Noninvasive measurement of steroid homones and effect of hormonal manipulation on behaviour in the gecko Paroedura picta

Hormones influence life of all animals. Not only they affect physiological changes in organisms, but also impact their behaviour. This work focuses at two main groups of steroid hormones: glucocorticoids and androgens. Glucocortiods are activated in response to stress. Their levels can be measured using non-invasive methods, which have a range of advantages. The main advantage is the feedback-free sample collection for enzyme immunoassay. As the measurement involves metabolites of the hormones rather than the hormones themselves, prior validation of the method is, however, necessary. This work reports on a study aiming to validate non-invasive measurement on the Madagascar Ground Gecko (Paroedura Picta). The validation was based on ACTH challenge test: Synacthen Depot was injected, which should lead to increased blood level of glucocorticoids. The validation, however, was not successful. The measurement did not discover significant increase in the levels of the metabolites of glucocorticoids. In addition, the work focuses on behavioural effects of testosterone, the primary androgen. The effect of social factors on reproduction and stress in spiny mouse (Acomys cahirinus)

1 Summary This thesis deals with relationships among social factors, behaviour, physiology and reproduction in spiny mouse (Acomys cahirinus). Spiny mice are social rodents with that should be kept in families consisting of an adult male, multiple females and their descendants which mimic their wild social system. In contrast to many other muroid rodents, spiny mice produce after an extended gestation period only small litters consisting of relatively large and well developed (precocoial) newborns. The first part of the thesis is focused on effects of selected social parameters on glucocorticoid levels, which are widely used as a marker of stress. We focused on non-invasive monitoring of glucocorticoid metabolites (GCM) in faeces via enzyme immunoassay. For this purpose two specific antibodies were validated via an ACTH challenge test. Further, we constructed a special experimental apparatus which allowed us routine repeated collection of faecal samples in socially living rodents without noticeable disturbance to studied animals. Next, we monitored baseline concentrations of faecal GCM levels of individuals in family groups of commensal and non-commensal population. This Bachelor thesis focuses on a complete description of the logistics flow in VUES Brno s.r.o. It explains basic and technical logistics concepts and understandable indicates readers to the problem. In the next section analyzes and evaluates the current status of logistics business. At the conclusion summarizes the suggestions for improvement across the entire logistics chain, which should lead to more efficient passage of the contract, and more flexible responses to problems that arise.
