Moreover, the results from transcript analysis showed that non-typical missense mutation c.1725G>A (p.T575=) which does not directly change the number of cysteine residues, can cause the CADASIL disease via missplicing and subsequent causing deletion including cysteine residues. Molecular genetic basis of CADASIL disease

CADASIL is a hereditary late-onset disease which is caused by a mutation in NOTCH3 gene. This gene belongs to the notch gene family that is conserved among Metazoa. The notch genes code transmembrane receptors which play role in Notch signal pathway during organism development. CADASIL is characterized by the impairing of small and medium vessels, especially cerebral arteries. The first symptoms appear in the middle age and the main symptoms are migraines with aura, recurrent strokes, cognitive impairment and dementia. The causes of this disease are mostly missense mutations altering the number of conserved cysteine residues in EGF-like domains of Notch3 protein. This thesis is focused on molecular genetic basis of CADASIL disease, it describes causative mutations and compares hypothesis about pathogenic mechanism of mutations. The penetration of the disease is not clarified yet but the thesis summarizes all current findings about genotype-phenotype correlations which can help to elucidate it. The phenotype differs between families and also between members of the same family. There is described the difference between men and women and the envi- ronmental influence too. Technologie a jakostní parametry ovocných vín

Bachelor thesis of Technology and parameters of fruits wines, it was prepred of Department of Post-Harvest Technology of Holticultural Products on Faculty of Holticulture Mendel University in Brno in ages 2014 -- 2015. The main aim of thesis was to focus for wood sortments fruit wines in the retail network and describe the legal requirements for quality fruit wines. Thesis describes technology the production of fruit wines and select retailers. The thesis includes the most commonly used fruit species for the of fruit wines in retail Tesco and Albert and in wineries Chateau Lednice, Pereg and Pankovo.
