Fair trade cotton on the czech market

The theoretical part deals with system fair trade, cultivate and produce of fair trade and organic cotton. Following there are describe standards for small fair trade cotton producers and fair trade textile standard. There are presented some czech producers of organic cotton and some fair trade cotton farmers. The practical part contains results of questionnaires, which show views of consumers on organic and fair trade cotton. Futhermore the are analyse offers of organic and fair trade cotton in selected retail chains.
Kryoprezervace a transplantace spermatogonií kapra obecného

Cryopreservation and transplantation of germ cells in fish provides a suitable tool for preserving genetic information. By method of surrogate reproduction, the offspring with characters of the chosen donor can be obtained. In this case of our commercially important species common carp. However, for the successful cryopreservation of the germ cells, a suitable protocol for each species must be established. Several cryoprotectants were tested. The best of them, Me2SO, regarding the viability of spermatogonia, was tested for its different concentrations depending also on the rate of freezing. Further testing, related to the effect of tissue size, incubation time and added sugar, was performed. The result of the assay identified best cryomedium composed of 2.5M dimethylsulfoxide, added sugar of 0.3M glucose, 1.5% BSA and 25nM Hepes dissolved in PBS. The most suitable size of tissue was 100 mg, incubation time was 30 min and coolig rate was -1 ° C/min. This protocol ensures the highest viability rate of cryopreserved spermatogonia of common carp. The second part of the work was to verify the success of the transplantation of cryopreserved and fresh spermatogonia into a suitably chosen recipient, the goldfish, which shares similar reproductive characteristics with carp, but also offers reduction of space requirements or resistance to koi-herpes virus. The transplanted germ cells colonized the germ line and started gametogenesis in 42.5% (cryopreserved spermatogonia) and 52.5% (fresh spermatogonia) goldfish recipients, which demonstrated that the transplantation of cryopreserved spermatogonia of common carp can be successfully achieved.
Interspecies transplantations of embryonic germ cels in fish

Primordial gonocytes (PGC) are embryonic germ cells and they are precursors of sperm cells and oocytes. We can identify, isolate and grow them in cell's culture. We can store them in a frozen state in liquid nitrogen for long term. We can transplant these cells into recipient of the same or a close related species, which is sterile in the ideal case. The cells which are transplanted can develop in recipient's body so this recipient can produce the donor's gametes after maturation. Such individual is called germline chimera. The manipulation with germ cells can provide several benefits, for example preservation of germ cells from endangered or valuable species/stains in a gene bank. Or we can shorten generation interval that is convenient for late adolescent species in a case of appropriately chosen recipient. The aim of this thesis is to summarize knowledge about fish PGCs and a possible using of these cells for transplantations. Buying Behaviour with regards to the Local Production

The theoretical part deals with the explanation of basic concepts including factors which influence consumer behaviour. Futhermore is here explained local production and forms of purchase of local products. Practical part contains results of questionnaires, which show views and opinions of the Czech consumers. On the basic of the results are made recommendations for Czech producers and dealers.
The significance of sport events for tourism

The aim of the diploma thesis is to evaluate a significance of sport events for tourism and to analyze their impacts on tourism in the region. The thesis is divided into four chapters, the first two summarize theoretical knowledge - the economic importance of tourism, the inclusion of events into the context of the tourism industry and also specifics of sport events and their general impacts on the region, in which they are held. The next two chapters are devoted to the most important sport events in the Czech Republic as well as abroad and to the evaluation of their impacts on tourism in the destination. The final chapter focuses fully on the analysis of impacts of the Biathlon World Championship 2013 in Nové Město na Moravě on tourism in the region.
