High-performance liquid chromatography coupled with high-resolution mass spectrometry (HPLC-HRMS) is an analytical technique very often used in the new drugs development. This technique is widely used, from the confirmation of the proposed structure and identification of present impurities, through the assessment of absorption, distribution, metabolism and excretion to the monitoring of poisoning and intoxications. In the theoretical part, the presented dissertation thesis is focused on the basic characteristics of Alzheimer's disease, its currently available treatment and new treatment strategies. In addition, attention is paid to areas closely related to the experimental part, namely: the study of pharmacokinetics, analytical tools for determining pharmacokinetic parameters, liquid chromatography, mass spectrometry and the specifics of the bioanalytical methods. Sexism and objectification of women in Czech and Slovak rap video clips

This bachelor's thesis examines the attitudes of rappers, female rappers, and rap video producers towards the issues of sexism and objectification of women in Czech and Slovak rap music videos. The aim of this thesis is to determine whether these issues are perceived by rap artists and video producers and subsequently describe how women are portrayed in rap music videos. To achieve these goals, semi-structured interviews were conducted. The theoretical part of the thesis delves into the history and different approaches in researching subcultures, describing the main currents and significant representatives. It also explores the history and evolution of the hip-hop subculture, defining key concepts. The work also addresses the divergent development of the hip-hop subculture in the USA and Europe, highlighting the main differences. Furthermore, the thesis examines the history of rap in the USA, the Czech Republic, and Slovakia. Another section of this thesis discusses gender and hip-hop, sexism, and the objectification of women, outlining problematic portrayals of women in rap music videos.

Remembering in postapocalyptic world: Anticipatory memory in postapocalyptic series about pandemic

This thesis aims to explore the relationship between future, present and past in the context of memory creation in post-apocalyptic media content. Using the concept of anticipatory memory, the aim is to discover how characters from distant post-apocalyptic future marked by a catastrophic pandemic remember the time before the pandemic, meaning their past and our present. More specifically, this work will focus on concepts of memory and remembering and clarify key theories about memory that include collective and cultural memory, nostalgia or trauma. Next, I will describe temporal deviations in the narrative structure, anachronies of analepsis and prolepsis, and their use in fictional creation, and I will define the concept of anticipatory memory. Finally, I will characterize the genre of post-apocalyptic fiction, its narrative scheme and ways of depicting memory. I will also focus on characteristic motifs of post-apocalyptic fiction, which include, for example, the fight to live, changes in social structures, or the existence of remains of the old world in a post-apocalyptic world. The practical part is dedicated to qualitative research that uses elements of narrative and discursive analysis in the analysis of anticipatory memory in the post-apocalyptic series Station 11, Sweet Tooth and 12 Monkeys.

Noise against the binary - How women, queer, and non-binary people use electronic music, and synthesizers as the emancipation tool

This practical bachelor's thesis maps the connections between gender and the production of electronic experimental music, with an emphasis on the element of synthesizers, in domestic conditions. The thesis documents whether and how electronic experimental music and synthesizers can provide a more inclusive and accessible musical infrastructure in the Czech Republic. It also builds on an area that has been neglected by historical/academic scholarship and the media for several decades, and which has received more attention in recent years, especially in the context of the amplification of voices from gendered communities. This is evidenced, for example, in the music documentary Sisters with Transistors (2020) by filmmaker Lisa Rovner, which documents the pioneers of experimental electronic music, whose contributions largely underpin what we now perceive as electronic music. The practical output of the work is a series of seven interviews with figures from the Prague and Brno electronic scene who incorporate synthesizers in their work, divided into several categories according to the role they play within the electronic scene. Dramaturgy of Radio Wave programs - discursive analysis

The diploma thesis focuses on the critical analysis of dicourses in three selected programs of Radio Wave. The aim is to find out how these discourses are constructed depending on Ruth Wodak' discoursive strategies and how they reflect social reality. At the theoretical level, the work defines the historical consequences that led to the foundation of the Radio Wave and the concept of alternativness. These theory sections open the parts of the work that will focus on the internal compostion of the radio, its format and dramaturgy. To fulfill the goal of the work, we will use the method of qualitative analysis, specifically critical discourse analysis. We well present the methodology of the work and the analysis procedure of the individual programs: Studovna, Podhoubí, Záložka. We follow the aktivity in the "action field", the probable overlap of the douscourses and individual speeches. The aim of the work is to analyze individual statements, their argumentation, possible falsity and truthfulness, to find out what connotations individual social actors acquire, but also to declare how our chosen public law station participates in formuling the topics of individual discourses. For this purpose, individual episodes are selected, which are based on the concept of journalistic radio interview.

Transmedia Storytelling in the World of Harry Potter

The master thesis focuses on the analysis of the World of Harry Potter (Wizarding World) in a transmedia context. Currently, Harry Potter is among the best instances of successful transmedia franchising. The thesis will focus on official media channels and extensions which expand on the original narrative. The most significant contribution to transmedia research is the producer's approach to narrative structure in a transmedia context. In the theoretical framework, the Literature Review is focused on transmedia storytelling, media platform narratology, as well as participatory culture. All these elements are a crucial factor for spreading the narrative through all media channels. The main goal of the master thesis is to analyze the Wizarding World from the producer's point of view. The analysis will not be a semio-narrative analysis of primary extensions (the books and original films, including the Fantastic Beasts and other spinoff literature and film series), but rather a holistic analysis of all official extensions comprising the transmedia narrative and its structure and strategic deployment. Dance floor as a safe space: interviews with women of Czech club culture

The practical bachelor's thesis Dance Floor as a Safe Space: interviews with women in Czech club culture was written as a response to the ongoing debate about safe spaces in club cultures, in which the part of society most affected by the discussion, i.e. women, the queer community and other marginalized groups, rarely get a place to express themselves. The issue is more serious in the context of club cultures and the predominantly specific rave culture because these cultures present themselves as egalitarian cultures without any marginalization based on gender, sex, race, nation, or religion. In addition to gender diversity, the thesis also deals with the issue of safe spaces within club cultures; these two issues are closely and inextricably linked. The theoretical part of the thesis introduces the concepts that relate to the topic from a historical and contemporary context, it deals with the theory of the genre of the journalistic interview, gender and gender stereotyping, safe space, and last but not least club cultures and the rave culture under study. It will also delineate the functioning and differences between the scenes of both cities, Prague and Brno. Pirates in the post-revolutionary Czech airwaves: A case study of Radio Podzemí

The thesis "Pirates in the post-revolutionary Czech airwaves: a case study of Radio Podzemí" deals with the six-month-long broadcast of Radio Podzemí from České Budějovice, one of the first pirate stations in Czechoslovakia. The aim of the work is to introduce the period of the station's broadcasting, to place it in the historical context of the post-revolutionary airwaves and to illustrate the concepts of alternative media. Emphasis is then placed on aspects of community and pirate media, as well as the transformation of the medium from pirate to commercial radio. The qualitative research uses in-depth interviews with several founding members to collect data. The data collected is analysed using the grounded theory method. During its existence, Radio Podzemí exhibited a combination of elements of pirate, community and commercial media. The station's early broadcasts illustrated the strong anti-system motivation that is characteristic of pirate media. As time progressed, the station became dominated by elements and values of community media, eventually to be fully replaced by the station's commercial management. The contribution of this thesis is to complement the historical and media research that has been devoted to Radio Podzemí to date.

Popularity decline of the Clubhouse application in the Czech Republic and its reasons

This thesis examines the reasons for the rapid decline in popularity of the Clubhouse platform and the needs of emerging social networks to be competitive in the long term. The aim of the thesis is to use qualitative research to find out what reasons and circumstances led former users of the Clubhouse platform to use it and then abandon it, whether and under what conditions a renewal of its popularity is possible, and what respondents believe an emerging platform needs to fulfill in order to retain user attention. The theoretical part is devoted to the introduction of the Clubhouse platform, analysis of the most currently used social platforms, historical development of social networks and their influence on society. Data collection was conducted by the author through in-depth interviews with former users of the Clubhouse platform who had been actively using it for at least two months. The data collected was analysed using thematic analysis. The research results show that the rise of the Clubhouse platform was closely linked to the circumstances of the Covid-19 pandemic and the artificially created FOMO. The abandonment of the platform by users was mainly due to the departure of inspirational speakers and its lack of attractiveness in the long run. The research revealed, among other things, that...
